The cryoprotective effect of polyols on rat embryos was measured after two-step freezing, and the mechanism of action of polyols on embryo survival was examined. Rat embryos frozen in solution of polyol by two-step method at the morula stage showed higher survival than that obtained using DMSO. As the number of hydroxyl groups increased, the cryoprotective effect of the polyol increased. However, this was true only when the additive could permeate the cell membrane. Of the additives tested, four or five carbon polyols were most effective at concentrations of 0.3 or 1.0 M than two, three, six, or seven carbon polyols. The highest survival rate was obtained with adonitol, which yielded 83% embryo survival at 1.0 M and 67% even at 0.3 M. Embryos frozen in 0.3 M adonitol and transferred directly into foster mothers without any dilution of the additive after thawing developed into live young. During slow cooling below -40 degrees C, embryonic blastomeres exhibited cell fusion only in the presence of adonitol. These findings suggest that one cryoprotective action of polyols is that the hydroxyl groups act both on the cell surface and the cytoplasm to stabilize the bound water on the embryonic membrane, and that the length of the C-chain determines the permeability of the membrane to the additive. 相似文献

Effects of sialodacryoadenitis virus infection on the reproduction of Wistar and Sprague-Dawley (SD) rats were studied. Estrous cycle was considerably out of order in about 40% of infected rats of both strains, starting on days 4 to 20 postinfection and persisting for 2 to 12 days. Of Wistar and SD dams infected on day 0 of gestation about 30% and 10% of the fetuses were found dead, while only 4 and 3% of non-infected dams. In severely diseased Wistar and SD dams infected on day 15 of gestation, the offspring showed death rates of 57% and 13%, respectively, because of inadequate nursing. 相似文献

The conformations of fibroblast and E. coli-derived recombinant human interferon-beta s were studied by circular dichroism and nuclear magnetic resonance spectroscopy in the acidic pH region of 4.6 to 1.6. Both interferons have very similar conformations with high alpha-helix contents (approximately 70%). These results suggest that glycosylation does not appreciably change the conformation of human interferon-beta. Moreover, a slow conformational change is observed below pH 2.0, which induces the disruption of beta-sheets. 相似文献

Using an avirulent strain of herpes simplex virus (HSV), SKa, and a methylcholanthrene induced sarcoma cell line, Meth A cells, we have developed a reliable target cell system for detection of cell-mediated cytotoxicity directed against HSV-infected cells. SKa-infection in Meth A produced no progeny virus but induced HSV-specific surface antigens as revealed by radioimmunoassay using 125I-labeled HSV antibody. Spontaneous release of 51Cr from the SKa-infected Meth A cells was no more than that from uninfected control cells but a strong spontaneous 51Cr release was produced in Meth A cells infected with KOS, a virulent strain which produced a progeny virus in Meth A and was lytic for the cells. When used as a target, SKa-infected Meth A cells could detect HSV-specific cytotoxicity by spleen and lymph node lymphocytes of mice immunized with SKa and KOS. This system also detected effector cytotoxic lymphocytes stimulated in vitro by mixed cultures of immune spleen cells and KOS-infected Meth A cells. Thus, the system should be valuable in studies of cell-mediated cytotoxicity directed against HSV-infected cells. 相似文献

For period from 1971 to 1975, 18 cases of osteosarcoma were experienced among 9132 spontaneously hypertensive rats. The onset was at 72 to 145 days of age and death occurred before 423 days of age. The tumors were multiple being distributed in the skull (11 of 14 cases), caudal vertebrae (8 of 14 cases), and fore- and hind-limb. All the tumors were mainly composed of sarcomatous tissue with some osteoid and cartilage. Metastasis was never observed. Serum A1-P levels of tumor bearing animals were 2 times higher than those without tumor. Bronchiectasis and abscess formation of submaxillary lymph nodes were noted in most affected animals. 相似文献

The immunogenic capacity of herpes simplex virs (HSV)-infected cells and their subcellular membrane fractions was investigated by assessing the anti-HSV cytotoxic T lymphocyte (CTL) response in cultures of spleen lymphocytes from HSV-primed BALB/c mice. Methylchloranthrane-induced fibrosarcoma (Meth A) cells infected with HSV (HSV-Meth A) were fixed either with glutaraldehyde or by heating at 56 C to preserve their immunogenic competence and then used as a stimulator. Microsomes and plasma membranes were prepared from HSV-Meth A and their immunogenic activities were determined. Though the recovery of stimulatory activity in the plasma membrane fraction was half of that in the microsome fraction, the activity in the former was much more stable than in the latter and the plasma membrane fraction proved to be well qualified as an immunogen for anti-HSV CTL induction. The immunopotentiating property of spin-labeled lipophilic muramyl dipeptide (SL-MDP) in liposomes was studied as to the plaque-forming cell (PFC) response to glycophorin, as an antigen, in membranes. The effect of SL-MDP depended on the densities of both SL-MDP itself and the antigen. The addition of the optimal amount of SL-MDP to liposomes containing a low density (0.016 mol%) of the antigen increased the PFC response by three times, whereas the presence of SL-MDP in optimal antigen density (0.032 0.127 mol%) membranes was rather inhibitory. In these liposomes, the amounts and molecular states of SL-MDP were determined from ESR spectra and are discussed in connection with its immunopotentiating property. 相似文献

In an earlier report [H. Utsumi and M. M. Elkind, Radiat. Res. 119, 534-541 (1989)], it was shown that the survival of V79 Chinese hamster cells treated with bleomycin was significantly reduced by a posttreatment with anisotonic phosphate-buffered saline in a manner that was qualitatively similar to what had been observed with X rays [H. Utsumi and M. M. Elkind, Radiat. Res. 77, 346-360 (1979)]. This similarity suggested that similarities might exist in the cyclic variation in the suppression of the repair of potentially lethal damage following treatment with bleomycin or X rays. Accordingly, the age-response variations of survival, with or without a posttreatment challenge with hypertonic buffer, were compared in the same experiment when cells were treated with either agent. Although a significant difference was observed near the G1/S-phase border, in general the damage induced by the two agents showed a similar dependence on cell age, and posttreatment with hypertonic buffer enhanced cell killing appreciably following either treatment.
