Dissection and manipulation of metabolic signalling pathways   总被引:2,自引:0,他引:2  
The partitioning of resources between different plant organs and compounds is an important determinant of crop quality. We are attempting to change resource partitioning in crop plants by manipulating the cellular mechanisms involved in metabolite sensing and signalling. One of the proteins involved is SnRK1 (sucrose nonfermenting‐1‐related protein kinase 1), so‐called because of its homology and functional similarity with sucrose non‐fermenting 1 (SNF1) of yeast. SnRK1 is a protein kinase that plays a key role in the global control of plant carbon metabolism. Here we review studies on the characterisation of SnRK1 gene families, SnRK1 regulation and function, and the identification of SnRK1‐interacting proteins. We also describe some potential applications of manipulating SnRK1 activity, including controlling sprouting in stored potato tubers, inducing male sterility in barley and increasing sterol levels in oilseeds.  相似文献

1. Parasite effects on host behaviour frequently alter their hosts' trophic interactions. There are many compelling examples of such effects in herbivore‐based trophic interactions, but less attention has been paid to how parasite effects on host behaviour can alter mutualistic interactions. 2. Pollination mutualisms depend greatly on pollinator behaviour, and many pollinators are attacked by a wide range of parasites and parasitoids. 3. To investigate whether parasites affect pollination service via changes in host behaviour, natural variation in conopid fly parasitism was used to investigate the relationship between infection and Bombus impatiens Cresson behaviour foraging on arrays of Trifolium pretense L. flowers in the laboratory. The consequences of infection for seed set and seed mass were also examined. 4. Conopid parasitism was not related to any measured behavioural response; however, flowers visited by conopid‐parasitised bees set significantly heavier seeds than those visited by unparasitised bees. Larger bees were more likely to be parasitised, but the relationship between parasitism and seed set still held after accounting for body size. 5. The present results demonstrate that parasitoids may have positive impacts on per‐visit pollination, but, because larger bees were more frequently parasitised, parasitism could also affect pollination by removing the largest pollinators from the population.  相似文献

1. Food web interactions are complex and can respond to environmental changes in unpredictable ways that do not necessarily equate to the individual responses of each of the components of the food web. 2. Biomass can be used to evaluate the productivity of the three individual trophic levels, in the form of the Net Generational Productivity (NGP) and the performance of the entire food web with the newly developed tri‐trophic food web performance ratio (?3t). 3. These parameters were used to evaluate the performance of nine plant‐based tri‐trophic food webs composed of: potato, Solanum tuberosum L. and two cultivars of bell pepper Capsicum annuum L; three biotypes of the aphid Macrosiphum euphorbiae Thomas (Hemiptera: Aphididae) and the parasitoid wasp Aphidius ervi (Haliday) (Hymenoptera: Braconidae). 4. The NGP showed that the thermal window for biomass productivity for each trophic level is different and is reduced by approximately 4 °C with respect to the inferior level. Aphidius ervi had the smallest thermal window for biomass productivity and development. 5. The present results showed that the performance (?3t) of the tested food webs is influenced in a top‐down fashion, where the intra‐specific variation of the food web, namely the host plant, played a major role in the productivity of each of the subsequent trophic levels. 6. The ?3t suggested that exposure to high and low temperatures might severely affect the effectiveness of A. ervi as a biocontrol agent of the aphid M. euphorbiae in bell pepper and potato crops.  相似文献

Odonates were the first group of organisms where sperm competition and last male sperm precedence have been identified. With the development of 10 microsatellites for the emperor dragonfly Anax imperator, the function and priority patterns of the multiple sperm storage organs of females can be studied and compared between species in natural populations. In addition, two microsatellite loci developed for the sister species Anax parthenope, are also highly polymorphic in A. imperator. For the presented 12 microsatellite loci, the number of alleles per locus ranged from two to 24. Observed heterozygosity ranged from 0.07 to 0.88.  相似文献

