Characterizing the carbon turnover in terrestrial ecosystems is critical for understanding and predicting carbon dynamics in ecosystems. We used in situ13C pulse labeling to track photosynthetic carbon fluxes from shoot to roots and to soil in a Kobresia humilis meadow on the Qinghai‐Tibet Plateau. We found that about 36.7% of labeled carbon was translocated out from the shoots within the first 24 h after photosynthetic uptake. This is equivalent to 66.1% of total 13C moving out from the shoot during the 32‐day chase period, indicating a rapid and large translocation of newly fixed carbon to belowground parts in these alpine plants. 58.7% of the assimilated 13C was transferred belowground. At the end of the chase phase, 30.9% was retained in living roots, 3.4% in dead roots, 17.2% lost as belowground respiration and 7.3% remained in the soil. In the four carbon pools (i.e., shoots, living roots, dead roots, and soil pools), living roots consistently had the highest proportion of 13C in the plant–soil system during the 32 days. Based on the 13C partitioning pattern and biomass production, we estimate a total of 4930 kg C ha?1 was allocated belowground during the vegetation growth season in this alpine meadow. Of this, roots accumulated 2868 kg C ha?1 and soils accumulated 613 kg C ha?1. This study suggests that carbon storage in belowground carbon pools plays the most important role in carbon cycles in the alpine meadow.  相似文献

Polymorphic microsatellite markers can provide essential information on genetic analyses of sociality and mating behaviour of insects. We developed eight microsatellite loci in the small carpenter bee, Ceratina flavipes, using a magnetic bead hybridization enrichment protocol. These loci showed two to eight alleles with expected heterozygosity of 0.21–0.87, and also seemed useful to such congeneric species as Ceratina okinawana distributed in southern Japan. By using these loci, the sociality and mating systems in Ceratina species, that are still controversial, are possible to be analysed.  相似文献

We developed seven polymorphic microsatellite loci for Myrmica kotokui from RAPD (rapid analysis of polymorphic DNA) fragments. These loci showed two to six alleles with expected and observed heterozygosity of 0.13–0.73 and 0.14–0.78, respectively. These loci are transferable to the congeneric species as Myrmica rubra, Myrmica luteola and Myrmica taediosa from northern Japan. These loci will allow analyses of genetic structure of Myrmica species at both the colony level and population levels.  相似文献

Rice internodes are vital for supporting high‐yield panicles, which are controlled by various factors such as cell division, cell elongation and cell wall biosynthesis. Therefore, formation and regulation of the internode cell‐producing intercalary meristem (IM) are important for determining the shape of internodes. To understand the regulation of internode development, we analysed a rice dwarf mutant, dwarf 50 (d50). Previously, we reported that parenchyma cells in the elongated internodes of d50 ectopically deposit cell wall phenolics. In this study, we revealed that D50 encodes putative inositol polyphosphate 5‐phosphatase (5PTase), which may be involved in phosphoinositide signalling required for many essential cellular functions, such as cytoskeleton organization, endocytosis and vesicular trafficking in eukaryotes. Analysis of the rice genome revealed 20 putative 5PTases including D50. The d50 mutation induced abnormally oriented cell division, irregular deposition of cell wall pectins and thick actin bundles in the parenchyma cells of the IM, resulting in abnormally organized cell files of the internode parenchyma and dwarf phenotype. Our results suggest that the putative 5PTase, encoded by D50, is essential for IM formation, including the direction of cell division, deposition of cell wall pectins and control of actin organization.  相似文献

