Understanding of leaf stomatal responses to the atmospheric CO2 concentration, [CO2], is essential for accurate prediction of plant water use under future climates. However, limited information is available for the diurnal and seasonal changes in stomatal conductance (gs) under elevated [CO2]. We examined the factors responsible for variations in gs under elevated [CO2] with three rice cultivars grown in an open‐field environment under flooded conditions during two growing seasons (a total of 2140 individual measurements). Conductance of all cultivars was generally higher in the morning and around noon than in the afternoon, and elevated [CO2] decreased gs by up to 64% over the 2 years (significantly on 26 out of 38 measurement days), with a mean gs decrease of 23%. We plotted the gs variations against three parameters from the Ball‐Berry model and two revised versions of the model, and all parameters explained the gs variations well at each [CO2] in the morning and around noon (R2 > 0.68), but could not explain these variations in the afternoon (R2 < 0.33). In vertebrates, melanin production is restricted to pigment cells. This cell type-specific melanogenesis is considered to involve cell type-specific expression of the tyrosinase gene. Recently, there have been several reports that sequences in the 5’ flanking region of the mouse tyrosinase gene are responsible for cell type-specific expression of the transgene in mice. As the first step in the study of the evolution of the regulatory mechanisms for tyrosinase gene function in vertebrates, we constructed a fused gene, hg-Tyrs-J which includes a 1.0-kb 5’ flanking sequence of the human tyrosinase gene fused with mouse tyrosinase cDNA. By introducing the fused gene into fertilized eggs of albino mice, we obtained two mice that exhibited pigmentation in the skin and eyes and established a transgenic line from one of them. Further analyses revealed that the transgene was expressed cell type-specifically in these transgenic mice. We conclude, therefore, that the 1.0 kb 5’ upstream region of the human tyrosinase gene contains conserved cis-elements essential for cell type-specific expression of the tyrosinase genes in mice and humans. Results of our study may provide a clue to elucidate the evolutionary process of regulatory mechanisms of the tyrosinase gene. 相似文献

Isolation and characterization of microsatellite markers in Japanese pear (Pyrus pyrifolia Nakai) 总被引:1,自引:0,他引:1
Fourteen microsatellite markers were developed from an enriched genomic library of Japanese pear (Pyrus pyrifolia) by selective hybridization. They were characterized using 17 Japanese pear cultivars. The expected heterozygosity and observed heterozygosity ranged from 0.21 to 0.74 and from 0 to 0.88, respectively. Two to 11 alleles were detected per locus, with IPPN09 and IPPN15 judged to amplify multiple loci. IPPN17 was the most informative locus with the lowest probability of identity (0.19). The rhythm of adult eclosion in the Indian meal moth Plodia interpunctella Hübner (Lepidoptera: Pyralidae) is investigated under various photoperiods and temperatures aiming to determine the nature of the temperature compensation and the free‐running period. Insects that are committed to a nondiapause larval development show diel rhythms of adult eclosion at 30, 25 and 20 °C. At 30 °C, the eclosion peak (i.e. the mean time of eclosion) occurs approximately 20 h after lights off under an LD 4 : 20 h photocycle, and at approximately 15 h under an LD 20 : 4 h photocycle. At 25 °C, the peak of eclosion occurs approximately 19 h after lights off under an LD 2 : 20 h photocycle and at approximately 16 h under an LD 20 : 4 h photocycle. At 20 °C, the eclosion peak is significantly advanced under long days of >12 h (i.e. approximately 20 h after lights off under an LD 4 : 20 h photocycle and approximately 9 h under an LD 20 : 4 h photocycle), indicating an effect of both lights‐off and lights‐on signals on the timing of the adult eclosion. To determine the involvement of a self‐sustained oscillator, the rhythm of adult eclosion is examined under darkness at different temperatures (30 to 21 °C). The mean free‐running periods are 22.4, 22.8, 22.0 and 22.5 h at 30, 24, 23 and 22 °C, respectively, indicating that the eclosion rhythm is temperature‐compensated. However, this rhythm does not free‐run under constant darkness at 21 °C. Because a clear diel rhythm is observed under 24‐h photocycles at 20 °C, the oscillator might be damped out within 24 h at the lower temperature. 相似文献

Exceptionally high levels of intraspecific variation of mitochondrial DNA (mtDNA) sequences encoding 16S ribosomal RNA (16SrRNA) were detected in populations of the land snail Euhadra peliomphala from the eastern part of Japan. Geographical isolation has occurred within small areas and has created many genetically distinct local populations. These are morphologically segregated into two groups: the first consists of populations from the southern part of the Izu Peninsula, and the second of populations from other areas within the peninsula. They are separated by mountains in the middle of the peninsula. Sequence divergences between these two groups were 7.6–9.5%. The sequence divergences and geological ages of the Izu peninsula and its mountains suggest that the divergence rate of mitochondrial 16SrRNA sequences of this species is extremely rapid, approximately 10% per million years. This rapid evolutional")' rate of mtDNA and vicariance events due to tectonic events and sea level changes has also contributed to creating exceptional geographical variation in the mtDNA of this species. 相似文献

