A Ralstonia pickettii species able to degrade chlorobenzene (CB) as the sole source of carbon and energy was isolated from a biotrickling filter used for the removal of CB from waste gases. This organism, strain L2, could degrade CB as high as 220 mg/L completely. Following CB consumption, stoichiometric amounts of chloride were released, and CO2 production rate up to 80.2% proved that the loss of CB was mainly via mineralization and incorporation into cell material. The Haldane modification of the Monod equation adequately described the relationship between the specific growth rate and substrate concentration. The maximum specific growth rate and yield coefficient were 0.26 h−1 and 0.26 mg of biomass produced/mg of CB consumed, respectively. The pathways for CB degradation were proposed by the identification of metabolites and assay of ring cleavage enzymes in cell extracts. CB was degraded predominantly via 2-chlorophenol to 3-chlorocatechol and also partially via phenol to catechol with subsequent ortho ring cleavage, suggesting partially new pathways for CB-utilizing bacteria.  相似文献

The heterotrimeric G protein subunit Gsα couples receptors to activate adenylyl cyclase and is required for the intracellular cAMP response and protein kinase A (PKA) activation. Gsα is ubiquitously expressed in many cell types; however, the role of Gsα in neural crest cells (NCCs) remains unclear. Here we report that NCCs-specific Gsα knockout mice die within hours after birth and exhibit dramatic craniofacial malformations, including hypoplastic maxilla and mandible, cleft palate and craniofacial skeleton defects. Histological and anatomical analysis reveal that the cleft palate in Gsα knockout mice is a secondary defect resulting from craniofacial skeleton deficiencies. In Gsα knockout mice, the morphologies of NCCs-derived cranial nerves are normal, but the development of dorsal root and sympathetic ganglia are impaired. Furthermore, loss of Gsα in NCCs does not affect cranial NCCs migration or cell proliferation, but significantly accelerate osteochondrogenic differentiation. Taken together, our study suggests that Gsα is required for neural crest cells-derived craniofacial development.  相似文献

在农业景观中,传粉昆虫的生存繁衍与半自然生境的面积大小有关。集约化生产方式使半自然生境比例逐渐减少,农田比例不断增加,随着景观简化梯度的变化(农田比例逐渐增大),传粉昆虫群落多样性将会发生怎样的变化?选择黄河中下游典型农区巩义市为研究区域,采用诱捕盘法(Pan traps)进行农田、林地的传粉昆虫取样,以21个样点作为景观简化梯度(农田比例范围5%—86%)的呈现,基于每个样点的传粉昆虫多度和丰富度变化来探究景观简化对传粉昆虫多样性的影响。结果显示:区内累计捕获传粉昆虫39660头,优势类群包括双翅目(Diptera)、膜翅目(Hymenoptera)、鞘翅目(Coleoptera)等。采用逐步回归分析及线性拟合后发现景观简化程度与传粉昆虫多度和丰富度呈显著负相关(P0.05);景观简化对传粉昆虫类群间的影响也是有差异的,其中对鞘翅目多度的影响最为密切(R~2=0.27),同时对膜翅目和双翅目也有较大影响(R~2=0.14、R~2=0.11),景观简化与鳞翅目多度呈正相关;随景观简化的程度加深,农田生境中膜翅目多度呈显著下降趋势(P0.05),而林地中膜翅目多度变化不明显。在未来的景观规划中,应着重考虑传粉昆虫中鞘翅目类群的栖息地变化及食物资源状况。依据研究结果建议林地生境中应注重保护现有的自然植被群落,在人工林中可以种植一定面积的蜜粉源植物;农田生境内对杂草群落、半自然生境斑块进行合理规划的基础上,还可以种植线性景观植物作为传粉昆虫的食物源。  相似文献

2000年3月-2001年2月对黄河三角洲盐碱土壤中5种优势植物柽柳(Tamarix chinensis),芦苇(Phragmites communis),碱蓬(Suaeda glauca),獐毛(Aeluropus littralis var.sinensis) 和刺儿菜(Cirsium setosum)根围内丛枝菌根(arbuscular mycorrhiza,AM)真菌进行了长期定点调查,结果表明,AM真菌在这5种植物根围土壤内都有分布,但多样性较低,尤其在冬季,AM真菌种的丰度和孢子密度分别在夏季降低了18.0%和61.6%,不同深度土层中AM真菌的分布存在诸多差异。  相似文献

作者以界面铺张-硝酸银染色技术,对东北马鹿和东北梅花鹿的F_1可育杂种的精母细胞联会复合体进行亚显微观察及分析。在减数分裂前期,杂种鹿精母细胞中形成31条完整的常染色体联会复合体、一个端着丝粒染色体/中着丝粒染色体的三价体和XY双价体。这进一步证明,两种亲本鹿的染色体具有高度的同源性,其差别仅在于一个罗伯逊易位。三价体的顺式构型可能和杂种鹿的可育性有关。  相似文献

Long noncoding RNAs (lncRNAs) play roles in the development and progression of many cancers; however, the contributions of lncRNAs to human gallbladder cancer (GBC) remain largely unknown. In this study, we identify a group of differentially expressed lncRNAs in human GBC tissues, including prognosis‐associated gallbladder cancer lncRNA (lncRNA‐PAGBC), which we find to be an independent prognostic marker in GBC. Functional analysis indicates that lncRNA‐PAGBC promotes tumour growth and metastasis of GBC cells. More importantly, as a competitive endogenous RNA (ceRNA), lncRNA‐PAGBC competitively binds to the tumour suppressive microRNAs miR‐133b and miR‐511. This competitive role of lncRNA‐PAGBC is required for its ability to promote tumour growth and metastasis and to activate the AKT/mTOR pathway. Moreover, lncRNA‐PAGBC interacts with polyadenylate binding protein cytoplasmic 1 (PABPC1) and is stabilized by this interaction. This work provides novel insight on the molecular pathogenesis of GBC.  相似文献