Immunohistochemical localization of tyrosinase was examined with a monoclonal antibody (MoAb MAT-1) against human tyrosinase on routine formalin-fixed paraffin-embedded sections of 3 normal skin specimens, 15 melanocytic tumors (6 pigmented nevi, 3 juvenile melanomas and 6 malignant melanomas) and 3 non-melanocytic tumors. In the melanotic melanomas, almost all tumor cells were clearly stained with the antibody. In the nevocytic nevi, the nevus cells in lower epidermis and upper dermis were positive for MoAb MAT-1, but negative in middle and lower dermis. All three juvenile melanomas, one amelanotic melanoma, and three non-melanocytic tumors were entirely negative for MoAb MAT-1. Thus, MoAb MAT-1 could recognize the cells with melanogenic activity on routine formalin-fixed paraffin-embedded sections. However, the staining quality was not adequate for normal epidermal melanocytes, indicating that small technical innovations in the immunostaining process such as formalin fixation after PBS washing are required. Nevertheless, MoAb MAT-1 can be expected to be very useful for identifying melanogenic cells on paraffin-embedded sections, because we have to date no other antibody available for it.  相似文献

The development of the gonad in Oryzias celebensis was studied by light and electron microscopy. The path of PGC-migration, the time of the sex-differentiation, and the pattern of germ cell proliferation were almost identical to those in O. latipes. The most conspicuous difference was in the distribution of germ cells after migration. The gonadal anlage in O. celebensis developed only on the right side of the dorsal mesentery, although PGCs were stituated on the both sides of the embryos before migration. The testis retained its unilateral condition throughout development and acquired a unilobed shape. In the female, the right presumptive ovary developed over the mesentery, and the ovary became bilobed. Thereafter, right and left parts of the presumptive ovary fused to develop into a single ovary in the adult fish. The situation appears to be comparable to the gonadal asymmetry in birds, but the present observations suggest that the developmental processes of the asymmetrical gonads in this fish are quite different from those in birds.  相似文献

Phytoplasmas are plant-pathogenic bacteria that are associated with numerous plant diseases. We have previously reported the complete genomic sequence of Candidatus Phytoplasma asteris, OY strain, OY-M line, which causes mild symptoms. The phytoplasma genome lacks several important metabolic genes, implying that the consumption of metabolites by phytoplasmas in plants may cause disease symptoms. Here we show that the approximately 30-kb region including the glycolytic genes was tandemly duplicated in the genome of OY-W phytoplasma, which causes severe symptoms. Almost duplicated genes became pseudogenes by frameshift and stop-codon mutations, probably because of their functional redundancy. However, five kinds of genes, including two glycolytic genes, remained full-length ORFs, suggesting that it is advantageous for the phytoplasma to retain these genes in its lifestyle. In particular, 6-phosphofructokinase is known as a rate-limiting enzyme of glycolysis, implying that the different number of glycolytic genes between OY-W and OY-M may influence their respective glycolysis activities. We previously reported that the phytoplasma population of OY-W was higher than that of OY-M in their infected plants. Taking this result into account, the higher consumption of the carbon source may affect the growth rate of phytoplasmas and also may directly or indirectly cause more severe symptoms.  相似文献

目的是克隆milbemycin C5-O-甲基转移酶基因。以阿维菌素(avermectin)产生菌S.avermitilis中C5-O-甲基转移酶基因aveD为探针,利用核酸杂交分析法将aveD的主要同源区定位在(22)g-8-E-10阳性克隆所整合的.3kb SatⅠ-BgⅢ片段上。最后对aveD同源区及附近的核苷酸序列进行了测序分析和相应区域的基因缺失分析。结果表明,该区域存在一个月与aveD同源性较强的基因milD。与aveD功能相似,milD参与milbemycin的生物合成,在milbemycin生物合成中可能催化C5位OH的甲基化反应。  相似文献

Trehalase activities were found in several tissues of the adult American cockroach, Periplaneta americana. Among these, male accessory glands, fat body, thoracic muscle, hepatic cecum, blood and mid-gut contained high trehalase activity; activity in the male accessory gland was especially high. The enzymic properties of soluble trehalases were investigated and the enzymes from the male accessory gland were highly purified.

The properties of these enzymes were electrophoretically and kinetically distinct from each other. The presence of enzymes with somewhat different properties in different tissues suggests that trehalose utilization and trehalase activity may be regulated by way of a tissue-specific mechanism. The detailed properties of these enzymes are presented with a discussion of their regulation.  相似文献