Background: Restoration of virus-specific immunity by virus specific T cells (VSTs) offers an attractive alternative to conventional drugs, and can be highly effective in immunocompromised patients, including hematopoietic stem cell transplant (HSCT) recipients. However, conventional VSTs manufacture requires preparation of specialized antigen-presenting cells (APCs), prolonged ex vivo culture in serum-containing medium and antigen re-stimulation with viruses or viral vectors to provide viral antigens for presentation on APCs. Methods: To simplify this complex process, we developed a method to generate multiple VSTs by direct stimulation of peripheral blood mononuclear cells (PBMCs) with overlapping peptide libraries in serum-free medium. Results: We generated VSTs that targeted seven viruses (cytomegalovirus [CMV], Epstein-Barr virus [EBV], adenovirus [AdV], human herpesvirus 6 [HHV-6], BK virus [BKV], JC virus [JCV] and Varicella Zoster virus [VZV]) in a single line. The phenotype, growth and specificity of multiple VSTs produced in serum-free medium were equivalent to those generated in conventional serum-containing medium. Discussion: The use of serum-free medium allows this approach to be readily introduced to clinical practice with lower cost, greater reproducibility due to the absence of batch-to-batch variability in serum and without concerns for infectious agents in the serum used. This simplified approach will now be tested in recipients of Human Leukocyte Antigen (HLA)–matched sibling HSCT. 相似文献

1. Plants take nutrients from the rhizosphere via two pathways: (i) by absorbing soil nutrients directly via their roots and (ii) indirectly via symbiotic associations with nutrient‐providing microbes. Herbivorous insects can alter these pathways by herbivory, adding their excrement to the soil, and affecting plant–microbe associations. 2. Little is known, however, about the effects of herbivorous insects on plant nutrient uptake. Greenhouse experiments with soybean, aphids, and rhizobia were carried out to examine the effects of aphids on plant nutrient uptake. 3. First, the inorganic soil nitrogen and the sugar in aphid honeydew between aphid‐infected and ‐free plants were compared. It was found that aphid honeydew added 41 g m?2 of sugar to the soil, and that aphids decreased the inorganic soil nitrogen by 86%. This decrease may have been caused by microbial immobilisation of soil nitrogen followed by increased microbial abundance as a result of aphid honeydew. 4. Second, nitrogen forms in xylem sap between aphid‐infected and ‐free plants were compared to examine nitrogen uptake. Aphids decreased the nitrogen uptake via both pathways, and strength of the impact on direct uptake via plant roots was greater than indirect uptake via rhizobia. The reduced nitrogen uptake by the direct pathway was as a result of microbial immobilisation, and that by the indirect pathway was probably because of the interaction of microbial immobilisation and carbon stress, which was caused by aphid infection. 5. The present results demonstrate that herbivorous insects can negatively influence the two pathways of plant nutrient uptake and alter their relative importance. 相似文献

Transect counts of butterflies were conducted in the northern part of Ibaraki, central Japan, from 1997 to 2001 at 11 census sites, composed of successive stages of deciduous forest development: grassland (one site, early abandoned stage); cutover land (one site, 1–5 years after clear‐cutting); secondary forests (very young (two sites, 6–9 years), middle (two sites, 16–22 years) and old (two sites, 47–51 years)) and old‐growth natural forests (three sites, ≥124 years old). A total of 86 species and 8858 individual butterflies were recorded by 29 sets (406 times) of transect counts. The species richness (number of species), abundance (number of individuals) and diversity indices (Shannon–Wiener H′ and Simpson's 1–λ) of butterflies were high in the early stages (grassland, cutover land and very young secondary forests) of secondary succession. Typical natural forest species increased with the progress of succession, and the old secondary forests and old‐growth natural forests had similar species composition. In contrast, most of the typical natural grassland species were recorded only in the grassland site. In the cutover land site, the number of individuals of grassland species considerably decreased from the first to the second year; furthermore, only one typical natural grassland species was recorded. Thus, the suitable stage for grassland butterfly species lasts for only 1–2 years after clear‐cutting. Old secondary forests (approximately>50 years old) would be able to give refuge to the forest butterfly species, including typical natural forest species. Based on the results, a practical, forestry‐based plan to conserve butterfly diversity was proposed. 相似文献

1. Species richness in a habitat patch is determined by immigration (regional) and extinction (local) processes, and understanding their relative importance is crucial for conservation of biodiversity. In this study, we applied the Island Biogeography concept to spring ponds connected to a river in southwestern Japan to examine how immigration and extinction processes interact to determine fish species richness in temporally variable environments. 2. Fish censuses were conducted 15 times in 13 study ponds at 1–4 month intervals from August 1998 through October 2000. Effects of habitat size (pond area), isolation (distance from the river) and temporal environmental variability (water level fluctuation) on (i) species richness, (ii) immigration and extinction rates and (iii) population size and persistence of each fish species were assessed. 3. The results revealed predominant effects of distance on species richness, immigration/extinction rates and population size and persistence. Species richness decreased with increasing distance but was not related to either pond area or water level fluctuation. A negative effect of distance on immigration rate was detected, while neither pond area nor water level fluctuation had significant effects on extinction rate. Further, population size and persistence of four species increased with decreasing distance, suggesting that, in ponds close to the river, immigrants from the river reduce the probability of extinction (i.e. provide a rescue effect), contributing to the maintenance of high species richness. 4. Overall results emphasise the importance of immigration processes, rather than extinction, in shaping patterns of species richness in our system. The predominant importance of immigration was probably because of (i) high temporal variability that negates habitat‐size effects and (ii) continuous immigration that easily compensates for local extinctions. Our results suggest that consideration of regional factors (e.g. connectivity, locations of source populations and barriers to colonisation) is crucial for conservation and restoration of local habitats. 相似文献
