Cercariae of many digenean trematodes target particular developmental stages of their hosts. For some digeneans that are parasites of amphibians, infection appears timed to host metamorphosis. The success and timing of metamorphosis is itself affected by a number of factors, including host density. We investigated the degree to which rearing density of Rana pipiens larvae influenced time to metamorphosis and snout-to-vent length and mass at metamorphosis, as well as establishment of cercariae of the trematode Manodistomum syntomentera Stafford, 1905. As expected, individuals metamorphosed later, were smaller, and weighed less at metamorphosis if they were reared under intermediate to high densities compared with low densities, in experimental outdoor mesocosms. Cercariae establishment was higher in smaller metamorphs that took longer to metamorphose within the low-density treatment. Additionally, cercariae establishment was lower in larvae from the low-density tanks compared with larvae from the intermediate- to high-density tanks. However, more tadpoles had failed to metamorphose in the intermediate to high rearing densities by the time cercariae were no longer available from natural collections of first intermediate hosts, Physa spp. Larval amphibians under crowded conditions should experience increased susceptibility to trematode establishment in nature, but only if they metamorphose within the time period when cercariae are still available.  相似文献

Background

As part of a formal evaluation of the Quantiferon-Gold in-tube assay (QFT-IT) for latent TB infection we compared its sensitivity to the tuberculin skin test (TST) in confirmed adult TB cases in Indonesia. Smear-positive TB disease was used as a proxy gold standard for latent TB infection.

Methods and Findings

We compared the sensitivity of QFT-IT and TST in 98 sputum smear and chest x-ray positive TB cases and investigated risk factors for negative and discordant results in both tests. Both tests showed high sensitivity; (QFT-IT; 88.7%: TST; 94.9%), not significantly different from each other (p value 0.11). Very high sensitivity was seen when tests were combined (98.9%). There were no variables significantly associated with discordant results or with a negative TST. For QFT-IT which particular staff member collected blood was significantly associated with test positivity (p value 0.01). Study limitations include small sample size and lack of culture confirmation or HIV test results.

Conclusions

The QFT-IT has similar sensitivity in Indonesian TB cases as in other locations. However, QFT-IT, like the TST cannot distinguish active TB disease from LTBI. In countries such as Indonesia, with high background rates of LTBI, test specificity for TB disease will likely be low. While our study was not designed to evaluate the QFT-IT in the diagnosis of active TB disease in TB suspects, the data suggest that a combination of TST and QFT-IT may prove useful for ruling out TB disease. Further research is required to explore the clinical role of QFT-IT in combination with other TB diagnostic tests.  相似文献

The electronic properties of the Mn(4)O(x)Ca cluster in the S(2) state of the oxygen-evolving complex (OEC) were studied using X- and Q-band EPR and Q-band (55)Mn-ENDOR using photosystem II preparations isolated from the thermophilic cyanobacterium T. elongatus and higher plants (spinach). The data presented here show that there is very little difference between the two species. Specifically it is shown that: (i) only small changes are seen in the fitted isotropic hyperfine values, suggesting that there is no significant difference in the overall spin distribution (electronic coupling scheme) between the two species; (ii) the inferred fine-structure tensor of the only Mn(III) ion in the cluster is of the same magnitude and geometry for both species types, suggesting that the Mn(III) ion has the same coordination sphere in both sample preparations; and (iii) the data from both species are consistent with only one structural model available in the literature, namely the Siegbahn structure [Siegbahn, P. E. M. Accounts Chem. Res.2009, 42, 1871-1880, Pantazis, D. A. et al., Phys. Chem. Chem. Phys.2009, 11, 6788-6798]. These measurements were made in the presence of methanol because it confers favorable magnetic relaxation properties to the cluster that facilitate pulse-EPR techniques. In the absence of methanol the separation of the ground state and the first excited state of the spin system is smaller. For cyanobacteria this effect is minor but in plant PS II it leads to a break-down of the S(T)=? spin model of the S(2) state. This suggests that the methanol-OEC interaction is species dependent. It is proposed that the effect of small organic solvents on the electronic structure of the cluster is to change the coupling between the outer Mn (Mn(A)) and the other three Mn ions that form the trimeric part of the cluster (Mn(B), Mn(C), Mn(D)), by perturbing the linking bis-μ-oxo bridge. The flexibility of this bridging unit is discussed with regard to the mechanism of O-O bond formation.  相似文献

