Mitotic PtK1cells were treated both during mid-anaphase and at furrow initiation with the potent microtubule (MT) stabilizing agent, taxol, to determine the role of MTs in the rate of cytokinetic events. Rates of cytokinesis (μm/min) were measured by changes in furrow diameter. Incubation of PtK1cells during mid-anaphase with 5 μg/ml taxol slows the rate of cytokinesis by an average of 43%. Instead of furrow initiation to midbody formation taking an average of 10.7 min (1.6 μm/min), furrowing to midbody formation was completed in an average of 19.0 min (0.9 μm/min), which does not include the 7-min period between taxol application in mid-anaphase and furrow initiation. Application of 5 μg/ml taxol to cells at furrow initiation had a reduced effect on decreasing the rate of cytokinesis and midbody formation; furrowing to midbody formation took an average of 14.6 min (1.2 μm/min). These data suggest that delays in the rate of cytokinesis is dependent on the mitotic stage at which taxol is applied. Ultrastructural analysis shows that taxol treatment of anaphase cells prevents midbody formation during early G1, yet MT number and organization in the furrowed region is not significantly altered from untreated cells. There is little change in the organization and amount of contractile ring microfilaments, yet filaments are also found parallel to midbody MTs. Our results may be explained by the fact that taxol tends to stabilize MTs which probably affects the rate at which they depolymerize in the terminal phases of cytokinesis. The composition and morphology of leaves exposed to elevated [CO2 ] usually change so that the leaf nitrogen (N) per unit dry mass decreases and the leaf dry mass per unit area increases. However, at ambient [CO2 ], leaves with a high leaf dry mass per unit area usually have low leaf N per unit dry mass. Whether the changes in leaf properties induced by elevated [CO2 ] follow the same overall pattern as that at ambient [CO2 ] has not previously been addressed. Here we address this issue by using leaf measurements made at ambient [CO2 ] to develop an empirical model of the composition and morphology of leaves. Predictions from that model are then compared with a global database of leaf measurements made at ambient [CO2 ]. Those predictions are also compared with measurements showing the impact of elevated [CO2 ]. In the empirical model both the leaf dry mass and liquid mass per unit area are positively correlated with leaf thickness, whereas the mass of C per unit dry mass and the mass of N per unit liquid mass are constant. Consequently, both the N:C ratio and the surface area:volume ratio of leaves are positively correlated with the liquid content. Predictions from that model were consistent with measurements of leaf properties made at ambient [CO2 ] from around the world. The changes induced by elevated [CO2 ] follow the same overall trajectory. During the past century, the upland breeding areas of Hen Harriers in Ireland have been extensively afforested. There is no evidence that this species avoids breeding in heavily forested landscapes and, indeed, young commercial forests in their second rotation are often selected as nest‐sites. However, Hen Harriers have coexisted with these forested areas for only a few decades and it is possible that such landscapes are suboptimal. We examined the relationship between breeding success and habitat using a dataset spanning three years and four study areas in the south and west of Ireland. We assessed whether nest success and fledged brood size were related to habitat type, both at the nest‐site and in the surrounding landscape. Neither measure of breeding productivity was related to total forest cover or to percentage cover of closed canopy forest in the landscape. However, in a subset of areas, high cover of second‐rotation pre‐thicket (young forests planted on land from which a first rotation has already been harvested) in the surrounding landscape was associated with low levels of breeding success. This may be due to factors related to predation, disturbance or prey availability. The fact that second‐rotation pre‐thicket is a preferred habitat for nesting in Ireland suggests that Hen Harriers may be making suboptimal decisions in the landscapes available to them. 相似文献

Abstract In Gryllus bimaculatus, more digestive enzymes (amylase, trypsin, aminopeptidase) are secreted in the caecum of fed crickets than in unfed crickets, but the enzymes are released continuously at a basal rate in unfed animals. The rate of synthesis of the enzymes appears to parallel their rate of release. Digestive enzymes are released in response to a specific ratio of nutrients, although a high nutrient component in the food does not necessarily induce a high digestive enzyme release for that component. Rinsed flat‐sheet preparations of the caecum are incubated with specific nutrients (carbohydrates and proteins) and various concentrations of a neuropeptide (type‐A allatostatin), which affects generally the basal rates of secretion. Both maltose and glucose increase the release of amylase in vitro, but starch produces an inhibition of amylase release at lower concentrations. Bovine serum albumin (BSA), peptone and a mixture of amino acids have almost no effect on the release of aminopeptidase or carboxypeptidase, and only low concentrations of peptone increase trypsin release. High concentrations of both BSA and peptone strongly inhibit trypsin activity, perhaps by excess substrate binding to the trypsin active site. The allatostatin Grybi‐AST 5 elevates the release of amylase in vitro, but not of trypsin or aminopeptidase, in 2‐day‐old fed females. In the caeca from 1‐day‐old unfed crickets, both amylase and the trypsin release are stimulated in the presence of AST 5. The paracrine AST 5 is probably released from the gut endocrine cells and binds to the enzyme‐producing caecal cells. 相似文献

