Archaeological investigations of pastoral economies often emphasize exchange relations with agricultural populations, though for Bronze Age Eurasia the notion of a ubiquitous ‘pastoral realm’ has masked various forms of mixed subsistence economies. In Central Asia, there are few attempts to specifically identify the domestic crops utilized by mobile pastoralists or what they may suggest about the role of agriculture in mobile pastoral production or subsistence strategies. This study reports the macrobotanical remains from two Late/Final Bronze Age (ca. 1950–1300 bc) mobile pastoralist habitation sites in the Murghab alluvial fan region of southern Turkmenistan. We compare our results with published macrobotanical data from contemporary agricultural settlements in the Murghab region, as well as with other sites in broader prehistoric Eurasia. We find that mobile pastoralists in the Murghab utilized some of the same domestic crops as their sedentary neighbors. While the data presented here do not preclude the possibility that mobile pastoralists may have practiced some low-investment cultivation (particularly of millet), we hypothesize an economic model that places mobile pastoralists in direct contact with nearby sedentary farming communities through exchange for pre-processed grains. These results highlight one of the possible strategies of mobile pastoral subsistence in Central Asia, and are a further step toward identifying the various degrees of agricultural involvement in the conceptually outdated pastoral realm of Eurasia.  相似文献

Cytochrome P450 enzymes are found in all life forms. P450s play an important role in drug metabolism, and have potential uses as biocatalysts. Human P450s are membrane-bound proteins. However, the interactions between P450s and their membrane environment are not well-understood. To date, all P450 crystal structures have been obtained from engineered proteins, from which the transmembrane helix was absent. A significant number of computational studies have been performed on P450s, but the majority of these have been performed on the solubilised forms of P450s. Here we present a multiscale approach for modelling P450s, spanning from coarse-grained and atomistic molecular dynamics simulations to reaction modelling using hybrid quantum mechanics/molecular mechanics (QM/MM) methods. To our knowledge, this is the first application of such an integrated multiscale approach to modelling of a membrane-bound enzyme. We have applied this protocol to a key human P450 involved in drug metabolism: CYP3A4. A biologically realistic model of CYP3A4, complete with its transmembrane helix and a membrane, has been constructed and characterised. The dynamics of this complex have been studied, and the oxidation of the anticoagulant R-warfarin has been modelled in the active site. Calculations have also been performed on the soluble form of the enzyme in aqueous solution. Important differences are observed between the membrane and solution systems, most notably for the gating residues and channels that control access to the active site. The protocol that we describe here is applicable to other membrane-bound enzymes.  相似文献

5-Methoxymethyl-2'-deoxyuridine (MMUdR), a drug with potent antiviral activity in vitro against Herpes simplex virus, was investigated for its immunosuppressive effects. Doses as high as 2000mg/kg given daily for 9 days were not immunosupporessive as judged by the fact that treated animals produced normal immune responses to sheep erythrocytes, Brucella bacteria, and Herpes simplex virus.  相似文献

A peptide corresponding to residues 1 to 23 of glycoprotein D of herpes simplex virus type 1 was chemically synthesized and coupled to a fatty acid carrier by standard Merrifield synthesis procedures. The resulting peptide-palmitic acid conjugate (acylpeptide) exhibited enhanced immunogenicity in mice as compared with that exhibited by the free form of the peptide. Incorporation of the acylpeptide into liposomes further increased the immunogenicity of the peptide, while inclusion of the immunomodulators muramyl tripeptide phosphatidylethanolamine and monophosphoryl lipid A into the same liposome stimulated the strongest response. The humoral immune responses induced by the acylpeptide-liposome construct were greater than those induced by peptide in Freund complete adjuvant, and cellular responses were equal. The acylpeptide-immunomodulator-liposome formulation also induced significant levels of protective immunity, although the immunity was less than that induced by herpes simplex virus infection. Acylated peptides, especially in liposomes, were taken up more effectively by draining lymph nodes, which possibly accounts in part for the enhanced immunogenicity of the peptides. Since the acylpeptide-immunoliposome formulation used was nontoxic, it could represent a useful way to enhance immunogenicity of subunit peptides used for vaccine purpose in humans and animals.  相似文献

Young, visually symptomless leaves from potato (Solanum tuberosum) plants infected with Verticillium dahliae exhibited reduced carbon assimilation rate, stomatal conductance, and intercellular CO2, but no increase in dark respiration, no change in the relationship between carbon assimilation rate versus intercellular CO2, and no change in light use efficiency when intercellular CO2 was held constant. Therefore, the initial decrease in photosynthesis caused by V. dahliae was caused by stomatal closure. Errors in the intercellular CO2 calculation caused by uneven distribution of carbon assimilation rate across the leaf were tested by 14CO2 autoradiography. Patchiness was found at a low frequency. Low stomatal conductance was correlated with low leaf water potentials. Infection did not affect leaf osmotic potentials.  相似文献

We have employed two-color staining with monoclonal anti-T-cell markers and a broadly reactive, nonbinding site monoclonal anti-idiotype to permit direct visualization of idiotype-bearing T cells in mouse lymph nodes following immunization with phosphorylcholine. Double positive cells peak in incidence on Days 8 to 12, as 17% of total idiotype+ cells but as only 0.7% of T cells. Such cells are antigen-specific, appearing in peripheral lymph nodes only following phosphorylcholine challenge. While Lyt-2+ and L3T4+ subsets are represented and both subsets reexpress the idiotypic determinant following its enzymatic removal, the L3T4+ subset represents the majority of idiotype+Thy-1+ cells. These findings raise the possibility that antigen-specific receptors on T and B cells, encoded by entirely different genetic information, may exhibit similarities in tertiary structure in portions of the molecules not directly involved in antigen binding. This is the first determination of the phenotype and chronology of appearance of idiotype-bearing normal T cells following immunization. It is consistent with previous reports of functionally defined idiotype-bearing T cells and provides direct support for the existence of such cells.  相似文献

The inflammatory neuropeptide substance P acted as a costimulant for macrophage CSF-1-induced clonal proliferation of murine marrow-derived two signal-dependent mononuclear phagocyte progenitors. Substance P had no effect on clonal proliferation by progenitors responding solely to CSF-1. Substance P fragment 2-11 had no costimulatory activity; however, SP fragment 1-4 retained the full activity of the parent undecapeptide. Fragment 1-4 (ARG-PRO-LYS-PRO), a peptide containing a PRO residue between two positive charges, is a tuftsin-like (THR-LYS-PRO-ARG) tetrapeptide, and tuftsin exerted an identical costimulatory effect. Substance P, SP:1-4, and tuftsin were optimally effective as costimulants at 10(-7) to 10(-6) M. (ALA1)-tuftsin, an inhibitory analog of tuftsin, was a potent negative regulator of two signal-dependent colony formation. (ALA1)-tuftsin at concentrations less than or equal to 10(-9) M exerted dose-dependent inhibition of the positive effects of optimal concentrations of all of the co-stimulants tested, including bacterial LPS. The inhibitory tetrapeptide was equivalent in activity to ferritin, an established inhibitor of two signal-dependent colony formation. The results indicated that SP may influence myelopoiesis in addition to its other inflammatory and immunopotentiating properties. In addition, a potentially valuable modulator of SP and LPS responses in this system, (ALA1)-tuftsin, was identified.  相似文献
