In laboratory research, the rainbow trout has become a counterpart to the white rat, because that fish is an adaptable species available in much of the developed world and stocks from egg through adult are available throughout the year. Moreover, many strains are recognized, and their propagation and laboratory maintenance are not particularly demanding. Also, knowledge of rainbow trout nutrition, husbandry, diseases, immune responses, toxicology, and carcinogenesis exceeds that of any other salmonid or coldwater teleost. Three satellite DNA families were identified in three species of burying
beetles, Nicrophorus orbicollis, N. marginatus, and N. americanus. Southern
hybridization and nucleotide sequence analysis of individual randomly
cloned repeats shows that these satellite DNA families are highly abundant
in the genome, are composed of unique repeats, and are species-specific.
The repeats do not have identifiable core elements or substructures that
are similar in all three families, and most interspecific sequence
similarity is confined to homopolymeric runs of A and T. Satellite DNA from
N. marginatus and N. americanus show single-base-pair indels among repeats,
but single-nucleotide substitutions characterize most of the repeat
variability. Although the repeat units are of similar lengths (342, 350,
and 354 bp) and A + T composition (65%, 71%, and 71%, respectively), the
average nucleotide divergence among sequenced repeats is very low (0.18%,
1.22%, and 0.71%, respectively). Transition/transversion ratios from the
consensus sequence are 0.20, 0.69, and 0.70, respectively.

Differential migration is a widespread, but poorly understood, phenomenon in birds. In this paper, we present the first detailed field study of differential migration in the Old World warbler (Sylviidae) family. We studied two chiffchaff Phylloscopus [collybita] semispecies: the common chiffchaff P. [c.] collybita and the Iberian chiffchaff P. [c.] ibericus. Using data collected at several latitudes in Europe and Africa, we present convincing evidence for differential distance migration of sexes in chiffchaffs, with females moving further than males. Interestingly, while there was a pronounced gradient in the sex‐ratios in Europe and North Africa (with an increasing proportion of females with declining latitude), no clear pattern was found south of the Sahara, where sex‐ratios were more male‐biased than predicted by a simple latitude model. This suggests that, amongst the chiffchaffs wintering in West Africa, a large proportion is composed by Iberian birds, and provides support to previous suggestions that Iberian chiffchaffs are long distance migrants. Results from detailed studies in Senegal also show that chiffchaffs display differential timing of spring migration, with males leaving the winter quarters considerably earlier than females. The results are discussed in the framework of the three main (non‐mutually exclusive) hypotheses attempting to explain the latitudinal segregation of the sexes. Given the relative failure of standard comparative studies to discriminate between competing single‐factor hypotheses to explain differential migration, it is argued that the chiffchaff species complex might be particularly suited to study this issue using a new approach suggested by Cristol et al. (1999) : detailed (further) comparisons between closely related species (such as the common and the Iberian chiffchaffs) could help identifying the key factors to be incorporated into optimality models that can predict relative distance of migration of different sex or age classes. 相似文献

Energetics of the structure and chain tilting of antiparallel beta-barrels in proteins 总被引:3,自引:0,他引:3
The preferred structural pattern of antiparallel beta-barrels in proteins, described as the right-handed tilting of the peptide strands with respect to the axis of the barrel, is accounted for in terms of intra- and interchain interaction energies. It is related to the preference of beta-sheets for right-handed twisting. Conformational energy computations have been carried out on three eight-stranded antiparallel beta-barrels composed of six-residue strands, in which L-Val and Gly alternate, and having a right-handed, a left-handed, or no tilt. After energy minimization, the relative energies of these structures were 0.0, 8.6, and 46.1 kcal/mol, respectively; i.e., the right-tilted beta-barrel is favored energetically, in agreement with anti-parallel beta-barrels observed in proteins. Tilting of the barrel is favored, relative to the nontilted structure, by both intra- and interstrand interactions, because tilting allows better packing of the bulky side chains. On the other hand, the energy difference between the left- and right-tilted barrels arises essentially from intrachain interactions. This is a consequence of the preference of beta-sheets for a right-handed twist. Space limitations inside the barrel are satisfied if there is an alternation of bulky residues and residues with small or no side chain (preferably Gly) in neighboring positions on adjacent strands. Such a pattern is seen frequently in antiparallel beta-barrels of globular proteins. The electrical properties of the frog taste cells during gustatory stimulations with distilled water and varying concentrations of NaCl were studied with intracellular microelectrodes. Under the Ringer adaptation of the tongue, two types of taste cells were distinguished by the gustatory stimuli. One type, termed NaCl-sensitive (NS) cells, responded to water with hyperpolarizations and responded to concentrated NaCl with depolarizations. In contrast, the other type of cells, termed water-sensitive (WS) cells, responded to water depolarizations and responded to concentrated NaCl with hyperpolarizations. The membrane resistance of both taste cell types increased during the hyperpolarizing receptor potentials and decreased during the depolarizing receptor potentials, Reversal potentials for the depolarizing and hyperpolarizing responses in each cell type were a few millivolts positive above the zero membrane potential. When the tongue was adapted with Na-free Ringer solution for 30 min, the amplitude of the depolarizing responses in the NS cells reduced to 50% of the control value under normal Ringer adaptation. On the basis of the present results, it is concluded (a) that the depolarizing responses of the NS and WS cells under the Ringer adaptation are produced by the permeability increase in some ions, mainly Na+ ions across the taste cell membranes, and (b) that the hyperpolarizing responses of both types of taste cells are produced by a decrease in the cell membrane permeability to some ions, probably Na+ ions, which is slightly enhanced during the Ringer adaptation. 相似文献

