Resting vegetative buds of Norway spruce, Picea abies (L.) Karst., were induced to form adventitious bud primordia when cultured on medium -containing cytokinin. After transfer of the induced buds to medium lacking cytokinin, adventitious buds developed. The adventitious buds arose from meristems formed de novo in the needle primordia. No differences were found in the ability to form adventitious buds among buds collected from trees ranging from 5–50 years old.  相似文献

Adventitious buds were induced when isolated embryos of Norway spruce (Picea abies L.) were cultured on a defined medium containing 2iP. Anatomically different bud primordia were formed at high and low cytoldnin concentrations. The highest percentage of embryos forming bud primordia was obtained after 4–5 weeks on 2iP medium. Buds developed after transfer of the induced embryos to medium without growth regulators. Many of the buds developed into elongated shoots. No root primordia were observed in any of the induced embryos.  相似文献

The radiation of Partula land snails has produced a large array of distinct morphological, ecological and behavioural types occupying many tropical volcanic islands in the Pacific Ocean. Within the Society Islands of French Polynesia, the mode of evolution is thought to have involved a single colonization event on each island, with later speciation occurring largely in situ. The present study examines genetic variation in the mitochondrial cytochrome b gene among taxa within the Society Island archipelago. Levels of intraspecific variation are found to be high, but variation among species is sometimes small. Mitochondrial variants do not always cluster according to island and some species are found to be polyphyletic in the cytochrome b tree, despite other morphological and molecular evidence that strongly supports their monophyly. A possible explanation for the polyphyly of species is that different variants are derived from ancestral mitochondrial polymorphisms that have been retained despite speciation events. Although it is possible that there has been some gene flow among islands, the distribution of mitochondrial lineages across islands strongly indicates that their origins predate the colonization of the islands in the study, and that they are very unlikely to have evolved entirely in situ.  相似文献

A phylogenetic analysis of a combined data set for 560 angiosperms and seven outgroups based on three genes, 18S rDNA (1855 bp), rbcL (1428 bp), and atpB (1450 bp) representing a total of 4733 bp is presented. Parsimony analysis was expedited by use of a new computer program, the RATCHET. Parsimony jackknifing was performed to assess the support of clades. The combination of three data sets for numerous species has resulted in the most highly resolved and strongly supported topology yet obtained for angiosperms. In contrast to previous analyses based on single genes, much of the spine of the tree and most of the larger clades receive jackknife support 250%. Some of the noneudicots form a grade followed by a strongly supported eudicot clade. The early‐branching angiosperms are Amborellaceae, Nymphaeaceae, and a clade of Austrobaileyaceae, Illiciaceae, and Schi‐sandraceae. The remaining noneudicots, except Ceratophyllaceae, form a weakly supported core eumagnoliid clade comprising six well‐supported subclades: Chloranthaceae, monocots, WinteraceaeICanellaceae, Piperales, Laurales, and Magnoliales. Ceratophyllaceae are sister to the eudicots. Within the well‐supported eudicot clade, the early‐diverging eudicots (e.g. Proteales, Ranunculales, Trochodendraceae, Sabiaceae) form a grade, followed by the core eudicots, the monophyly of which is also strongly supported. The core eudicots comprise six well‐supported subclades: (1) Berberidopsidaceae/Aextoxicaceae; (2) Myrothamnaceae/ Gunneraceae; (3) Saxifragales, which are the sister to Vitaceae (including Leea) plus a strongly supported eurosid clade; (4) Santalales; (5) Caryophyllales, to which Dilleniaceae are sister; and (6) an asterid clade. The relationships among these six subclades of core eudicots do not receive strong support. This large data set has also helped place a number of enigmatic angiosperm families, including Podostemaceae, Aphloiaceae, and Ixerbaceae. This analysis further illustrates the tractability of large data sets and supports a recent, phylogenetically based, ordinal‐level reclassification of the angiosperms based largely, but not exclusively, on molecular (DNA sequence) data.  相似文献

Seven polymorphic microsatellite loci were isolated and characterized from the grapsid crab Pachygrapsus marmoratus using FIASCO (fast isolation by AFLP of sequences containing repeats) protocol. Twenty‐seven primer pairs were designed, 14 of which worked well in polymerase chain reaction (PCR), amplifying a fragment of the expected size. Variability was tested in 18 specimens collected along the coast of Tuscany (Italy). Five loci were discarded due to stutter products during their amplification, and two resulted to be monomorphic. The remaining seven loci, showed a number of alleles ranging from two to 14 and an observed heterozygosity ranging from 0.12 to 0.67.  相似文献