Abstract: A collection of very early brephic juveniles from the Upper Aalenian of the Cotswolds, England, has provided the first opportunity to study the ontogeny of thecideoids prior to the recognizable development of a median septum. Traced towards their origin, the enduring characters of thecideides appear to be cementation, a delthyrium closed by a pseudodeltidium, an almost circular dorsal valve with a prominent erect bilobed cardinal process, well‐defined inner socket ridges, widely spaced lateral adductor muscle fields, tubercles, fibrous secondary shell lining both valves and probably endopunctae. The subperipheral rim and median septum so characteristic of adult thecideoids are undeveloped in the earliest juveniles. Initiation of the development of the free ventral wall in the ventral valve is identified as an important event in thecideoid ontogeny. Further important discoveries arising from the study are that the crura‐like outgrowths which form the brachial bridge in thecideides are not structurally homologous with the crura of spiriferides or terebratulides and that during their earliest ontogenetic development thecidellinids and thecideids are indistinguishable. Also, the identification of morphological characters correlative with both palaeontological and neontological approaches to thecideoid phylogeny has important implications for thecideide taxonomy. Interpretation of the morphology exhibited by the Cotswold specimens introduces the probability that during their earliest ontogeny moorellinin, thecidein and lacazellin dorsal valves follow the same development pattern and the described ontogenetic sequence has been corroborated by evidence from Jurassic rocks of Argentina, North America and France, from Cretaceous rocks of England, Central Europe and Czechoslovakia, also from extant species from the Atlantic, Indian and Pacific oceans, and to an extent which suggests the pattern of ontogenetic development revealed has been typical of thecideoids throughout their history. 相似文献

1. We measured the hyporheic microbial exoenzyme activities in a floodplain river to determine whether dissolved organic matter (DOM) bioavailability varied with overlying riparian vegetation patch structure or position along flowpaths. 2. Particulate organic matter (POM), dissolved organic carbon (DOC), dissolved oxygen (DO), electrical conductivity and temperature were sampled from wells in a riparian terrace on the Queets River, Washington, U.S.A. on 25 March, 15 May, 20 July and 09 October 1999. Dissolved nitrate, ammonium and soluble reactive phosphorus were also collected on 20 July and 09 October 1999. Wells were characterised by their associated overlying vegetation: bare cobble/young alder, mid‐aged alder (8–20 years) and old alder/old‐growth conifer (25 to >100 years). POM was analysed for the ash‐free dry mass and the activities of eight exoenzymes (α‐glucosidase, β‐glucosidase, β ‐N‐acetylglucosaminidase, xylosidase, phosphatase, leucine aminopeptidase, esterase and endopeptidase) using fluorogenic substrates. 3. Exoenzyme activities in the Queets River hyporheic zone indicated the presence of an active microbial community metabolising a diverse array of organic molecules. Individual exoenzyme activity (mean ± standard error) ranged from 0.507 ± 0.1547 to 22.8 ± 5.69 μmol MUF (g AFDM)?1 h?1, was highly variable among wells and varied seasonally, with the lowest rates occurring in March. Exoenzyme activities were weakly correlated with DO, DOC and inorganic nutrient concentrations. 4. Ratios of leucine aminopeptidase : β‐glucosidase were low in March, May and October and high in July, potentially indicating a switch from polysaccharides to proteins as the dominant component of microbial metabolism. 5. Principal components analysis indicated that there were patch effects and that these effects were strongest in the summer. 6. DOM degradation patterns did not change systematically along hyporheic flowpaths but varied with overlying forest patch type in the Queets River hyporheic zone, suggesting that additional carbon inputs exist. We hypothesise that the most likely input is the downward movement of DOM from overlying riparian soils. Understanding this movement of DOM from soils to subsurface water is essential for understanding both the hyporheic metabolism and the carbon budget of streams and rivers. 相似文献
