Two cell populations in sunflower root meristems are described. Most cells stop in G1 after being cultured in sucrose-deficient medium, but “leaky” cells continue through DNA synthesis and stop in G2. A comparison of “leaky” and arrested cells is reported on the basis of radiosensitivity, and cytological and biochemical responses to metabolic inhibitors. “Leaky” cells are randomly distributed throughout primary meristematic tissues. They are not inhibited from initiating DNA synthesis by exposure to doses of γ-irradiation ranging from 300–7200 R; arrested cells, depending upon the dose, are inhibited partially or completely. “Leaky” cells do, however, show a dose-dependent mitotic delay in G2, which is the same as arrested cells. Treatment with puromycin and actidione does not inhibit “leaky” cells from initiating DNA synthesis but does inhibit them from mitosis. Arrested cells are inhibited from advancing to S and M by both inhibitors. Also, puromycin and actidione cause a decrease in protein and RNA synthesis, demonstrating a possible protein dependent RNA synthesis necessary for cell cycle progression. Actinomycin D (10 μg/ml) inhibits neither “leaky” nor arrested cells from entering S and M. At 30 μg/ml, however, arrested cells are partially inhibited. “Leaky” cell metabolism is unique in preparation for and initiation of DNA synthesis but similar to that of the remaining cells of the meristem in terms of requirements for progression through the rest of the mitotic cycle.  相似文献

We present results from a field study of inorganic carbon (C) acquisition by Ross Sea phytoplankton during Phaeocystis‐dominated early season blooms. Isotope disequilibrium experiments revealed that HCO3? was the primary inorganic C source for photosynthesis in all phytoplankton assemblages. From these experiments, we also derived relative enhancement factors for HCO3?/CO2 interconversion as a measure of extracellular carbonic anhydrase activity (eCA). The enhancement factors ranged from 1.0 (no apparent eCA activity) to 6.4, with an overall mean of 2.9. Additional eCA measurements, made using membrane inlet mass spectrometry (MIMS), yielded activities ranging from 2.4 to 6.9 U · [μg chl a]?1 (mean 4.1). Measurements of short‐term C‐fixation parameters revealed saturation kinetics with respect to external inorganic carbon, with a mean half‐saturation constant for inorganic carbon uptake (K1/2) of ~380 μM. Comparison of our early springtime results with published data from late‐season Ross Sea assemblages showed that neither HCO3? utilization nor eCA activity was significantly correlated to ambient CO2 levels or phytoplankton taxonomic composition. We did, however, observe a strong negative relationship between surface water pCO2 and short‐term 14C‐fixation rates for the early season survey. Direct incubation experiments showed no statistically significant effects of pCO2 (10 to 80 Pa) on relative HCO3? utilization or eCA activity. Our results provide insight into the seasonal regulation of C uptake by Ross Sea phytoplankton across a range of pCO2 and phytoplankton taxonomic composition.  相似文献

The availability of accurate and fast artificial intelligence (AI) solutions predicting aspects of proteins are revolutionizing experimental and computational molecular biology. The webserver LambdaPP aspires to supersede PredictProtein, the first internet server making AI protein predictions available in 1992. Given a protein sequence as input, LambdaPP provides easily accessible visualizations of protein 3D structure, along with predictions at the protein level (GeneOntology, subcellular location), and the residue level (binding to metal ions, small molecules, and nucleotides; conservation; intrinsic disorder; secondary structure; alpha-helical and beta-barrel transmembrane segments; signal-peptides; variant effect) in seconds. The structure prediction provided by LambdaPP—leveraging ColabFold and computed in minutes—is based on MMseqs2 multiple sequence alignments. All other feature prediction methods are based on the pLM ProtT5. Queried by a protein sequence, LambdaPP computes protein and residue predictions almost instantly for various phenotypes, including 3D structure and aspects of protein function. LambdaPP is freely available for everyone to use under embed.predictprotein.org , the interactive results for the case study can be found under https://embed.predictprotein.org/o/Q9NZC2 . The frontend of LambdaPP can be found on GitHub ( github.com/sacdallago/embed.predictprotein.org ), and can be freely used and distributed under the academic free use license (AFL-2). For high-throughput applications, all methods can be executed locally via the bio-embeddings ( bioembeddings.com ) python package, or docker image at ghcr.io/bioembeddings/bio_embeddings , which also includes the backend of LambdaPP.  相似文献

EEG changes were studied in the case of real movements (wrestlers training) and also in the case of imagination of motor activity (ideomotor training). Statistically significant increases were found of the average frequency of oscillations in the EEG alpha-range and simultaneously typical vegetative changes. Thus, imagination of motor activity may serve as a model for studying the CNS activation processes in connection with the realization of real motor activity.  相似文献

The goal is to present how shared decision-making in paediatric oncology occurs from the viewpoints of parents and physicians. Eight Swiss Pediatric Oncology Group centres participated in this prospective study. The sample comprised a parent and physician of the minor patient (<18 years). Surveys were statistically analysed by comparing physicians’ and parents’ perspectives and by evaluating factors associated with children’s actual involvement. Perspectives of ninety-one parents and twenty physicians were obtained for 151 children. Results indicate that for six aspects of information provision examined, parents’ and physicians’ perceptions differed. Moreover, parents felt that the children were more competent to understand diagnosis and prognosis, assessed the disease of the children as worse, and reported higher satisfaction with decision-making on the part of the children. A patient’s age and gender predicted involvement. Older children and girls were more likely to be involved. In the decision-making process, parents held a less active role than they actually wanted. Physicians should take measures to ensure that provided information is understood correctly. Furthermore, they should work towards creating awareness for systematic differences between parents and physicians with respect to the perception of the child, the disease, and shared decision-making.  相似文献

Parkinson's disease is characterized by progressive cell death in the substantia nigra pars compacta, which leads to dopamine depletion in the striatum and indirectly to cortical dysfunction. Increased glutamatergic transmission in the basal ganglia is implicated in the pathophysiology of Parkinson's disease and glutamate receptor mediated excitotoxicity has been suggested to be one of the possible causes of the neuronal degeneration. In the present study, the effects of serotonin, gamma-aminobutyric acid and bone marrow cells infused intranigrally to substantia nigra individually and in combination on unilateral 6-hydroxydopamine induced Parkinson's rat model was analyzed. Scatchard analysis of total glutamate and NMDA receptor binding parameters showed a significant increase in Bmax (P < 0.001) in the cerebral cortex of 6-hydroxydopamine infused rat compared to control. Real Time PCR amplification of NMDA2B, mGluR5, bax, and ubiquitin carboxy-terminal hydrolase were up regulated in cerebral cortex of 6-hydroxydopamine infused rats compared to control. Gene expression studies of GLAST, ά-Synuclien and Cyclic AMP response element-binding protein showed a significant (P < 0.001) down regulation in 6-OHDA infused rats compared to control. Behavioural studies were carried out to confirm the biochemical and molecular studies. Serotonin and GABA along with bone marrow cells in combination showed reversal of glutamate receptors and behaviour abnormality shown in the Parkinson's rat model. The therapeutic significance in Parkinson's disease is of prominence.  相似文献