Low levels of dissolved oxygen (DO) are potential stressors of fishes and invertebrates in freshwater and marine systems. Because of the incredibly nutrient-rich and warm nature of the Atchafalaya River Basin (ARB), microbial decomposition in backwater areas with limited flow often results in potentially stressful, if not lethal, DO levels during and after the annual flood pulse. As water temperatures increase and flood levels fall, DO levels often stratify, restricting fish habitat. In order to better guide development of water management projects, we investigated the relationship between DO and DO stratification (DOD; surface DO − bottom DO) and a suite of environmental variables potentially linked to DO and DOD. Based on continuous monitoring data collected from 2000 to 2007, we used a variable selection procedure to construct statistical generalized linear models to describe the relationship between DO levels and DOD and several potentially important predictor variables at three spatial scales: the entire sampled area of the ARB; individual water management units (WMUs) or subunits (WMSs); and individual sampling sites. We conducted these analyses using models that assume the normal distribution and compared them to models that incorporate alternate error terms based on other error term distributions. Goodness of fit statistics suggested that our analyses best described the relationship between DO and structuring variables at the WMU/WMS scale. At this scale, water temperature, river stage height, current velocity, and to a lesser extent, water depth appeared to be the most important measured variables that influence DO levels. We suggest that management decisions at the WMU and WMS scales will offer the greatest potential benefits for improvement of DO conditions in the ARB.  相似文献

The early post-fire plant succession in fynbos vegetation in the Mediterranean-type climate area of South Africa was studied. Relatively little has been published on this early stage of plant succession in fynbos. Annual sampling over the first three post-fire years confirmed a steady, but relatively slow increase in plant canopy cover of shrubs and graminoids (mainly Restionaceae), whereas cover of geophytes and other herbs peaked in the first year and declined significantly, thereafter. Cover of annual plants increased each year, which may relate to the persistence of a relatively open vegetation cover by the third year. The responses of reseeder and resprouter species of the Restionaceae to the post-fire environment appeared to be habitat dependent. Cover of the reseeders increased rapidly in seep areas, but their recovery was distinctly delayed in dryland areas outside the seeps. Re-establishment of the many reseeder Erica species appeared to be delayed until the second post-fire year. Seed banks of these species were possibly negatively impacted by the fire, and required dispersal of seed from unburnt areas for recruitment. In contrast to some current generalisations, species richness appeared to increase after the fire; less certainly from the first to the second year, but more certainly from the second to the third year. Therefore, this study does not support a short-term monotonic decline in species richness after fire in fynbos.  相似文献

Cochlear inner hair cells (IHCs) develop from pre‐sensory pacemaker to sound transducer. Here, we report that this involves changes in structure and function of the ribbon synapses between IHCs and spiral ganglion neurons (SGNs) around hearing onset in mice. As synapses matured they changed from holding several small presynaptic active zones (AZs) and apposed postsynaptic densities (PSDs) to one large AZ/PSD complex per SGN bouton. After the onset of hearing (i) IHCs had fewer and larger ribbons; (ii) CaV1.3 channels formed stripe‐like clusters rather than the smaller and round clusters at immature AZs; (iii) extrasynaptic CaV1.3‐channels were selectively reduced, (iv) the intrinsic Ca2+ dependence of fast exocytosis probed by Ca2+ uncaging remained unchanged but (v) the apparent Ca2+ dependence of exocytosis linearized, when assessed by progressive dihydropyridine block of Ca2+ influx. Biophysical modeling of exocytosis at mature and immature AZ topographies suggests that Ca2+ influx through an individual channel dominates the [Ca2+] driving exocytosis at each mature release site. We conclude that IHC synapses undergo major developmental refinements, resulting in tighter spatial coupling between Ca2+ influx and exocytosis.  相似文献
