A procedure is described which unequivocally demonstrates the presence of adenosine 3′:5′-cyclic monophosphate in Phaseolus vulgaris. Its concentration was determined spectrophotometrically at 2·6–9·2 nmol g?1 of tissue (dry wt) for 6-day-old seedlings and about one-tenth of this in 13-day-old plants.  相似文献

Adult female white-footed mice, Peromyscus leucopus, were exposed to long (LP) or short (SP) photoperiods for 6 weeks (experiment I). Another group of animals was kept for 6 weeks in SP, then injected SC with 30 μg prolactin twice daily for 2, 3, 4, or 6 days (experiment II). Ovaries from the mice in both experiments were weighed and serially sectioned for light microscopic examination of regressing corpora lutea. In experiment I, it was observed that vessels supporting corpora lutea were dilated, and that their endothelium was either undergoing necrosis or it was missing. Pronounced changes of luteal capillaries led to rupture and intraluteal hemorrhage, thus opening the capillary bed. Regressing luteal cells became segregated and seemed to invade the vascular system passively. They were seen as luteal cell thrombi in medullary veins. This luteolytic course termed “rapid luteolysis” was most apparent in SP ovaries. It differed from “retarded luteolysis,” which represents the well-established luteolytic model of auto- and heterophagocytosis. In experiment II, there was a statistically significant decrease in ovarian weight 4 days after prolactin treatment in comparison with saline-treated controls. At the light microscopic level, signs of both rapid and retarded luteolysis were present, but not intensified. It is concluded: (1) The concept of rapid luteolysis represents a reasonable working hypothesis. (2) Prolactin, though luteolytic at the macroscopic level, failed to produce evidence of increased rapid or retarded luteolysis at the light microscopic level.  相似文献

To determine whether the fip gene of Escherichia coli, which is required for filamentous phage assembly, is required for cell viability, we replaced the chromosomal copy of the gene with an inactive copy introduced on a plasmid. We found that the fip gene is dispensable. The method we devised, which should be generally useful, was also tested with an inactivated rho gene. As expected, the rho gene is essential.  相似文献

Some Perturbations That Disturb the Circadian Melatonin Rhythm   总被引:3,自引:0,他引:3  
The circadian melatonin rhythm is highly reproducible and generally not easily altered. The few perturbations that are capable of significantly changing either the amplitude or the pattern of the 24-h melatonin rhythm are summarized herein. Aging alters cyclic melatonin production by decreasing the amplitude of the nocturnal melatonin peak in all species in which it has been studied. The best known acute suppressor of nocturnal melatonin is light exposure. The brightness of light required to acutely depress pineal melatonin production is species dependent; of the visible wavelengths, those in the blue range (∼500-520 nm) seem most effective in suppressing melatonin production. Nonvisible, nonionizing radiation in the extremely low frequency range (e.g., 60 Hz) seems also capable of altering pineal melatonin synthesis. Hormones have relatively little influence on the circadian production of melatonin, although either adrenalectomy or hypo-physectomy does attenuate the amplitude of the melatonin cycle. Exercise at the time of high melatonin production rapidly depresses pineal concentrations of the indole without influencing its synthesis; the mechanism of this suppression remains unknown.  相似文献

The structure and crystallography of the internal shell of the pulmonate gastropod slug Limax maximus were studied at the levels of light and scanning electron microscopy, revealing patterns of shell ontogeny and morphogenesis. The calcified portion of the slightly convex ovoid shell is composed of a single palisade layer of calcitic crystals. Numerous projections, 100 μm in width at the dorsal tip, are found on the dorsal surface of the shell and coincide with local nucleation sites of primordial calcium salt deposition onto the periostracum. With continued calcification these projections coalesce ventrally, forming the single crystalline shell layer. The organic portion of the shell includes the periostracum and an extensive PAS-staining conchiolin. In EDTA-etched preparations, conchiolin appears as a spongy network of fibers throughout the shell. Both horizontal and vertical components of the conchiolin are present, the former of variable thickness and occurring in an intercrystalline manner, the latter always occurring normal to the horizontal set. Macromorphogenic growth is characterized by three distinct temporal stages. Primary growth occurs radially from the umbonal region. Secondary growth is synonymous with shell thickening. Tertiary growth is characterized by both a lateral component, in which the shell extends beyond the primary growth boundaries, and a ventral component, in which the shell continues to grow in thickness. SEM of the ventral shell surface reveals a pattern of growth at the crystalmatrix interface. Proteinaceous fibers of the conchiolin occur unidirectionally in horizontal rows. Zones of incipient calcitic crystallization onto these hypostracal fiber bundles are contrasted by zones of increasing crystallization until the fibrous template (reduced hypostracum) is completely covered by crystals.  相似文献
