Nucleoside diphosphate kinase (NDP kinase) from Paramecium was purified to homogeneity. The native enzyme was 80 kDa (by gel filtration), with subunits of 18 and 20 kDa. Near the amino terminus, 15 of 20 residues were identical with those in human NDP kinase, and 17 of 20 with the awd gene product from Drosophila. NDP kinase bound α-labeled ATP and GTP, and a photoreactive GTP analog labeled both subunits. Purified NDP kinase underwent autophosphorylation on a histidine and a serine residue using either ATP or GTP as a substrate. The enzyme also catalyzed acid-stable phosphorylation of casein and phosvitin. This protein kinase activity is distinct from the histidine phosphorylation that is part of the NDP kinase catalytic cycle. Antiserum against the purified protein from Paramecium cross-reacted with 16- to 20-kDa proteins in most species tested, and with a larger protein (44 kDa) in Paramecium, Xenopus, and two human lines. The multiple forms (20 and 44 kDa) of the NDP kinase in Paramecium and its protein kinase activity, suggest that the protein is more than a housekeeping enzyme; it may have regulatory roles such as those of the NDP kinase-like awd protein of Drosophila and Nm23 protein of humans.  相似文献

Electron microscopic examinations of Glugea hertwigi and Spraguea lophii spores indicated the presence of a single plasma membrane; however, this membrane remained in the spore during the discharge of the sporoplasm from the spore. Although discharged spores retained the old plasma membrane, the extruded sporoplasms acquired a new plasma membrane. In order to determine where the new plasma membrane came from, we used two fluorescent probes with membrane affinities. The markers were tested on unfired and discharged spores. The probe, N-phenyl-1-naphthylamine (NPN), labeled the polaroplast membrane in addition to the apolar groups in the posterior vacuoles of unfired spores. After spore discharge, NPN label disappeared from the spore ghosts except for a slight fluorescence on residual plasma membranes. Much of the NPN-labeled membrane reappeared after spore discharge on the outer envelope of discharged sporoplasms. The probe chlorotetracycline (CTC) labeled calcium-associated membranes of spore polaroplasts. During spore discharge, the CTC fluorescence shifted from the polaroplast organelle of unfired spores to the outer envelope of discharged sporoplasms. These results indicate that the polaroplast organelle may provide the new plasma membrane for discharged microsporidian sporoplasms.  相似文献

Effects of prochloraz, imazalil, diclobutrazol, triadimefon and propiconazole were studied. Compared with most systemic fungicides previously tested they were all relatively toxic to R. trifolii on agar. Effects of soil residues of the compounds on the growth and nitrogen fixing capacity of white clover were measured. The lowest concentration of the fungicides used was that which would be present if all the active ingredient from a single application remained unchanged and evenly distributed in the top 5 cm of soil. This concentration of diclobutrazol decreased the size and weight of 11-wk-old clover plants. The rate of nitrogen fixation was reduced by twice this concentration and root nodulation by four times this amount. None of the other fungicides had any significant effect. The possibility of field application of diclobutrazol to cereals causing damage to clover crops is discussed. It is questionable whether harmful soil residues will ever accumulate from normal usage of the fungicide but exceeding the recommended rate of application, or application to cereals undersown with clover, might cause undesirable effects on the clover.  相似文献

Rat tracheal explants maintained as organ cultures exhibited a normal mucociliary epithelium for at least 46 days in the presence of retinyl acetate. In the absence of vitamin A the explant epithelium became quiescent or underwent a metaplastic change to a keratinizing squamous epithelium. This process was accelerated if explants were derived from vitamin A-deficient animals. Autoradiographic examination showed that [3H]glucosamine label accumulated in various cell types in the explant, but especially in the epithelium. It was found that the explants secreted mucous glycoproteins into the medium and that the production and biochemical characteristics of a specific mucin fraction were dependent upon the vitamin A status of the explant.  相似文献

Quantitative anatomical and other measurements were made onfully expanded flag leaves of a series of diploid, tetraploidand hexaploid Triticum and Aegilops species, and photosyntheticrates per unit leaf area were measured at light saturation (Pmax). Diploids had the highest Pmax, hexaploids the lowest with tetraploidsbeing intermediate. The anatomical features of tetraploids andhexaploids were generally similar, but different from the diploids.The diploids had thinner leaves with less dry matter and chlorophyllper unit area. The surface area of the mesophyll cells per unitvolume of mesophyll tissue was similar for all ploidy levels,as was the ratio mesophyil cell surface area per unit leaf area.It is argued that while these anatomical features are unlikelyto account for the observed variation in Pmax, it is possiblethat other structural factors with which they are correlatedmay causally influence Pmax. One such feature is the averagediffusion path length from the plasmalemma at the cell surfaceto the sites of carboxylation. Manipulating resource availability to assess the strength and effects of resource limitation on an animal population is relatively straightforward, and thus, common in the scientific literature. Resource quality, however, is rarely manipulated, as this requires a priori knowledge of a relationship between some measurable variation in the resource and individual preference and/or fitness. Recent research on nest‐site selection was used to inform the design of custom‐built nest‐boxes for an endangered hollow‐nester, the Gouldian finch (Erythrura gouldiae). By provisioning breeding populations with these ‘high quality’ artificial nest sites over 3 years, we experimentally investigated the strength and effects of nest‐site limitation in this species. Breeding pairs using nest‐boxes initiated nesting earlier, produced larger clutches and fledged more offspring per season than those in natural hollows. Total and mean reproductive output also increased in nest‐box provisioned sites over the duration of the study. All of these effects were predominantly driven by pairs initiating clutches earlier, potentially because of the reduced intra‐ and interspecific competition for nest sites at optimal breeding times. Our findings suggest that reproduction in wild Gouldian finch populations may be limited by the availability of high quality nest sites in the landscape, and that nest‐boxes could be used as a tool for enhancing reproduction in recovering populations. Furthermore, we conclude that resource quality is equally, if not more important than quantity when supplementing resource‐limited habitats.  相似文献

Twelve polymorphic microsatellite loci were isolated from polyploid alpine wood‐sorrel, Oxalis alpina (Oxalidaceae), and optimized for future studies of its breeding system. The loci were screened for variability among 72 individuals from Pinos Altos, New Mexico. The primers amplified loci with allele number ranging from two to 17 per locus and with estimates of Nei's genetic diversity varying from 0.10 to 0.99. These primers provide an opportunity to use polymorphic DNA markers to study the causes of breeding system variability in this species.  相似文献

Although microsatellite markers have become exceedingly popular in molecular studies of wild organisms, their development in some taxonomic groups is challenging. This is partly because of repetitive flanking sequences, which lead to the simultaneous amplification of alleles from multiple loci. Until now, these microsatellite DNA families have been considered unsuitable for population genetics studies, but here we describe our development of these repetitive flanking sequences (ReFS) as novel molecular markers. We illustrate the utility of these markers by using them to address an outstanding taxonomic question in the moth genus Schrankia.  相似文献

Twelve polymorphic microsatellite markers were developed from microsatellite‐enriched DNA libraries and mined from an expressed sequence tags library of Diaphorina citri, the vector of the citrus greening disease (huanglongbing). Analysis of 288 individuals from Florida, Texas, and Brazil showed that allelic diversity ranged from three to eight alleles per locus and observed and expected heterozygosities ranged from 0.014 to 0.569 and from 0.052 to 0.653, respectively. These variable microsatellite loci can provide means for assessing overall genetic variation and migration patterns for this agriculturally important pest species. This information can be used to aid in developing successful management strategies.  相似文献
