The influence of (1H-1,2,3-triazol-1-yl)-1,2,5-oxadiazole derivatives: 4-amino-3-(5-methyl-4-ethoxycarbonyl-(1H-1,2,3-triazol-1-yl)-1,2,5-oxadiazole (TF4CH3) and 4,4′-bis(5-methyl-4-ethoxycarbo-nyl-1H-1,2,3-triazol-1-yl)-3,3′-azo-1,2,5-oxadiazole (2TF4CH3) on stimulation of human platelet soluble guanylate cyclase by YC-1, NO donors (sodium nitroprusside, SNP, and spermine NONO) and on a synergistic increase of NO-dependent activation of the enzyme in the presence of YC-1 has been investigated. Both compounds increased guanylate cyclase activation by YC-1, potentiated guanylate cyclase stimulation by NO donors and increased the synergistic effect of YC-1 on the NO-dependent activation of soluble guanylate cyclase. The similarity in the properties of the examined 1,2,3-triazol-1-yl-1,2,5-oxadiazole derivatives with that of YC-1 and a possible mechanism underlying the recognized properties of compounds used are discussed.  相似文献

The characteristic patterns of EEG spatial organization at different stages of natural sleep and the hypnotic state were studied in 26 volunteers aged 18–22 years. EEGs were recorded using 12 monopolar leads, and EEG cross-correlation coefficient matrices were calculated for consecutive epochs (4 and 8 s). Matrices averaged for each state were treated using factor analysis. The EEG correlation matrices were compared element by element for the states studied and the waking state. Relatively similar changes in the spatial structure of EEG correlations were observed at different stages of natural sleep, with the correlations tending to intensify, especially in the posterior temporal region of the right hemisphere. In the light and deep (somnambular) phases of hypnosis, the interaction between cortical zones that was characteristic of distant relationships of the EEGs of frontal regions, especially the posterior inferior frontal region of the right hemisphere, decreased. The systemic reorganization of the interregional EEG correlations during natural sleep was considerably more pronounced than in the hypnotic state. Notwithstanding, the highly orderly spatial organization of the cortical biopotential field that was typical of the waking state was retained at different stages of natural sleep and hypnosis. Doppler sonography and rheoencephalography were used to study cerebral hemodynamics of young schoolchildren in the village of Klimovskoe (Konosha raion, Arkhangel’sk oblast, Russia). Mean linear blood flow velocities (LBVs) in the major cerebral vessels did not differ significantly from the velocities measured in age-matched children living in St. Petersburg. The index of resistivity was lower in the residents of the North than in their counterparts in St. Petersburg, which suggests a decreased tone of resistive vessels of the children living in the North. Asymmetry of the LBVs through the homonymous cerebral arteries of the left and right hemispheres was found in 67% of children examined. Asymmetry of the LBVs in the anterior, middle, and posterior cerebral and vertebral arteries (VAs) was correlated with a malformed posture (such as scoliosis or a slouch). In most children examined, sideward head rotation was accompanied by compression of VAs at the level of the C 1 and C 2 cervical vertebrae; in 71% of them, this condition could be regarded as a hemodynamically significant stenosis. In all children, a decreased LBV observed in one of the VAs was completely compensated by an increased LBV in its contralateral counterpart. Doppler sonographic signs of dystonic VAs and/or asymmetrical LBVs in the VAs were found in 79% of children examined. These children can be supposed to have latent disorders of blood flow in the VA basins caused by vertebral influences (related to irritation of the periarterial nervous plexus or cervical autonomic plexuses). The schoolchildren living in the North exhibited a lower reactivity of their cerebral vessels to hypo-and hypercapnia than their counterparts living in the central parts of the Russian Federation or in St. Petersburg. Regional differences were found in the sensitivity of cerebral vessels to hypocapnia. The volume of blood in the basins of the internal carotid arteries determined in a hypocapnic test was decreased; it was increased in the basins of VAs. This fact can be interpreted as a regional redistribution of the cerebral circulation caused by extreme environmental factors stimulating vital regulatory sites of the brainstem.  相似文献

It is shown that water-ion homeostasis in a water-biopolymer-salt system is realized when proportion of the concentrations of the components changes in the definite range. The mechanism of instantaneous osmotic regulating reaction is discussed, which is based on the ability of the system to physicochemical redistribution of the components of the solution and the formation of supramolecular structures in the form of dynamic biopolymer clusters. Quasiequilibrium biopolymer<-->cluster leads to changes in the properties of the hydration water phase that provide the stabilization of water activity and the ability of hydration water to dissolve electrolyte ions. It regulates osmotic pressure and surface tension in water-protein matrix. This process supports a balance of stabilizing and destabilizing forces that affect protein structure and determine its functional activity.  相似文献

An approach to measure the microscopic surface tension in a water-protein matrix using spin label EPR is proposed. The approach utilizes the parameters of the viscosity isotherms for the correlation time of the spin labels bound to a protein macromolecule. The pattern of changes in the specific surface energy in the protein hydration shell depending on the protein concentration, structure of reporter group, ligand state of protein, and the presence of salts, water-soluble polymers, and D2O in solution was studied using serum albumin, hemoglobin, and antibodies as examples. Possible causes underlying the observed changes in surface tension are discussed.  相似文献

Genetic diversity of wild reindeer (Rangifer tarandus) inhabiting the European part of Russia, including Komi Republic, Arkhangelsk oblast, Murmansk oblast, and the Republic of Karelia was characterized using sequence polymorphism of the mtDNA control region. Despite of currently low population number of wild reindeer, they were characterized by a high level of genetic diversity (?? = 0.018; H = 0.872 to 0.914). Phylogenetic analysis showed close relationships between European reindeer and wild reindeer of Siberia. In reindeer from Murmansk oblast a haplotype in common with the wild reindeer form Southwestern Norway was described. The reindeer sample examined contained no haplotypes earlier described for the reindeer of Central Norway. It is suggested that in recent past wild reindeer from the European north of Russia formed one population with the reindeer from the north of the Asian part of Eurasia.  相似文献

The authors demonstrated a marked dissociation of the Neisseria meningitis cultures and the capacity to their rapid change from S- to R-forms. Differential signs of the S- and R-forms are described. Differences in the degree of dissociation of the cultures depending on their origin were shown: strains isolated from the cerebrospinal fluid and the blood of patients with meningitis proved to be less dissociated, and those isolated from carriers--more dissociated. Transfers from media to media and adaptation of cultures to artificial nutrient media produced the greatest influence on dissociation of the freshly isolated strains.  相似文献

The previously derived formulas for the curves corresponding to sol-gel, liquid-liquid, and liquid-solid phase transitions, which correlate the critical molar composition of the water-protein-salt system with individual characteristic features of its component (protein charge z, the number of ions adsorbed v, the function of electrolyte activity A) are presented as curves in ordinary coordinates of protein solubility logS against salt concentration m3. Tendencies in changes in phase transition lines versus the v, z, and v/z ratio have been determined. Correlations of the salting-out curve and the salting-out coefficient with phase transitions are discussed.  相似文献
