Cell envelope phospholipid changes in a moderate halophile during phenotypic adaptation to altered salinity and osmotic stress

Abstract The increased content of negatively-charged phospholipids in membranes of Vibrio costicola grown at high salinities is mediated by increased phospholipid synthesis of phosphatidylglycerol relative to phosphatidylethanolamine. This phenomenon provides a system for investigating the factors involved in triggering and controlling haloadaptation in this moderately halophilic bacterium. We review recent experiments, which show that when subjected to sudden increases in external salinity, V. costicola senses both the absolute NaCl concentration and the magnitude of the salt shift. We show that the latter is sensed at least in part via osmotic pressure effects, since shift-up into sucrose-containing media triggers comparable changes in growth and in phospholipid composition and synthesis.     

INCIDENTAL MORTALITY OF NORTHERN SEA LIONS IN SHELIKOF STRAIT, ALASKA

The incidental catch of northern sea lions ( Eumetopias jubatus ) in the walleye pollock ( Theragra chalcogramma ) joint-venture fishery in Shelikof Strait, Alaska, was studied during 1982–1984 to assess the nature and magnitude of the catch. Data were obtained by placing U.S. observers on foreign processing vessels. Dead sea lions recovered from trawl nets were counted, sexed and measured, teeth were removed for age determination by dental laminae; and stomach contents were analyzed. Although the fishery has continued to expand both in number of boats and estimated total catch (74,136 metric tons [t] in 1982 to 171,539 t in 1984), the estimated incidental catch of northern sea lions has declined (ranging from 958 to 1,436 in 1982, 216 to 324 in 1983 and 237 to 355 in 1984). Of the sea lions processed, 73 percent were caught between 2000 and 0500 h, probably during net retrieval. Most caught sea lions were females ranging in age from 1–25 yr with a mean age of 6.43 yr; 79 percent of the females were sexually mature and probably part of the reproducing population. Males had a mean age of 4.8 yr and only 12 percent were old enough to obtain and defend territories. Analysis of stomach contents showed that the sea lions consumed pollock the same size as that taken by the commercial fishery. The impact of the incidental catch on the Gulf of Alaska sea lion population is unknown.     

Relationship of seminal vesicle size and measures of libido in yearling beef bulls

Ninety-five yearling beef bulls were given routine Breeding Soundness Examinations, two libido tests and palpated rectally for internal genital disease and measurement of seminal vesicle size. Finger tips were premeasured, and then used as "glandometers". The bulls were examined at the end of a 140-day performance test. Sixteen lines of breeding were examined including 13 Hereford, 2 Angus and 1 Red Angus. Average age was 384 days and average weight was 961 lbs. Line and breed differences (P<.05) were observed for scrotal circumference, scrotal circumference score and second libido test. Similar differences were observed for SV length (P<.01), depth and volume both (P<.05). There were no significant correlations between seminal vesicle (SV) size and libido scores. There were, however, significant correlations between SV size and scrotal circumference (P<.05), BSE score (P<.01), body weight (P<.01), and semen morphology score (P<.05). Proximal droplets and midpiece abnormalities, respectively, were the most common spermatozoal abnormalities observed in semen from this group of bulls.     

Effect of Glutaraldehyde on the Outer Layers of Escherichia coli

S ummary : Sodium lauryl sulphate (SLS) at pH 3 and 8 lysed cell walls of Escherichia coli. Pretreatment with glutaraldehyde at pH 3 and at pH 8 prevented this lysis. SLS induced maximum lysis of E. coli cells at 40°; pretreatment of cells with glutaraldehyde prevented this lysis also. Electrophoretic studies indicated that glutaraldehyde accumu lated on the surface of E. coli cells more rapidly in acid than in alkaline conditions, and that it blocked amino groups on the surface layer of Bacillus subtilis spores. The relationship of these findings to the bactericidal efficiency of glutaraldehyde in acid and alkaline solution is discussed.
