Mini UV lamps were installed over antarctic plants at Léonie Island, Antarctic peninsula, and shoot length measurements of Deschampsia antarctica were performed during the austral summer January–February 1999.We studied the response of the antarctic hairgrass, Deschampsia antarctica to enhanced UV-B. In a climate room experiment we exposed tillers of Deschampsia antarctica, collected at Léonie Island, Antarctic peninsula, to ambient and enhanced levels of UV-B radiation. In this climate room experiment with 0, 2.5 and 5 kJ m–2 day–1 UV-BBE treatments we observed that length growth of shoots at 2.5 and 5 kJ m–2 day–1 UV-BBE was markedly reduced compared to 0 kJ m–2 day–1 UV-BBE. In addition, there was an increased number of shoots and increased leaf thickness with enhanced UV-B. The Relative Growth Rate (RGR) was not affected by UV-B, possibly because reduced shoot length growth by enhanced UV-B was compensated by increased tillering. Light response curves of net leaf photosynthesis of plants exposed to 5 kJ m–2 day–1 UV-BBE did not differ from those exposed to 0 kJ m–2 day–1 UV-BBE. The content of UV-B absorbing compounds of plants exposed to increasing UV-B did not significantly change.Mini UV-B lamp systems were installed in the field, to expose the terrestrial antarctic vegetation at Léonie Island to enhanced solar UV-B. In that study, the increment of shoot length of tagged plants of Deschampsia antarctica during the January-February 1999 at Léonie Island, was recorded and compared to shoot length growth under controlled conditions.The consequences of enhanced UV-B radiation as a result of ozone depletion for the terrestrial antarctic ecosytems are discussed.  相似文献

The effects of enhanced UV-B are often overestimated in greenhouse studies due to low levels of photosynthetically active radiation (PAR). For this reason, we studied effects of enhanced UV-B (12 kJ m–2 d–1) at low and near ambient PAR levels on young vegetative plants of Vicia faba, in the greenhouse. It was hypothesized that near ambient PAR levels could reduce the negative UV-B effects on growth, due to higher amounts of UV-B absorbing compounds in the leaves and to morphological changes attenuating UV-B damage.We found that effects of enhanced UV-B on the growth were not negative. We found an increase in biomass in response to enhanced UV-B at low and near ambient PAR levels. The increase in biomass was related to increased branching, which leads to a higher interception of PAR. Enhanced irradiance of both PAR and UV-B had similar photomorphogenic effects: thicker and smaller leaves and reduced plant height and internode length. Moreover, the concentration of UV-B absorbing compounds was increased. We conclude that in this study effects of enhanced UV-B were mainly photomorphogenic effects, which were also induced by radiation in the PAR region.  相似文献

The charophycean algal species Chara aspera was exposed for 73 days to three levels of UV-B radiation (weighted according to Caldwell's generalized plant action spectrum): 1.9 kJ m–2 day–1 (`no UV-B'), 6.4 KJ m–2 day–1 (ambient) and 10.5 kJ m–2 day–1 (enhanced UV-B), the latter level simulating 30% ozone reduction in The Netherlands.Charophycean algae are mainly freshwater organisms and are thought to be the algae most closely related to higher land plants. Therefore we expected that responses of charophycean algae to UV-B radiation might be more related to those observed in the higher land plants than those of other `lower' algal groups.Under elevated UV-B radiation algal length was reduced. There was no induction of UV absorbing compounds under enhanced UV-B. This might relate to a sensitive response to UV-B radiation. The charophycean algae show similar adaptations to UV-B radiation as terrestrial plants, while not having UV-screens as occur in many angiosperms. Vegetative reproduction (bulbils) increased in the presence of UV-B radiation, while generative reproduction (antheridia and oogonia) decreased.  相似文献