Evaluating contributions of forest ecosystems to climate change mitigation requires well‐calibrated carbon cycle models with quantified baseline carbon stocks. An appropriate baseline for carbon accounting of natural forests at landscape scales is carbon carrying capacity (CCC); defined as the mass of carbon stored in an ecosystem under prevailing environmental conditions and natural disturbance regimes but excluding anthropogenic disturbance. Carbon models require empirical measurements for input and calibration, such as net primary production (NPP) and total ecosystem carbon stock (equivalent to CCC at equilibrium). We sought to improve model calibration by addressing three sources of errors that cause uncertainty in carbon accounting across heterogeneous landscapes: (1) data‐model representation, (2) data‐object representation, (3) up‐scaling. We derived spatially explicit empirical models based on environmental variables across landscape scales to estimate NPP (based on a synthesis of global site data of NPP and gross primary productivity, n=27), and CCC (based on site data of carbon stocks in natural eucalypt forests of southeast Australia, n=284). The models significantly improved predictions, each accounting for 51% of the variance. Our methods to reduce uncertainty in baseline carbon stocks, such as using appropriate calibration data from sites with minimal human disturbance, measurements of large trees and incorporating environmental variability across the landscape, have generic application to other regions and ecosystem types. These analyses resulted in forest CCC in southeast Australia (mean total biomass of 360 t C ha?1, with cool moist temperate forests up to 1000 t C ha?1) that are larger than estimates from other national and international (average biome 202 t C ha?1) carbon accounting systems. Reducing uncertainty in estimates of carbon stocks in natural forests is important to allow accurate accounting for losses of carbon due to human activities and sequestration of carbon by forest growth.  相似文献

The allocation and cycling of carbon (C) within forests is an important component of the biospheric C cycle, but is particularly understudied within tropical forests. We synthesise reported and unpublished results from three lowland rainforest sites in Amazonia (in the regions of Manaus, Tapajós and Caxiuanã), all major sites of the Large‐Scale Biosphere–Atmosphere Programme (LBA). We attempt a comprehensive synthesis of the C stocks, nutrient status and, particularly, the allocation and internal C dynamics of all three sites. The calculated net primary productivities (NPP) are 10.1±1.4 Mg C ha−1 yr−1 (Manaus), 14.4±1.3 Mg C ha−1 yr−1 (Tapajós) and 10.0±1.2 Mg C ha−1 yr−1 (Caxiuanã). All errors bars report standard errors. Soil and leaf nutrient analyses indicate that Tapajós has significantly more plant‐available phosphorus and calcium. Autotrophic respiration at all three sites (14.9–21.4 Mg C ha yr−1) is more challenging to measure, with the largest component and greatest source of uncertainty being leaf dark respiration. Comparison of measured soil respiration with that predicted from C cycling measurements provides an independent constraint. It shows general good agreement at all three sites, with perhaps some evidence for measured soil respiration being less than expected. Twenty to thirty percent of fixed C is allocated belowground. Comparison of gross primary productivity (GPP), derived from ecosystem flux measurements with that derived from component studies (NPP plus autotrophic respiration) provides an additional crosscheck. The two approaches are in good agreement, giving increased confidence in both approaches to estimating GPP. The ecosystem carbon‐use efficiency (CUEs), the ratio of NPP to GPP, is similar at Manaus (0.34±0.10) and Caxiuanã (0.32±0.07), but may be higher at Tapajós (0.49±0.16), although the difference is not significant. Old growth or infertile tropical forests may have low CUE compared with recently disturbed and/or fertile forests.  相似文献

Arctotis debensis R.J.McKenzie, a new species from the Albany Centre of Floristic Endemism, South Africa, is described and illustrated. It grows almost exclusively in grassland on kommetjies, a distinctive landform of depressions and mounds associated with giant earthworms, in a restricted area west of King William's Town, Eastern Cape Province. It is the first plant taxon reported to be endemic or near-endemic to this unusual habitat. The new species appears to have been collected rarely and previous collections examined have remained undetermined to species level. The new species is distinguished from closely allied species by the combination of its usually larger dimensions in all parts, prostrate habit, coriaceous leaves that are green, scabrous, and eglandular on the adaxial surface and densely lanate on the abaxial surface, and involucral-bract morphology.  © 2006 The Linnean Society of London, Botanical Journal of the Linnean Society , 2006, 151 , 581–588.  相似文献
