The localization of glycogen phosphorylase was studied during the differentiation of prespore and prestalk cells in Dictyostelium discoideum. Ultramicrotechniques were utilized to assay the enzyme activity in cell samples as small as 0.02 μg dry wt in reaction volumes of 0.1 μl. The activity was assayed using an amplification procedure employing the enzymatic cycling of pyridine nucleotides. Glycogen phosphorylase from individual organisms was assayed during the developmental period. Early in development, activity was low but gradually increased to a maximum value at culmination. From culmination to sorocarp, enzyme activity decreased rapidly. Cell-specific assays of spores showed that phosphorylase activity increased slightly to culmination, and then decreased. Prestalk cells showed the greatest activity in the area of stalk sheath construction and elongation. Stalk cells showed a decreasing gradient of enzyme activity from the tip of the stalk to the base. Enzyme activity in the spores may be sufficient to provide glucose units for trehalose synthesis and spore coat production. The prestalk enzyme may degrade glycogen to provide glycosyl units for production of the stalk sheath and trehalose. Possible models of cell-specific biochemical events in Dictyostelium discoideum are discussed.  相似文献

We studied methods for the automatic segmentation of neonatal and developing brain images into 50 anatomical regions, utilizing a new set of manually segmented magnetic resonance (MR) images from 5 term-born and 15 preterm infants imaged at term corrected age called ALBERTs. Two methods were compared: individual registrations with label propagation and fusion; and template based registration with propagation of a maximum probability neonatal ALBERT (MPNA). In both cases we evaluated the performance of different neonatal atlases and MPNA, and the approaches were compared with the manual segmentations by means of the Dice overlap coefficient. Dice values, averaged across regions, were 0.81±0.02 using label propagation and fusion for the preterm population, and 0.81±0.02 using the single registration of a MPNA for the term population. Segmentations of 36 further unsegmented target images of developing brains yielded visibly high-quality results. This registration approach allows the rapid construction of automatically labeled age-specific brain atlases for neonates and the developing brain.  相似文献

Nitric oxide (NO) is an important mediator in both health and disease. In addition to its effects on vascular tone and platelet function, it plays roles in inflammation and pain perception that may be of relevance in osteoarthritis. Many patients with osteoarthritis take nonsteroidal anti-inflammatory drugs (NSAIDs) long term for pain control. Over recent years concern has been raised about the possible cardiovascular side effects of NSAIDs. The reasons for this possible increased cardiovascular risk with NSAIDs are not yet entirely clear, although changes in blood pressure, renal salt handling and platelet function may contribute. Recently, drugs that chemically link a NSAID with a NO donating moiety (cyclo-oxygenase-inhibiting NO-donating drugs [CINODs]) were developed. NO is an important mediator of endothelial function, acting as a vasodilator and an inhibitor of platelet aggregation, and having anti-inflammatory properties. The potential benefits of CINODs include the combination of effective analgesic and anti-inflammatory actions with NO release, which might counterbalance any adverse cardiovascular effects of NSAIDs. Effects of CINODs in animal studies include inhibition of vasopressor responses, blood pressure reduction in hypertensive rats and inhibition of platelet aggregation. CINODs may also reduce ischemic damage to compromised myocardial tissue. In addition, endothelial dysfunction is a recognized feature of inflammatory arthritides, and therefore a drug that might provide slow release of NO to the vasculature while treating pain is an attractive prospect in these conditions. Further studies of the effects of CINODs in humans are required, but these agents represent a potential exciting advance in the management of osteoarthritis.  相似文献

1. Three beta-fructofuranosidases were separated by chromatography on a DEAE-cellulose column from the soluble protein extracted from dandelion (Taraxacum officinale Weber) roots. 2. One enzyme, which acted on sucrose, was characterized as an invertase, with a K(m) of 2.00x10(-2)m and pH optimum of 7.5. 3. The other two enzymes are hydrolases (A and B), which act on the inulin series of oligosaccharides [general formula glucose-fructose-(fructose)(n)]. They both have a pH optimum of 4.0 and K(m) of 1.54x10(-2)m but differ in their chromatographic behaviour on DEAE-cellulose. Neither of the hydrolases is inhibited by sucrose. 4. The physiological role of these three hydrolytic enzymes is discussed.  相似文献

Acute arterial obstruction may result not only from arterial injuries but also from thrombosis or embolism. The fate of the extremity is generally decided in the first few hours following the obstruction of the major artery. It is therefore essential that physicians should be able to recognize acute ischemia of the extremities and institute treatment which will prevent permanent damage and facilitate the reestablishment of normal circulation.Direct application of heat should be scrupulously avoided since the reduced blood flow is unable to supply increased metabolic demand. The extremity should be placed at or just below heart level so as to avoid both the ischemia of elevation and the edema of dependency. Pressure in the form of either encircling plaster or even compression bandages should be avoided and the extremity simply immobilized by a posterior splint. General supportive treatment is essential to maintain adequate pressure and composition of the arterial blood. In selected cases, agents to overcome vascular spasm or to prevent extension of intravascular thrombosis are indicated.  相似文献