As a reference for ongoing studies reconstructing past vegetation, climate and environment, pollen spectra in tundra peat profiles from Svalbard, were investigated. The base of tundra peat cores collected from Ny Ålesund, Stuphallet, Blomstrand and Isdammen has been 14C dated to 350–490 BP, 5710 BP, 4670 BP and 700–900 BP, respectively. The Stuphallet and Blomstrand (Brøggerhalvøya) peat profiles were composed of a peat developed in a nutrient enriched and wet tundra environment of steep birdcliffs. Pollen concentrations were low, Brassicaceae pollen dominated the whole profile. In contrast, the Ny Ålesund and Isdammen profiles contained high pollen concentrations and suggest a nutrient-poor, dry tundra environment. Pollen of the polar willow, Salix polaris, occurred commonly throughout all four peat profiles. In the relatively high resolution (10 years per peat core sample) analysis of the Ny Ålesund core, starting before or at the beginning of the Little Ice Age (LIA, 16th-mid 19th century), dominance of Saxifraga oppositifolia indicates a cold and dry climate, followed by a decline of Saxifraga oppositifolia and gradual increase of Salix polaris after the LIA, which indicates a moist and milder climate.  相似文献

UV-B radiation currently represents c. 1.5% of incoming solar radiation. However, significant changes are known to have occurred in the amount of incoming radiation both on recent and on geological timescales. Until now it has not been possible to reconstruct a detailed measure of UV-B radiation beyond c. 150 yr ago. Here, we studied the suitability of fossil Pinus spp. pollen to record variations in UV-B flux through time. In view of the large size of the grain and its long fossil history, we hypothesized that this grain could provide a good proxy for recording past variations in UV-B flux. Two key objectives were addressed: to determine whether there was, similar to other studied species, a clear relationship between UV-B-absorbing compounds in the sporopollenin of extant pollen and the magnitude of UV-B radiation to which it had been exposed; and to determine whether these compounds could be extracted from a small enough sample size of fossil pollen to make reconstruction of a continuous record through time a realistic prospect. Preliminary results indicate the excellent potential of this species for providing a quantitative record of UV-B through time. Using this technique, we present the first record of UV-B flux during the last 9500 yr from a site near Bergen, Norway.  相似文献

We report a long-term experiment on the photosynthetic response of natural vegetation of Deschampsia antarctica (Poaceae) and Turgidosculum complicatulum (Lichenes) to altered UV-B levels on Léonie Island, Antarctica.UV-B above the vegetation was reduced by filter screens during two seasons. Half of the screens were transparent to UV-A and UV-B (ambient treatment) or absorbing UV-B and part of the UV-A (below-ambient treatment). Half of the wedge- shaped filters had side walls leading to an enhancement of the daily mean temperature in summer by 2–4 °C, simulating rising mean air temperature on the Antarctic Peninsula. The other half of the filters were without side walls resulting in close-to-ambient temperature underneath. Plots without filters served as controls.UV-B supplementation of an extra 1.3 kJ UV-BBE was achieved using UV-mini-lamp systems during 15 days in the second season.We found no evidence that altered incident UV-B levels and temperature had an effect on maximum photosystem II efficiency (F v/F m) and effective photosystem II efficiency (F/F m) in both species. UV-B reduction did not influence contents of chlorophyll, carotenoids and methanol-soluble UV absorbing compounds in D. antarctica.Flowering shoot length of D. antarctica was not affected by UV-B reduction. Temperature enhancement tended to result in longer inflorescence axes. Results of two austral summer seasons of UV- reduction in natural stands of D. antarctica and T. complicatulum suggest that current ambient levels of UV-B do not have a direct effect on the photosynthetic performance and pigment contents of these species. Cumulative effects on growth have not been recorded after two years but can not be excluded on a longer term.  相似文献