The deciphering of the process of genetic differentiation of species with insular distributions is relevant for biogeographical and conservation reasons. Despite their importance as old gondwanic islands and part of the western Indian Ocean biodiversity hotspot, little is known about the genetic structure of taxa from the Seychelles Islands. We have examined the patterns of structure and isolation within Urocotyledon inexpectata (Reptilia: Geckkonidae), an endemic species from this archipelago. Genetic diversity was screened from populations across the archipelago for both mitochondrial and nuclear genes. Gene genealogies and model‐based inference were used to explore patterns and timings of isolation between the main lineages. High levels of genetic diversity were found for the mitochondrial and some of the nuclear markers. This species harbours at least two highly differentiated lineages, exclusively distributed across the northern and southern groups of the islands. The main split between these was dated back to the Miocene–late Pliocene, but isolation events throughout the Pliocene and Pleistocene were also inferred. Migration between groups of islands was apparently nonexistent, except for one case. The low dispersal capabilities of this species, together with the intrinsic fragmented nature of its geographical distribution, seem to have resulted in highly structured populations, despite the cyclic periods of contact between the different island groups. These populations may currently represent more than one species, making U. inexpectata another example of a morphologically cryptic lineage with deep genetic divergence within gekkonids. The observed patterns suggest a hypothetical biogeographic scenario (of a main north–south phylogeographic break) for the Seychelles that can be further tested with the exploration of the phylogeographic structure of other Seychellois taxa. © 2011 The Linnean Society of London, Biological Journal of the Linnean Society, 2011, 104 , 177–191.  相似文献

Four species of swiftlet (Aves: Apodidae), two of which produce 'edible nests', occur sympatrically in Malaysia. Food boluses were analysed to investigate (1) diet differences and resource partitioning among the four species and (2) the influence of habitat on the diet of the Glossy Swiftlet Collocalia esculenta. Hymenoptera and diptera were the most abundant prey in all diets. Black-nest Swiftlets Aerodramus maximus were extremely selective; 89% of the items taken were hymenoptera, of which 97% were flying ants. White-nest Swiftlets A. fuciphagus and Mossy-nest Swiftlets A. salanganus took more than twice as many prey items as either of the other species. Their diets were taxonomically diverse and differed only in the average prey size taken, Mossy-nest Swiftlets taking the larger prey. Glossy Swiftlets took a higher proportion of coleoptera than did the other species. For the habitat comparisons, diptera were the main constituents of the Glossy Swiftlet diet in an urban habitat (71% of total prey items), whereas hymenoptera predominated in forest habitat (42% of total prey items). The results suggest that diet separation occurs among the four species of Malaysian swiftlet, in relation to both species and habitat. The implications of potentially habitat-driven dietary changes are discussed for the edible-nest swiftlets.  相似文献

The Euterpe edulis embryo consists of a prominent single cotyledon, a very short radicle-hypocotyl axis and an epicotyl. The epicotyl is obliquely angled with respect to the cotyledon; consequently it corresponds to one of the two categories recognized for palm seeds by DeMason (1988 ). Parenchyma, protoderm and procambium can be distinguished on the basis of position and shape of their cells, which are highly vacuolated with one central vacuole and the cytoplasm restricted to a thin parietal layer. Initial cells from both apical meristems are also vacuolated but they have small vacuoles distributed around the nuclei. Silica occurs in cell walls of some protodermal cells. Raphides, silica bodies and tannins all occur occasionally in vacuoles, especially in the basal cotyledon region. Most embryo cells lack storage reserves and exhibit an active state, with numerous mitochondria, RER cisternae and Golgi apparatus, indicating a strategy of continuous development without the interposition, at maturity, of a dry state. The endosperm consists of living cells with very large nuclei and thickened cell walls. Similar to the endosperm of other studied palm species, their cells exhibit a quiescent appearance with lipid, protein, minerals (in the cytoplasm) and mannans (in the cell walls) as the insoluble storage reserves.  © 2004 The Linnean Society of London, Botanical Journal of the Linnean Society , 2004, 145 , 445–453.  相似文献

1. While the effect on animals of handling them and fitting them with radio tags has been investigated, little work has been reported testing the effect of the presence of a human observer tracking an animal. 2. The activity of wood mice fitted with radio collars and confined to a semifield pen was measured in the presence and the absence of a human observer. Free‐ranging wood mice were radio tracked by using two protocols: ‘fixes’ taken periodically, and periods of continuous tracking. 3. There was no significant difference in any quantified aspect of the animals’ behaviour. This suggests that the presence of a human tracker has a negligible effect on the behaviour of wood mice.  相似文献
