Sb‐doped SnO2 (ATO) is used as an alternative support material to replace carbon in the highly corrosive environment of a fuel cell cathode. Two ATO powders with different morphologies are decorated with Pt nanoparticles and afterwards used as the cathode catalyst. The commercial ATO powder exhibits crystallites in the nanometer range, while the home‐made ATO powder, which was synthesized by ultrasonic spray pyrolysis, consists of polycrystalline hollow spheres. The spheres have diameters in the micrometer range and are composed of individual nanocrystallites. The unusual morphology of the home‐made ATO offers nano‐ and microporosity at the same time and opens up new possibilities for the controlled design of electrode structures in low‐temperature polymer electrolyte fuel cells. Both materials are characterized by XRD, SEM, and TEM and tested in a single cell set‐up. While almost no current is gained from the membrane electrode assembly with the commercial ATO support, the cell with the home‐made ATO achieves a mediocre performance. This higher activity, however, is obtained with approximately half the Pt content compared to the catalyst with the commercial support. The different behaviours of both ATO powders can therefore mainly be attributed to differences in the specific support morphology.  相似文献

Cystic fibrosis (CF) is caused by mutations in the apical chloride channel cystic fibrosis transmembrane conductance regulator (CFTR) with 90% of patients carrying at least one deletion of the F508 (ΔF508) allele. This mutant form of CFTR is characterized by a folding and trafficking defect that prevents exit from the endoplasmic reticulum. We previously reported that ΔF508 CFTR can be recovered in a complex with Hsp90 and its co-chaperones as an on-pathway folding intermediate, suggesting that Δ508 CF disease arises due to a failure of the proteostasis network (PN), which manages protein folding and degradation in the cell. We have now examined the role of FK506-binding protein 8 (FKBP8), a component of the CFTR interactome, during the biogenesis of wild-type and ΔF508 CFTR. FKBP8 is a member of the peptidylprolyl isomerase family that mediates the cis/trans interconversion of peptidyl prolyl bonds. Our results suggest that FKBP8 is a key PN factor required at a post-Hsp90 step in CFTR biogenesis. In addition, changes in its expression level or alteration of its activity by a peptidylprolyl isomerase inhibitor alter CFTR stability and transport. We propose that CF is caused by the sequential failure of the prevailing PN pathway to stabilize ΔF508-CFTR for endoplasmic reticulum export, a pathway that can be therapeutically managed.  相似文献

Oocyte membrane fragments bind specifically radioiodinated VLD lipoprotein (very-low density lipoprotein) and LD lipoprotein (low-density lipoprotein). Competitive binding assays showed 2-3 times more VLD lipoprotein than LD lipoprotein bound at 4 degrees C. Equilibrium-binding data revealed the presence of one class of non-interacting sites for VLD lipoprotein (kD 12 microgram/ml) and co-operative binding for LD lipoprotein. The binding of VLD lipoprotein showed a distinct pH maximum at 5.3, whereas an indistinct maximum at about pH 7.3 was observed for LD lipoprotein. Unlabelled VLD lipoprotein did compete with 125I-labelled LD lipoprotein binding, but unlabelled LD lipoprotein did not compete with 125I-labelled VLD lipoprotein binding. VLD lipoprotein binding was inhibited by HD lipoprotein (high-density lipoprotein), but not by lysozyme, collagen, poly-L-lysine or poly-L-arginine; LD lipoprotein binding was inhibited by lysozyme and collagen, but not by HD lipoprotein. On the basis of these studies, we suggest that: (1) VLD lipoprotein and LD lipoprotein enter the oocytes by a receptor-mediated transport mechanism; (2) the receptors for VLD lipoprotein and LD lipoprotein are distinct; and (3) the binding of LD lipoprotein to chicken oocyte membranes differs from that to other cell types.  相似文献

Previous analysis of Rendille household herd composition revealed a transition from camel to cattle ownership for sedentary impoverished Rendille pastoralists of northern Kenya. In an attempt to delineate determinants of livestock holdings, logistic regression analysis of 112 household herds from the Rendille settlement of Korr, Marsabit District, Kenya was undertaken. Results indicated that household wealth, measured in present livestock holdings, past drought losses, and livestock sales, formed better predictors of cattle ownership than did household characteristics pertaining to labor supply, wage earners, age-set membership, and birth order of household head. These results are discussed in light of pastoral strategies designed to minimize risk.  相似文献

Studies of population dynamics, movement patterns and animal behavior usually require identification of individuals. We evaluated the reliability of using whisker spot patterns to noninvasively identify individual polar bears Ursus maritimus . We obtained the locations of polar bear whisker spots from photographs taken in western Hudson Bay, tested the independence of spot locations, estimated the complexity of each spot pattern in terms of information and determined whether each whisker spot pattern was reliable from its information content. Of the 50 whisker spot patterns analyzed, 98% contained enough information to be reliable, and this result varied little among observers. Photographs taken <50 m from polar bears were most useful. Our results suggest that individual identification of polar bears in the field based on whisker spot pattern variations is reliable. Researchers studying polar bear behavior or estimating population parameters can benefit from this method if proximity to the bears is feasible.  相似文献

We have identified recD mutants of Salmonella typhimurium by their ability to support growth of phage P22 abc (anti-RecBCD) mutants, whose growth is prevented by normal host RecBCD function. As in Escherichia coli, the recD gene of S. typhimurium lies between the recB and argA genes at min 61 of the genetic map. Plasmids carrying the Salmonella recBCD+ genes restore ATP-dependent exonuclease V activity to an E. coli recBCD deletion mutant. The new Salmonella recD mutations (placed on this plasmid) eliminate the exonuclease activity and enable the plasmid-bearing E. coli deletion mutant to support growth of phage T4 gene 2 mutants. The Salmonella recD mutations caused a 3- to 61-fold increase in the ability of a recipient strain to inherit (by transduction) a large inserted element (MudA prophage; 38 kb). In this cross, recombination events must occur in the short (3-kb) sequences that flank the element in the 44-kb transduced fragment. The effect of the recD mutation depends on the nature of the flanking sequences and is likely to be greatest when those sequences lack a Chi site. The recD mutation appears to minimize fragment degradation and/or cause RecBC-dependent recombination events to occur closer to the ends of the transduced fragment. The effect of a recipient recD mutation was eliminated if the donor P22 phage expressed its Abc (anti-RecBC) function. We hypothesize that in standard (high multiplicity of infection) P22-mediated transduction crosses, recombination is stimulated both by Chi sequences (when present in the transduced fragment) and by the phage-encoded Abc protein which inhibits the host RecBCD exonuclease.  相似文献