Cationic membrane disruptive peptides such as melittin would appear to have attributes necessary for DNA delivery: DNA binding via electrostatic interactions and membrane lysis to enable cytoplasmic delivery. However, the relatively small overall charge of membrane disruptive peptides results in weak interactions with DNA. As a model of cationic membrane disruptive peptides, amphiphilic polyvinyl ethers were synthesized. The number of positively charged groups incorporated into these polymers is substantially greater than membrane-active peptides, which enables these polymers to form stable complexes with DNA. By varying the length of the hydrophobic groups incorporated into the polymer from one to four carbons, the dependence of membrane activity on side chain length was established. The ability of these polymers to transfect DNA in tissue culture was tested, and it was found that transfection efficiency is dependent upon the membrane disruptive activity of the polymer. Comparison of melittin and synthetic polymers suggests that transfection and toxicity appear to be dependent upon their affinity for DNA. This demonstration of relationships among membrane lysis, transfection, DNA binding, and polymer side-chain composition establishes a new class of transfection reagents and may guide in the design of polymers and formulations that will enable efficient in vivo transfection.  相似文献

Many proteins that contain a carboxyl-terminal CaaX sequence motif, including Ras and yeast a-factor, undergo a series of sequential posttranslational processing steps. Following the initial prenylation of the cysteine, the three C-terminal amino acids are proteolytically removed, and the newly formed prenylcysteine is carboxymethylated. The specific amino acids that comprise the CaaX sequence influence whether the protein can be prenylated and proteolyzed. In this study, we evaluated processing of a-factor variants with all possible single amino acid substitutions at either the a(1), the a(2), or the X position of the a-factor Ca(1)a(2)X sequence, CVIA. The substrate specificity of the two known yeast CaaX proteases, Afc1p and Rce1p, was investigated in vivo. Both Afc1p and Rce1p were able to proteolyze a-factor with A, V, L, I, C, or M at the a(1) position, V, L, I, C, or M at the a(2) position, or any amino acid at the X position that was acceptable for prenylation of the cysteine. Eight additional a-factor variants with a(1) substitutions were proteolyzed by Rce1p but not by Afc1p. In contrast, Afc1p was able to proteolyze additional a-factor variants that Rce1p may not be able to proteolyze. In vitro assays indicated that farnesylation was compromised or undetectable for 11 a-factor variants that produced no detectable halo in the wild-type AFC1 RCE1 strain. The isolation of mutations in RCE1 that improved proteolysis of a-factor-CAMQ, indicated that amino acid substitutions E139K, F189L, and Q201R in Rce1p affected its substrate specificity.  相似文献

Elymus athericus (Link) Kerguélen, a C3 grass, was grown in a greenhouse experiment to determine the effect of enhanced atmospheric CO2 and elevated UV-B radiation levels on plant growth. Plants were subjected to the following treatments; a) ambient CO2-control UV-B, b) ambient CO2-elevated UV-B, c) double CO2-control UV-B, d) double CO2-elevated UV-B. Elevated CO2 concentrations stimulated plant growth, biomass production was 67% higher than at ambient CO2. Elevated UV-B radiation had a negative effect on growth, biomass production was depressed by 31%. Enhanced CO2 combined with elevated UV-B levels caused a biomass depression of 8% when compared with the control plants. UV-B induced growth depression can be modified by a growth stimulus caused by high CO2 concentrations. Growth analysis has been performed and possible physiological mechanisms behind changing growth parameters are discussed.  相似文献

Cancer is the second most frequent cause of death in children. Because the prognosis for childhood malignancies has improved, attention has now focused on long‐term consequences of cancer treatment. The immediate effects of chemotherapy on soft tissues have been well described; however, there is less information about long‐term effects of chemotherapy on the development of dental tissues. To test the association between the effect of chemotherapy on enamel development, we examined two groups of rats: one that had received an intraperitoneal dose of 200 mg/kg of irinotecan, whereas the other (control) group had received vehicle only. Rats were killed at 6, 48 and 96 hr post‐injection; the mandibles dissected out, fixed for histological evaluation and scanned for mineralization defects by Micro‐CT. Our results showed structural changes in the ameloblast layer along with a significant reduction in mineralization and thickness of enamel at 96 hr after chemotherapy. These data demonstrate that irinotecan induces structural changes in forming enamel that become apparent after anticancer chemotherapy treatment.  相似文献
