Maturation proteins associated with desiccation tolerance in soybean   总被引:17,自引:2,他引:15       下载免费PDF全文
A set of proteins that accumulates late in embryogenesis (Lea proteins) has been hypothesized to have a role in protecting the mature seed against desiccation damage. A possible correlation between their presence and the desiccation tolerant state in soybean seeds (Glycine max L. Chippewa) was tested. Proteins that showed the same temporal pattern of expression as that reported for Lea proteins were identified in the axes of soybean. They were distinct from the known storage proteins and were resistant to heat coagulation. The level of these “maturation” proteins was closely correlated with desiccation tolerance both in the naturally developing and in the germinating seed: increasing at 44 days after flowering, when desiccation tolerance was achieved, and decreasing after 18 hours of imbibition, when desiccation tolerance was lost. During imbibition, 100 micromolar abscisic acid or Polyethylene glycol-6000 (−0.6 megapascals) delayed disappearance of the maturation proteins, loss of desiccation tolerance, and germination. During maturation, desiccation tolerance was prematurely induced when excised seeds were dried slowly but not when seeds were held for an equivalent time at high relative humidity. In contrast, maturation proteins were induced under both conditions. We conclude that maturation proteins may contribute to desiccation tolerance of soybean seeds, though they may not be sufficient to induce tolerance by themselves.  相似文献

The relative importance of stomatal and nonstomatal limitations to net photosynthesis (A) and possible signals responsible for stomatal limitations were investigated in unhardened Pinus taeda seedlings at low soil temperatures. After 2 days at soil temperatures between 13 and 7°C, A was reduced by 20 to 50%, respectively. The reduction in A at these moderate root-chilling conditions appeared to be the result of stomatal limitations, based on the decrease in intercellular CO2 concentrations (ci). This conclusion was supported by A versus ci analysis and measurements of O2 evolution at saturating CO2, which suggested increases in stomatal but not biochemical limitations at these soil temperatures. Nonuniform stomatal apertures, which were demonstrated with abscisic acid, were not apparent 2 days after root chilling, and results of our A versus ci analysis appear valid. Bulk shoot water potential (ψ) declined as soil temperature dropped below 16°C. When half the root system of seedlings was chilled, shoot ψ and gas-exchange rates did not decline. Thus, nonhydraulic root-shoot signals were not implicated in stomatal limitations. The initial decrease in leaf conductance to water vapor after root chilling appeared to precede any detectable decrease in bulk fascicle ψ, but may be in response to a decrease in turgor of epidermal cells. These reductions in leaf conductance to water vapor, which occurred within 30 minutes of root chilling, could be delayed and temporarily reversed by reducing the leaf-to-air vapor-pressure deficit, suggesting that hydraulic signals may be involved in initiating stomatal closure. By independently manipulating the leaf-to-air vapor-pressure deficit of individual fascicles, we could induce uptake of water vapor through stomata, suggesting that nonsaturated conditions occur in the intercellular airspaces. There was an anomaly in our results on seedlings maintained for 2 days at soil temperatures below 7°C. Lower A appeared primarily the result of nonstomatal limitations, based on large increases in calculated ci and A versus ci analysis. In contrast, measurements of O2 evolution at saturating CO2 concentrations implied nonstomatal limitations per se did not increase at these temperatures. One explanation for this paradox is that calculations of ci are unreliable at very low gas-exchange rates because of inadequate measurement resolution, and limitations of A are predominantly stomatal. An alternative interpretation is that increases in ci are real and the results from O2-evolution measurements are in error. The high CO2 concentration used in O2-evolution measurements (15%) may have overcome nonstomatal limitations by enzymes that were down-regulated by a feedback mechanism. In this scenario, carbohydrate feedback limitations may be responsible for nonstomatal reductions in A after 2 days at soil temperatures below 7°C.  相似文献

