Background and Aims
Mechanical perturbation is known to inhibit elongation of the inflorescence stem of Arabidopsis thaliana. The phenomenon has been reported widely for both herbaceous and woody plants, and has implications for how plants adjust their size and form to survive in mechanically perturbed environments. While this response is an important aspect of the plant''s architecture, little is known about how mechanical properties of the inflorescence stem are modified or how its primary and secondary tissues respond to mechanical perturbation.Methods
Plants of the Columbia-0 ecotype were exposed to controlled brushing treatments and then submitted to three-point bending tests to determine stem rigidity and stiffness. Contributions of different tissues to the inflorescence stem geometry were analysed.Key Results
Perturbed plants showed little difference in stem diameter, were 50 % shorter, 75 % less rigid and 70 % less stiff than controls. Changes in mechanical properties were linked to significant changes in tissue geometry – size and position of the pith, lignified interfascicular tissue and cortex – as well as a reduction in density of lignified cells. Stem mechanical properties were modified by changes in primary tissues and thus differ from changes observed in most woody plants tested with indeterminate growth – even though a vascular cambium is present in the inflorescence axis.Conclusions
The study suggests that delayed development of key primary developmental features of the stem in this ecotype of Arabidopsis results in a ‘short and flexible’ rather than a ‘short and rigid’ strategy for maintaining upright axes in conditions of severe mechanical perturbation. The mechanism is comparable with more general phenomena in plants where changes in developmental rate can significantly affect the overall growth form of the plant in both ecological and evolutionary contexts. 相似文献953.

1. The underlying basis of alternative male reproductive strategies is either genetic or environmental. Several non‐pollinating fig wasp lineages have dimorphic males, typically with winged males that disperse from natal figs to mate and flightless males that seek mating opportunities in natal figs. 2. Walkerella sp. from Ficus benjamina has dark and pale wingless males. Observations and experiments in Xishuangbanna, Southern China found that (i) the sex ratio of Walkerella sp. did not vary with foundress number or brood size. (ii) The frequency of dark males increased with brood size and foundress number and they were absent from figs with a single foundress. This produced a higher proportion of dark males at higher densities. (iii) Males of both morphs fought, but injuries to dark males were more frequent. (iv) Dark males were more likely to disperse away from their natal figs and (v) they were more resistant to dehydration. 3. Responses to selection are constrained by the genetic options available. Consequently, selection pressures acting on different lineages can produce similar outcomes that are achieved in different ways. Walkerella species lack winged males, but dark males display some of their features, dispersing from natal figs and displaying appropriate physiological and behavioural adaptations. Crossing experiments indicate that hybrid sterility barriers frequently have developed within diploid, circumpolar plant species of the genus Draba. To gain insight into the rapid evolution of postzygotic reproductive isolation in this system, we augmented the linkage map of one of these species, D. nivalis, and searched for quantitative trait loci (QTLs) associated with reproductive isolation. The map adds 63 new dominant markers to a previously published dataset of 31 co-dominant microsatellites. These markers include 52 amplified fragment length polymorphisms (AFLPs) and 11 sequence-specific amplified polymorphisms (SSAPs) based on retrotransposon sequence. 22 markers displaying transmission ratio distortion were further included in the map. We resolved eight linkage groups with a total map length of 894 cM. Significant genotype-trait associations, or quantitative trait loci (QTL), were detected for reproductive phenotypes including pollen fertility (4 QTLs), seed set (3 QTLs), flowering time (3 QTLs) and number of flowers (4 QTLs). Observed patterns of inheritance were consistent with the influence of both nuclear-nuclear interactions and chromosomal changes on these traits. All seed set QTLs and one pollen fertility QTL displayed underdominant effects suggestive of the involvement of chromosomal rearrangements in hybrid sterility. Interestingly, D. nivalis is predominantly self-fertilizing, which may facilitate the establishment of underdominant loci and contribute to reproductive isolation. 相似文献

The bone microenvironment (BME) is the main hub of all skeletal related pathological events in osteosarcoma leading to tumor induced bone destruction, and decreasing overall bone quality and bone strength. The role of extra-cellular membrane vesicles (EMVs) as mediators of intercellular communication in modulating osteosarcoma-BME is unknown, and needs to be investigated. It is our hypothesis that osteosarcoma-EMVs contain pro-osteoclastogenic cargo which increases osteoclastic activity, and dysregulated bone remodeling in the osteosarcoma-BME. In this study, EMVs were isolated from the conditioned media of 143B and HOS human osteosarcoma cell cultures using differential ultracentrifugation. Nano-particle tracking analysis determined EMVs in the size range of 50-200 nm in diameter. The EMV yield from 143B cells was relatively higher compared to HOS cells. Transmission electron microscopy confirmed the ultrastructure of 143B-EMVs and detected multivesicular bodies. Biochemical characterization of 143B-EMVs detected the expression of bioactive pro-osteoclastic cargo including matrix metalloproteinases-1 and -13 (MMP-1, -13), transforming growth factor-β (TGF-β), CD-9, and receptor activator of nuclear factor kappa-β ligand (RANKL). Detection of a protein signature that is uniquely pro-osteoclastic in 143B-EMVs is a novel finding, and is significant as EMVs represent an interesting mechanism for potentially mediating bone destruction in the osteosarcoma-BME. This study further demonstrates that 143B cells actively mobilize calcium in the presence of ionomycin, and forskolin, and induce cytoskeleton rearrangements leading to vesicular biogenesis. In conclusion, this study demonstrates that 143B osteosarcoma cells generate EMVs mainly by mechanisms involving increased intracellular calcium or cAMP levels, and contain pro-osteoclastic cargo. 相似文献

Increasing the eco‐efficiency of fishing fleets is currently a major target issue in the seafood sector. This objective has been influenced in recent years by soaring fuel prices, a fact particularly relevant to a sector whose vessels present high energy consumption rates. Efforts to minimize fuel consumption in fishing fleets result in economic benefits and also in important reductions regarding environmental impacts. In this article, we combine life cycle assessment (LCA) and data envelopment analysis (DEA) to jointly discuss the operational and environmental performances of a set of multiple, similar entities. We applied the “five‐step LCA + DEA method” to a wide range of vessels for selected Galician fisheries, including deep‐sea, offshore, and coastal fleets. The environmental consequences of operational inefficiencies were quantified and target performance values benchmarked for inefficient vessels. We assessed the potential environmental performance of target vessels to verify eco‐efficiency criteria (lower input consumption levels, lower environmental impacts). Results revealed the strong dependence of environmental impacts on one major operational input: fuel consumption. The most intensive fuel‐consuming fleets, such as deep sea trawling, were found to entail the diesel consumption levels nearest to the efficiency values. Despite the reduced environmental contributions linked to other operational inputs, such as hull material, antifouling paint, or nets, these may contribute to substantial economic savings when minimized. Finally, given that Galicia is a major fishing region, many of the conclusions and perspectives obtained in this study may be extrapolated to other fishing fleets at the international level. 相似文献
