ABSTRACT β-1,4-glucosaminoglucan (GG) was prepared from the sheath of a sulfur-oxidizing bacterium Thiothrix nivea. Recently, GG was found to be adsorbed by cellulose (paper) and is therefore potentially applicable as an aminating agent for cellulose. We attempted to increase the yield of GG using a fed-batch cultivation method. Furthermore, the behavior of GG molecules in water was theoretically and experimentally investigated. NMR analysis in combination with molecular dynamics calculation suggested that GG molecules tend to form soluble aggregates in water. It was experimentally revealed that the self-aggregation is enhanced by the addition of NaCl and reduced temperature. Adsorption of GG onto cellulose via hydrogen bonding was confirmed by molecular dynamics simulation. Adsorption was also promoted in the presence of NaCl but was inhibited by a reduction in temperature. Only 11% of the amino groups in the GG-treated paper was reactive, suggesting that GG molecules adsorbed by the paper were forming aggregates. 相似文献

Crayfish swimmeret system shows rhythmic, coordinated behavior when the command fibers are stimulated chronically by electrical pulses, and the oscillating frequency becomes faster with increasing stimulus frequency. This behavior is organized by the distributed neural oscillators in the abdominal ganglia. We investigated the dynamics of the neural oscillators which are controlled by command fibers. Phase resetting experiment technique was used for this purpose; a temporary cessation of commanding pulses, which was regarded as suppressive perturbation for the neural oscillator, was applied to the chronically stimulated oscillator, and phase transition curves (PTCs) were measured. For the short cessation of command pulses, type 1 PTCs were obtained. With increasing cessation length, the PTC shifted downward, and finally changed into type 0. We also measured PTCs for temporarily increased stimulus frequency, which was an excitatory perturbation for the neural oscillator and increased the frequency of the oscillation transiently. For the short excitatory perturbation, the PTCs were also type 1 and shifted upward. PTCs changed their shapes from type 1 into type 0, as increasing the perturbation length. These shapes of the PTCs contain important information about the properties of the neural oscillator. Four stereoisomers of 4-methyl-1-[N2-(3-methyl-1,2,3,4-tetrahydro-8-quinolinesulfonyl)-L-arginyl]-2-piperidinecarboxylic acid were synthesized and examined for the inhibitory effect on thrombin. The inhibitory potency varied largely with the stereo-configuration of the 4-methyl-2-piperidinecarboxylic acid portion. The (2R, 4R)-isomer was the most potent inhibitor with a Ki of 0.019 μM, while the (2R, 4S) and (2S, 4R)-isomers showed the values of Ki 0.24 and 1.9 μM, respectively. The least potent inhibitor, (2S, 4S)-isomer, showed a Ki of 280 μM which is approximately 15,000 times that of (2R, 4R)-isomer. 相似文献
