M. tuberculosis and M. leprae are considered to be prototypical intracellular pathogens that have evolved strategies to enable growth in the intracellular phagosomes. In contrast, we show that lysosomes rapidly fuse with the virulent M. tuberculosis- and M. leprae-containing phagosomes of human monocyte-derived dendritic cells and macrophages. After 2 days, M. tuberculosis progressively translocates from phagolysosomes into the cytosol in nonapoptotic cells. Cytosolic entry is also observed for M. leprae but not for vaccine strains such as M. bovis BCG or in heat-killed mycobacteria and is dependent upon secretion of the mycobacterial gene products CFP-10 and ESAT-6. The cytosolic bacterial localization and replication are pathogenic features of virulent mycobacteria, causing significant cell death within a week. This may also reveal a mechanism for MHC-based antigen presentation that is lacking in current vaccine strains.  相似文献

HIV-1 infection has significant effect on the immune system as well as on the nervous system. Breakdown of the blood-brain barrier (BBB) is frequently observed in patients with HIV-associated dementia (HAD) despite lack of productive infection of human brain microvascular endothelial cells (HBMEC). Cellular products and viral proteins secreted by HIV-1 infected cells, such as the HIV-1 Gp120 envelope glycoprotein, play important roles in BBB impairment and HIV-associated dementia development. HBMEC are a major component of the BBB. Using cocultures of HBMEC and human astrocytes as a model system for human BBB as well as in vivo model, we show for the first time that cannabinoid agonists inhibited HIV-1 Gp120-induced calcium influx mediated by substance P and significantly decreased the permeability of HBMEC as well as prevented tight junction protein down-regulation of ZO-1, claudin-5, and JAM-1 in HBMEC. Furthermore, cannabinoid agonists inhibited the transmigration of human monocytes across the BBB and blocked the BBB permeability in vivo. These results demonstrate that cannabinoid agonists are able to restore the integrity of HBMEC and the BBB following insults by HIV-1 Gp120. These studies may lead to better strategies for treatment modalities targeted to the BBB following HIV-1 infection of the brain based on cannabinoid pharmacotherapies.  相似文献

The species composition and aboveground biomass of plant and lichens and the composition of reindeer and musk ox diet in the arctic tundra of Wrangel Island were studied in 2004 to 2007. The above-ground phytomass in different areas of the island varied from 1105 to 2100 kg/ha. The composition of plants consumed by reindeer and musk oxen and their proportions in the diet were determined by standard micro-histological analysis of plant remains in their feces. The results showed that, either in winter or in summer, both species obviously preferred feeding on willows (Salicaceae), which comprised almost half of their diet. Moreover, their feeding was highly selective, especially with respect to sedges and rushes (Cyperaceae + Juncaceae) and legumes (Fabaceae). Although the contributions of these plant groups to the total aboveground phytomass were very small (less than 4 and 8%), their proportions in the diet reached 27 and 24%, respectively. Mosses were not a preferred forage: their proportion in the aboveground phytomass reached 40%, but that in the diets of both species was below 10% in summer and increased to 20% only in the winter diet of reindeer. At a high abundance of lichens (up to 20% of the aboveground phytomass), neither of the animals consumed them during the study period.  相似文献

In Russia, the conservation of anseriformes is possible through the creation of temporary huntingfree zones during hunting season, especially in spring. A justification for creating such zones and outlining their boundaries (by analogy with the experience derived from the countries on North America) each season must be based on data on annual waterfowl monitoring. The present paper describes census experience drawn from the use of ultralight aviation for further delineating the key staging sites of waterfowl in western Siberia. To refine the duration of monitoring, observation data were combined with those derived from geese equipped with GSM-GPS transmitters. In the spring and autumn of 2012–2014, we covered over 50000 km of aerial surveys of 25 waterfowl species. A new method is advanced for assessing their numbers based on visual observations, flock photography, and modern statistics. We estimated the species densities in 16 habitat types delineated on the basis of Landsat imagery. In terms of this, a system is proposed for extrapolating the survey data on 25 waterfowl species onto model sites in western Siberia. Drops in the numbers of several mass game species were noted. Based on an evaluation of the habitat quality, ten waterfowl hunting-free zones were suggested and delineated. A GIS project was launched that incorporated the main migration routes, boundaries of the key sites, places of mass bird aggregations, and sites for the observation of rare, Red Data Book. A program of long-term monitoring and sustainable use of waterfowl in the study region is offered. Such an approach must also be applied to other regions of Russia.  相似文献

CP-T1 is the only described generalized transducing bacteriophage for the intestinal pathogen Vibrio cholerae, yet many of its basic biological parameters remain unknown. Due to low frequencies of transduction and pseudolysogen formation, CP-T1 has not been widely used as a genetic tool. To overcome these limitations, we have isolated a conditional mutant of CP-T1 that exhibits temperature-sensitive plaque formation. Several biological properties of CP-T1ts were determined, including its restrictive temperature, adsorbance profile to host cells, burst time, and burst size. Based on these properties, an optimized transduction protocol was designed which resulted in several fold higher transduction frequencies for a variety of genetic markers from a number of chromosomal loci. Generalized transduction was also demonstrated between classical and E1 Tor biotype strains of V. cholerae.  相似文献

Individuals with autism constitute a variable population whose members are spread along the autism spectrum. Subpopulations within that spectrum exhibit other conditions, such as anxiety, intellectual disabilities, hyperactivity and epilepsy, with different severities and co‐occurrences. Among the genes associated with the increased risk for autism is the methylenetetrahydrofolate‐reductase (MTHFR) 677C>T polymorphism, which impairs one‐carbon (C1) metabolic pathway efficiency. The frequency of the MTHFR677TT homozygote is markedly higher among autism patients and their mothers than in the general population. Here, we report on the Mthfr heterozygous knockout (KO) mouse as a rodent model of autism that shows the contributions of maternal and offspring genotypes to the development of autistic‐like behaviors. Maternal Mthfr‐deficiency was associated with developmental delays in morphogenic features and sensory‐motor reflexes in offspring. In the adult male mouse, behaviors representing core autism symptoms, such as repetitive behavior and restricted interest, were affected by maternal genotype while social behaviors were affected by both maternal and offspring genotypes. In females and males, behaviors associated with autism such as memory impairment, social aggression and anxiety were affected by both the maternal and offspring Mthfr genotypes, with sex‐dependent differences. Mthfr‐deficient male mice with observable impacts on behavior presented a particular laminar disturbance in parvalbumin interneuron density and innervation in superficial and deep layers of the cingulate cortex. This mouse model of autism will help to elucidate the molecular mechanisms that predispose a significant subgroup of autistic patients to abnormal development and to distinguish between the in‐utero and autonomous factors involved in autism.  相似文献