Mouse t haplotypes are variant forms of chromosome 17 that exist at high
frequencies in worldwide populations of two species of commensal mice. To
determine both the relationship of t haplotypes to each other and the
species within which they exist, 35 representative t haplotypes were
analyzed by means of 10 independent molecular probes, including five DNA
clones and five polypeptide spots identified by means of two- dimensional
gel electrophoresis. All of the tested haplotypes were found to share
restriction fragments and polypeptide spots that are absent in mice
carrying wild-type forms of chromosome 17. This observation provides the
first direct evidence that all of the known t haplotypes are descendents of
a single ancestral chromosome. The absence of variation among t haplotypes
could mean that this ancestral chromosome existed relatively recently, in
which case it would be necessary to postulate introgressions of t
haplotypes across species lines to explain their presence in both Mus
domesticus and M. musculus. Alternatively, it is possible that the
ancestral chromosome existed prior to the split between M. domesticus and
M. musculus and that, by chance, our probes fail to detect polymorphisms
that exist among the t haplotypes. A further result of our analysis is the
characterization of a partial t haplotype in a wild population of Israeli
mice.

The holoparasitic angiosperm Orobanche minor parasitizes a diverse range of flowering plants from at least 16 orders in both the monocots and eudicots. However, populations of O. minor show host specificity at a local level. Our previous work identified the potential for host specificity to act as a catalyst for genetic divergence among populations of O. minor. Here we have extended this investigation by sampling populations from multiple hosts, across a broad geographic range. Sequence characterised amplified region (SCAR) data identified an exotic host-generalist lineage and a native host-specialist lineage of O. minor, suggesting genetic structure in this species is defined by both host specificity and geography. In addition, host-range overlap, discordant tree topologies, and cryptic morphology indicate the presence of gene flow between alien races and endemic populations. Therefore, repeated introductions of alien races of O. minor from disparate sources leading to introgression with native populations, and cryptic race formation, seem to have contributed to the taxonomic confusion associated with this species. We speculate that radiations associated with broad host range and divergent host ecologies may have promoted the unusually wide geographic distribution and diversification of this species. Finally, evidence of multiple shifts to exotic hosts, coupled with the predicted northward shift in climatic suitability, identify the potential for range expansion in alien races of O. minor, which may threaten nationally scarce native taxa with genetic assimilation. Our phylogenetic analysis provides a framework for identifying host races in Orobanche with a view to setting conservation priorities. 相似文献

AIMS: Production of the mosquito biolarvacide Lagenidium giganteum in solid culture has been proposed as an economic alternative to production in liquid culture because of observations of improved shelf life and efficacy upon storage. Understanding the differences between these production systems and estimating growth rate in solid culture are important for commercialization. In order to address these needs a logistic model was developed to describe the growth kinetics of L. giganteum produced in solid and liquid cultures. METHODS AND RESULTS: Kinetic parameters in the logistic model were estimated by nonlinear regression of CO2 evolution rate (CER) and biomass data from solid and liquid cultivation experiments. Lagenidium giganteum biomass was measured using DNA extracted directly from samples. The logistic model was fit to experimental biomass and CER data with low standard errors for parameter estimates. The model was validated in two independent experiments by examining prediction of biomass using on-line CER measurements. CONCLUSIONS: There were significant differences between maximum biomass density, maintenance coefficients, and specific growth rates for liquid and solid cultures. The maximum biomass density (mg dw ml-1) was 11 times greater for solid cultivation compared with liquid cultivation of L. giganteum; however, the maintenance coefficient (mg CO2 h-1 (mg dw)-1) was six times greater for liquid cultivation than in solid cultivation. The specific growth rate at 30 degrees C was approximately 30% greater in liquid cultivation compared with solid cultivation. Slower depletion of substrate and lower endogenous metabolism may explain the longer shelf life of L. giganteum produced in solid culture. SIGNIFICANCE AND IMPACT OF THE STUDY: A simple logistic model was developed which allows real-time estimation of L. giganteum biomass from on-line CER measurements. Parameter estimates for liquid and solid cultivation models also elucidated observations of longer shelf life for production in solid culture. 相似文献