利用光学显微镜和扫描电子显微镜对山西霍山罂粟科白屈菜属白屈菜、紫堇属延胡索、角茴香属角茴香、秃疮花属秃疮花 4种的花粉形态进行了观察和比较研究 ,以上 4种花粉差异大。角茴香花粉粒近球形 ,具合沟 ;延胡索花粉粒长球形 ,表面纹饰较光滑 ;白屈菜花粉粒长球形或近球形 ;秃疮花花粉粒近球形。角茴香花粉表面纹饰为刺状纹和细网状纹的复合纹 ,白屈菜花粉表面纹饰为细网状纹和颗粒状纹的复合纹 ,秃疮花花粉表面纹饰为细颗粒状纹。延胡索、白屈菜、秃疮花花粉粒的萌发沟均为三沟 ,但沟的形态均不相同  相似文献

A group of 224 recombinant inbred lines (RILs) was derived from a narrow cross between 2 cucumber (Cucumis sativus L.) lines, namely, S94 (Northern China type with weak lateral branch growth potential and early lateral branch sprouting time) and S06 (Northern European type with strong lateral branch growth potential and late lateral branch sprouting time). These lines were then used for investigating lateral branch-related traits. A total of 36 quantitative trait loci (QTLs) were detected for the following 4 lateral branch-related traits: lateral branch average length (LBAL), lateral branch total length (LBTL), lateral branch number (LBN), and first lateral branch node (FLBN). Further, each QTL explained 3.1% (lbtl2.1, spring) to 32.3% (lbn2.3, spring) of the observed phenotypic variance. Eleven QTLs (lbal1.1, lbtl1.1, lbn1.2, flbn1.2, etc.) for different traits were found to be clustered on the e23m18d-ME23EM6c section (7.4 cM) of linkage group (LG) 1; further, 15 QTLs (lbal2.1, lbtl2.1, lbn2.1, flbn2.1, etc.) were found to be clustered on the S94A1-ME4SA4a section (13.9 cM) of LG2. Twenty-one QTLs explained more than 10% of the phenotypic variance. Moreover, lbtl1.3 (autumn, 26.2%, logarithm of odds (LOD) = 17.4; spring, 26.9%, LOD = 17.9) had stable position and contribution in both seasons. Several sequence-anchor markers (CMBR40, F, CS30, S94A1, CSWTA11B, etc.) were closely linked with some QTLs for LBAL, LBTL, LBN, and FLBN, which can be used for the marker-assisted selection to improve the plant architecture in cucumber breeding.  相似文献

Phosphorylation of the cAMP response element binding protein (CREB) by cAMP-dependent kinase (PKA) is critical to memory formation. However, activation of PKA can also increase tau phosphorylation, which may contribute to memory impairment. Therefore, the regulation of PKA may be part of the mechanism by which glucocorticoids (GCs) influence memory. Additionally, the cellular response to GCs may be affected by the presence of human tau. The goal of this paper was to study GCs-mediated regulation of PKA as well as CREB and tau phosphorylation in wild-type HEK293 cells and HEK293 cells stably expressing human tau441 (HEK293/tau441 cells). By using dexamethasone (DEX) as GCs, we found that DEX induced a tau-dependent selective decrease in the level of PKA RIIβ subunit protein. The observed decrease in RIIβ expression was not due to alterations of mRNA levels and was reversed by inhibiting the proteasome with lactacystin. Moreover, the decrease in RIIβ did not diminish the co-localization of the catalytic subunit of PKA with tau and might contribute to the DEX-induced increase in tau phosphorylation at Ser-214. DEX also induced a tau-dependent decrease in CREB phosphorylation that could not be reversed by activating PKA with forskolin. Taken together, these results show that human tau protein may alter the GCs-mediated regulation of PKA activity and CREB phosphorylation.  相似文献

Dietary shifts can alter the relative availability of different nutrients and are therefore associated with metabolic adaptation in animals. The Coccinellidae (ladybirds) exhibits three major types of feeding habits and provides a useful model to study the effects of dietary changes on the evolution of mitogenomes, which encode proteins directly involved in energy metabolism. Here, mitogenomes of three coccinellid species were newly sequenced. These data were combined with other ten previously sequenced coccinellid mitogenomes to explore the relationship between mitogenome evolution and diets. Our results indicate that mitogenomic data can be effectively used to resolve phylogenetic relationships of Coccinellidae. Strong codon usage bias in coccinellid mitogenomes was predominantly determined by nucleotide composition. The 13 mitochondrial protein‐coding genes (PCGs) globally evolved under negative constraints, with some PCGs showing a stronger purifying selection. Six PCGs (nad3, nad4L, and nad5 from Complex I; cox1 and cox3 from Complex IV; and atp6 from Complex V) displayed signs of positive selection. Of these, adaptive changes in cox3 were potentially associated with metabolic differences resulting from dietary shifts in Coccinellidae. Our results provide insights into the adaptive evolution of coccinellid mitogenomes in response to both dietary shifts and other life history traits.  相似文献