Differentiation in morphometric traits among experimental populations of the housefly subjected to an experimental bottleneck was examined for replicate lines founded with one, four or 16 pairs of flies. Differentiation among lines within a bottleneck size was significantly greater than predicted by drift in relation to the additive genetic variation for these traits within the founding population. Two models of nonadditive genetic variance were investigated to interpret these results, one involving dominance of allelic effects within loci and another incorporating multiplicative epistasis. Both models generated more variation among lines as a direct result of sampling during the bottleneck than predicted by a model with additive gene action. The pattern of differentiation among our experimental lines in relation to these models conformed more to the model incorporating epistasis. Nevertheless, it may be difficult to distinguish differentiation among lines occurring during a bottleneck as a result of nonadditive gene action from that caused by diversifying selection among lines after the bottleneck.  相似文献

The photosynthetic growth action spectrum of a green alga at three bands of visible light (blue, orange, and red) at fixed quanta input and under light-limiting conditions was measured in a batch cultivation system. Quantum efficiencies (biomass dry weight increment per quanta absorbed) were better in the yellow-red region than in the blue region. Results served as a basis for the design and optimization of a dye system that would shift the energy of solar radiation to the required wavelength range by absorbing ultraviolet to blue radiation and emitting in the yellow-red, thus enhancing algae growth. Direct incorporation of dyes into the growth medium, although theoretically expected to enhance growth, in fact resulted in dye decomposition, toxicity to algae and consequently in growth inhibition. Indirect application of dyes in a double tubular reactor (algae inside and dye solution outside) demonstrated growth enhancement for certain dyes with high quantum yields and stability, which had suitable absorption/emission spectra for artificial light sources used. The maximum indirect growth enhancement was obtained using rhodamine 6G at a concentration of 3x10(-5)M with tungsten filament lamp sources.  相似文献

The charge-state model of electrophoretic variation was tested by comparing the distances between nearest electromorphs of five enzyme loci within polymorphic species and among pooled species of sea anemone. If the charge-state model is generally true, and in particular if it allows linear distance between electromorphs to be used as a measure of genetic distance, then electromorphs of different species should be on the same "mobility ladder". Therefore, distances between adjacent electromorphs should be approximately equal for the two sets of comparisons. It was found that distances between adjacent electromorphs for each locus were significantly smaller for the pooled comparisons than within polymorphic species. Thus, it was concluded that much of the variation detected among different species does not conform to the charge-state model, and therefore that distance between electromorphs per se would not be a good measure of genetic distance. However, the charge-state model does appear to adequately account for most of the variation existing as common polymorphisms within species, or between very closely related species. Possible reasons for this apparent difference in the nature of the variation seen within and among species are discussed.  相似文献

The effect of host plant cultivar on H2 evolution by root nodules was examined in symbioses between Pisum sativum L. and selected strains of Rhizobium leguminosarum. Hydrogen evolution from root nodules containing Rhizobium represents the sum of H2 produced by the nitrogenase enzyme complex and H2 oxidized by any uptake hydrogenase present in those bacterial cells. Relative efficiency (RE) calculated as RE = 1 − (H2 evolved in air/C2 H2 reduced) did not vary significantly among `Feltham First,' `Alaska,' and `JI1205' peas inoculated with R. leguminosarum strain 300, which lacks uptake hydrogenase activity (Hup). That observation suggests that the three host cultivars had no effect on H2 production by nitrogenase. However, RE of strain 128C53 was significantly (P ≤ 0.05) greater in symbiosis with cultivar JI1205 than in root nodules of Feltham First. At a similar rate of C2H2 reduction on a whole-plant basis, nearly 24 times more H2 was evolved from the Feltham First/128C53 symbiosis than from the JI1205/128C53 association. Root nodules from the Alaska/128C53 symbiosis had an intermediate RE over the entire study period, which extended from 21 to 36 days after planting. Direct assays of uptake hydrogenase by two methods showed significant (P ≤ 0.05) host cultivar effects on H2 uptake capacity of both strain 128C53 and the genetically related strain 3960. The 3H2 incorporation assay showed that strains 128C53 and 3960 in symbiosis with Feltham First had about 10% of the uptake hydrogenase activity measured in root nodules of Alaska or JI1205. These data are the first demonstration of significant host plant effects on rhizobial uptake hydrogenase in a single plant species.  相似文献
