Fucoxanthin chlorophyll a/c-binding protein (FCP) is a unique light-harvesting apparatus in diatoms. Several biochemical characteristics of FCP oligomer and trimer from different diatom species have been reported previously. However, the integration of information about molecular organizations and polypeptides of FCP through a comparison among diatoms has not been published. In this study, we used two-dimensional clear-native/SDS-PAGE to compare the oligomeric states and polypeptide compositions of FCP complexes from four diatoms: Chaetoceros gracilis, Thalassiosira pseudonana, Cyclotella meneghiniana, and Phaeodactylum tricornutum. FCP oligomer was found in C. gracilis, T. pseudonana, and C. meneghiniana, but not in P. tricornutum. The oligomerization varied among the three diatoms, although a predominant subunit having similar molecular weight was recovered in each FCP oligomer. These results suggest that the predominant subunit is involved in the formation of high FCP oligomerization in each diatom. In contrast, FCP trimer was found in all the diatoms. The trimerizations were quite similar, whereas the polypeptide compositions were markedly different. On the basis of this information and that from mass spectrometric analyses, the gene products in each FCP complex were identified in T. pseudonana and P. tricornutum. The cyprinid species of the genus Gnathopogon, exhibiting flexible morphological and ecological variation, include limnetic life forms. We examined the origin of the limnetic forms and the population divergence of the Japanese Gnathopogon species, using molecular phylogenetic and phylogeographic analyses. A Bayesian phylogenetic inference approach based on mtDNA cytochrome b sequence data revealed three major lineages in G. elongatus. One of them formed a monophyletic group with the limnetic species G. caerulescens, which is endemic to an ancient lake, Lake Biwa. The divergence of the G. caerulescens lineage was estimated to date back to the early Pleistocene. This precedes the formation of the extensive pelagic environment in the present Lake Biwa. However, the recent genetic divergence of G. caerulescens was inferred to originate in the present Lake Biwa in the late Pleistocene. Another lacustrine population in the Mikata Lakes was shown to belong to a different lineage from G. caerulescens. The majority of the population possessed unique, but non-monophyletic, haplotypes, suggesting a short evolutionary history. One of the cryptic lineages of G. elongatus discovered in the Ina Valley, the lower area of Lake Suwa, might be related to the extinct lacustrine subspecies G. elongatus suwae, which has been replaced by introduced congeners. The previous and ongoing introductions of Gnathopogon fishes would have produced genetic disturbance to the indigenous populations. 相似文献

Intact cells of Kluyvera citrophila KY 3641 produced enzymaticaily d(—)-α-aminobenzyl-penicillin from 6-arainopenicillanic acid and phenylglycine derivatives. The optimum pH of the acylase was 6.5. Among various phenylglycine derivatives examined as substrates, d-phenylglycine methylester HC1 was the best compound giving the yields of about 10.7 mg/ml of d(—)-α-aminobenzy]penicillin in the enzymic reaction mixture. Spinach monogalactosyldiacylglycerol (MGDG) and digalactosyldiacylglycerol (DGDG) were oxidized with singlet molecular oxygen by the use of chlorophyll a as the photosensitizer. The oxidation products were separated from the unoxidized MGDG and DGDG by reverse-phase high performance liquid chromatography (HPLC). The products separated by HPLC were identified to be mono- and di-hydroperoxides formed by 1O2 oxidation of the 16:3 or 18:3 component of MGDG and DGDG. Endogenous auxins in the shoots and ears of rice were investigated. Indole-3-acetic acid (IAA) and indole-3-carboxylic acid (ICA) was identified by GC-MS, the endogenous level of IAA being much higher than that of ICA. To analyze the fluctuation of endogenous IAA level throughout the life cycle of rice, a rapid and effective procedure, using HPLC and a fluorescence detector, was developed. The level of IAA in shoots was 10 —26ng/g fr. wt., while that in ears was 10 to 100 times higher. The level of IAA conjugate in ears was also much higher than that in shoots. These results show that the biosynthesis of IAA occurs at an early stage of seed development, and it is also suggested that IAA may play a role in regulating the reproductive growth of rice. 相似文献
