A 2-year study was designed to investigate the spatiotemporal distribution of sulphide in the water column of a saline lake, Lake Hiruga in Fukui, Japan. Water samples were collected in Lake Hiruga periodically between April 2014 and March 2016 at appropriate depths with simultaneous in situ measurements of physicochemical parameters such as temperature, salinity and dissolved oxygen (DO) concentration. Sulphide started accumulating in the hypolimnion in September 2014, whilst that did in June 2015. The sulphide concentrations increased toward the end of December in both 2014 and 2015. Then the sulphide disappeared from the hypolimnion in April 2015 and February 2016. The difference in starting season of the sulphide accumulation was closely connected with DO concentrations in the hypolimnion in circulation periods of winter season. The DO concentrations were regulated by water column stability before thermal stratification periods. Our analysis of the sulphide distribution for the 2 years of the study suggested that accumulation of sulphide in the hypolimnion during stratified periods appears to be associated with strength of water column stability in the winter-spring seasons before stagnant periods in Lake Hiruga. 相似文献

A new pest of Welsh onion and carrot, the dark-winged fungus gnat, Bradysia sp., was found in the northern region of Saitama Prefecture, Japan. DNA sequences of mitochondrial cytochrome c oxidase subunit I revealed that Bradysia sp. is identical to the Chinese chive gnat, Bradysia odoriphaga Yang and Zhang. This suggests that Bradysia sp. may be an invasive species that originates from continental East Asia. Direct sequencing analysis and PCR using a species-specific primer were found to be useful tools for discriminating Bradysia sp. from other native Japanese fungus gnats, such as Bradysia impatiens (Johannsen), Pnyxia scabiei (Hopkins), and Lycoriella ingenua (Dufour). 相似文献

Two marihuana constituents, cannabidiol (1) and cannabidiolic acid (4) were each converted mainly to two metabolites using tissue segments of Pinellia ternata tuber. The structures of the metabolites formed from 1 were determined to be 1-O-D-glucopyranoside and 1-O-D-diglucopyranoside by 1H nmr, 13C nmr and fabms. Those from 4 were determined as 4-O-D-glucopyranoside and 10-hydroxyl 4-O-D-glucopyranoside. In time course experiments, 1 was absorbed rapidly by the tissues and glucosylated. A "bound form" of cytokinin extracted from roots of the riceplant was purified by column and thin layer chromatography,then analysed by gas chromatography and IR spectrophotometry.Results strongly suggest that the bound cytokinin is zeatinglucoside. The extracellular protein MPB70 is a heat-stable immunogenic protein which was found in the culture filtrate of Mycobacterium bovis BCG Japanese. We determined the complete nt and aa sequences of MPB70 and correlated with the previously reported data. The N-terminal sequence revealed that the signal peptide (SP) consisted of 30 aa and that the mature protein had 163 aa with a molecular weight of 16,305. The SP displayed a characteristic feature of an Ala-rich property which would be efficient in a SP function. 相似文献

The mechanisms of intracellular calcium store depletion and store-related Ca2+ dysregulation in relation to apoptotic cell death in PC12 cells were investigated at physiological temperatures with a leak-resistant fluorescent indicator dye Fura-PE3/AM by a cooled CCD imaging analysis system. Electron microscopic observations have shown thapsigargin (TG; 100 nM)-induced apoptosis in PC12 cells. Thorough starvation of stored Ca2+ by BAPTA/AM (50 μM), or La3+ (100 μM) enhanced while dantrolene (100 μM) attenuated the TG-induced apoptosis by preventing a calcium release from internal stores. An immunoblotting analysis revealed an enhanced expression of GRP78, the hallmark of endoplasmic reticulum (ER) stress when cells were treated by TG along with BAPTA/AM. These results indicate that the depletion of the intracellular Ca2+ stores itself induces the ER stress and apoptosis in PC12 cells without any involvement of the capacitative calcium entry (CCE) or a sustained elevation of intracellular Ca2+ concentrations ([Ca2+]i). 相似文献

By developing gene-specific RT-PCR and using filters to allow transmission down to 290 nm (UV-B+) or blocking all radiation below 320 nm (UV-B–), the effect of UV-B+ and UV-B– light on expression of each of the presently known seven members of soybean chalcone synthase (CHS) gene family in dark-grown seedlings was analyzed. Dark expression was detectable already in 18 h dark-germinating embryos, with progressive increases on successive days, suggesting that chs belongs to a class of genes expressed very early during germination, and that the expression at this stage is either constitutive or induced by non-light-dependent factors present in the seed or made available following imbibition. Exposure of 18 h dark-germinating embryos to UV-B– or to UV-B+ light did not lead to an increase in chs signal. However, the 24 h dark-germinating embryos showed a distinct effect of UV-B+, interestingly coinciding with the stage when the head of seedlings was in the process of being pushed up above ground by stem elongation, suggesting the possibility of a developmental switch modulating the appearance of UV-B response. The response to UV-B– was most prominent in chs1 and almost silent in chs2, while the up-regulation by UV-B+ was most prominent in chs5 and chs6 and much less so in chs2. Interestingly, chs2 was noted to be the only member of the Gmchs gene family devoid of H-box, raising the possibility that the H-box may be a good indicator of the photo-inducibility of a chs gene. 相似文献
