Synopsis Aquatic and aerial oxygen uptake (̇O2), ventilation frequency, and oxygen transport properties of the blood were determined for the intertidal fish Helcogramma medium. Ventilation frequency increased in response to decreased environmental PO2 and aquatic ̇O2 was maintained down to a critical PO2 of 30–40 mm Hg. Below PO2 30 mm Hg fish intermittently gulped air and finally emerged into air at PO2 18 mm Hg. After 1 h exposure to air ̇O2 decreased to 60% of the aquatic rate and this was accompanied by an increase in blood lactate. Aerobic expansibility was reduced in air (×1.2) compared to water (× 5.5). The Hb concentration was 0.47 ± 0.13 mmol 1–1 and hematocrit 11.55 ± 3.61% indicating a moderate O2-carrying capacity. Oxygen affinity was not especially high (P50 = 19 mm Hg at pH 7.7 and 15°C) and ATP was the predominant acid-soluble phosphate regulating P50. The equilibrium curve was essentially hyperbolic (Hill's n = 1.2) with a marked Bohr effect = –1.06) and Root effect (saturation depressed by 50% at pH7.1). The pattern of respiration and the respiratory properties of the blood together with observations of the behaviour of the fish during aerial exposure indicated that Helcogramma is adapted to living in a well-aerated environment yet can adequately tolerate short term exposure to low aquatic PO2 or air. 相似文献

It is well known that alpha-chymotrypsin can exist in two major conformational states, only one of which is active. We have examined the pH (pH 2.0--11.0) and salt (ionic strength 0.01--1.0) dependence of the transition between the active and inactive forms in detail. At low pH (pH 2.0--6.0) the equilibrium is very dependent on salt concentration, with high salt concentrations effectively stabilizing the active conformation. This apparent stabilization is an artifact due to the salt-dependent dimerization of alpha-chymotrypsin, and our data show that only active species form dimers and higher aggregates. At neutral pH (6.0--8.0) dimerization is absent, yet an ionic strength dependence remains. The effects show no lyotropic order and appear to be due to preferential salt binding to the active conformation at one or possibly a few sites. Above pH 6 (pH 6.0--11.0), the pH dependence can be described by a two-ionization mechanism at all ionic strengths. We report values for all seven equilibrium constants in the proposed mechanism at four ionic strengths (mu = 0.01, 0.05, 0.2, and 1.0). The transition is the first "refolding" transition to be studied at high precision, but, even so, certain decisions about the mechanism must await higher experimental precision not available with present methods. 相似文献

Most attempts to model the evolution of parental care assume that caring and mating are mutually exclusive activities (i.e., individuals acquire and guard broods "sequentially"). However, in most fish and certain insects, males can keep mating and collecting additional eggs while continuing to guard broods obtained earlier (i.e., males guard "overlapping" broods). We present a model of parental care with overlapping broods in which males can mate and guard simultaneously, even though there is a trade-off between these two activities. Within this framework, we show that male care is favored by short female processing times and high population densities, which minimize the mating cost of care. Relatively low mortality while guarding is also important for the stability of male care. Female care, on the other hand, is favored by long female processing times and low populations densities, which lead to longer intermating intervals. Biparental care is stable only when the cost to benefit ratio of care was not biased toward either sex. We derive quantitative estimates of fitness for different strategies for two species of assassin bugs with male and female uniparental care and show that the model predicts the correct form of care for both species. We believe our model might help explain the prevalence of male uniparental care in certain taxa, such as fish. 相似文献

Endothelial cells are primary targets for pro-atherosclerotic stressors such as oxidized LDL (ox-LDL). The isoflavone genistein, on the other hand, is suggested to prevent a variety of processes underlying atherosclerosis and cardiovascular diseases. By analyzing the proteome of EA.hy 926 endothelial cells, here we show, that genistein reverses the ox-LDL-induced changes of the steady-state levels of several proteins involved in atherosclerosis. These alterations caused by genistein are functionally linked to the inhibition of ox-LDL induced apoptosis. 相似文献

A bioassay arena was developed to test the response of the Japanese beetle, Popillia japonica Newman (Coleoptera: Scarabaeidae), to various plant‐derived factors that may influence the selection of oviposition sites. In choice experiments with natural and artificial plant cues, females preferred to lay eggs in ryegrass rather than in artificial resources. Artificial grass stems received more eggs on average than bare ground, indicating the role of vertical objects on the soil surface as oviposition cues. Popillia japonica females were able to discriminate between the quality and quantity of the artificial stems, as more eggs were laid in soil blocks that had larger diameter stems or a higher number of stems per soil block. Beetles were capable of evaluating plant characteristics on the soil surface and adjusted their investment accordingly; more eggs were laid in resources that were chosen more frequently. Observations of females in choice arenas revealed how surface cues influence digging behavior and the time spent under the soil surface. The number of digging events was not significantly different among the three treatments with surface cues (ryegrass, grass roots, and artificial grass), but beetles spent significantly more time in the soil under ryegrass than any of the other treatments. These findings suggest that oviposition by P. japonica is selective and is influenced by plant‐derived cues evaluated before and after digging into the soil. 相似文献

1 The recent increase in planting of selected willow clones as energy crops for biomass production has resulted in a need to understand the relationship between commonly grown, clonally propagated genotypes and their pests. 2 For the first time, we present a study of the interactions of six willow clones and a previously unconsidered pest, the giant willow aphid Tuberolachnus salignus. 3 Tuberolachnus salignus alatae displayed no preference between the clones, but there was genetic variation in resistance between the clones; Q83 was the most resistant and led to the lowest reproductive performance in the aphid 4 Maternal effects buffered changes in aphid performance. On four tested willow clones fecundity of first generation aphids on the new host clone was intermediate to that of the second generation and that of the clone used to maintain the aphids in culture. 5 In the field, patterns of aphid infestation were highly variable between years, with the duration of attack being up to four times longer in 1999. In both years there was a significant effect of willow clone on the intensity of infestation. Benzo(a)pyrene diol-epoxide I (r-7,t-8,dihydroxy-t-9,10 oxy-7,8,9,10 tetrahydrobenzo(a)pyrene) was used to treat either human adenovirus 5 or cultures of human fibroblasts. The survival of diol-epoxide I treated adenovirus was greater when infecting fibroblasts from normal persons than when infecting fibroblasts from patients with xeroderma pigmentosum (XP). One diol-epoxide I molecule bound per viral genome correlated with one lethal hit as measured using XP fibroblasts.Normal fibroblasts blocked in semi-conservative DNA synthesis incorporated into their DNA more [3H]thymidine in response to diol-epoxide I treatment than did XP fibroblasts, and also excised more diol-epoxide I from their DNA. The physiological function of urate is poorly understood. It may act as a danger signal, an antioxidant, or a substrate for heme peroxidases. Whether it reacts sufficiently rapidly with lactoperoxidase (LPO) to act as a physiological substrate remains unknown. LPO is a mammalian peroxidase that plays a key role in the innate immune defense by oxidizing thiocyanate to the bactericidal and fungicidal agent hypothiocyanite. We now demonstrate that urate is a good substrate for bovine LPO. Urate was oxidized by LPO to produce the electrophilic intermediates dehydrourate and 5-hydroxyisourate, which decayed to allantoin. In the presence of superoxide, high yields of hydroperoxides were formed by LPO and urate. Using stopped-flow spectroscopy, we determined rate constants for the reaction of urate with compound I (k1 = 1.1 × 107
m−1 s−1) and compound II (k2 = 8.5 × 103
m−1 s−1). During urate oxidation, LPO was diverted from its peroxidase cycle because hydrogen peroxide reacted with compound II to give compound III. At physiologically relevant concentrations, urate competed effectively with thiocyanate, the main substrate of LPO for oxidation, and inhibited production of hypothiocyanite. Similarly, hypothiocyanite-dependent killing of Pseudomonas aeruginosa was inhibited by urate. Allantoin was present in human saliva and associated with the concentration of LPO. When hydrogen peroxide was added to saliva, oxidation of urate was dependent on its concentration and peroxidase activity. Our findings establish urate as a likely physiological substrate for LPO that will influence host defense and give rise to reactive electrophilic metabolites. 相似文献
