As expected from its short renewal time, the hypertrophic Lingese Reservoir responded rapidly but incompletely to external nutrient reduction in 1993. Although there was a sudden decline in lake phosphorus and chlorophyll concentrations, the ‘turbid regime’ revealed resistance in this weakly stratified reservoir of intermediate depth. Draining the reservoir in autumn 1995 provided an unprecedented opportunity for sediment treatment—the phosphorus-rich surface layer was inverted and covered with deeper nutrient-poor layers—and removal of the dense cyprinid fish stock which had not responded to sewage diversion, at least in the short term. Commencing with refilling in 1999, a new fish stock was built up from 2000 by only stocking predators (fingerlings of pike, pike-perch and larger specimens of rainbow-trout in the first years) in combination with catch restrictions. Concomitantly, with the appearance of daphnids in 1999, a ‘clear water regime’ was established and lake water phosphorus concentrations decreased at unchanged external loading. Reduced zooplanktivory as well as reduced fish-mediated phosphorus release from the sediments were driving mechanisms behind successful reduction of internal loading and achievement of a ‘clear water regime’. Hence, phosphorus concentrations were revealed to be a response variable not only to input management but also to food-web management. As the development of cyprinid dominance was prevented in the long-term, there is ample evidence that the fish community responded to the applied management measures as expected featuring successful food-web management. Overall, the biological structure was revealed to be of major importance for lake phosphorus availability and turbidity as in shallow lakes, without, however, the establishment of macrophyte dominance.  相似文献

A quantitative endonuclease assay, which relies on the introduction of single and double strand breaks into supercoiled plasmid DNA, was used to study the activity of the extracellular nuclease of Serratia marcescens SM6 in buffer and in groundwater. The parallel enzyme concentration-dependent production of relaxed and linear plasmid molecules suggests that the nuclease produces single and double strand breaks in duplex DNA. Bovine serum albumin stimulated the nuclease activity towards DNA and RNA and increased the stability of the enzyme against thermal inactivation. The DNase activity at 4 °C and 50 °C was almost half of that at the optimum temperature (37 °C). The nuclease was active in groundwater, although the specific activity was lower than in buffer. In a groundwater aquifer microcosm, mineral-adsorbed transforming DNA was substantially less accessible to the nuclease than was dissolved DNA. The data suggest that the extracellular nuclease of Serratia marcescens may contribute to DNA turnover in the environment and that adsorption of DNA to minerals provides protection against the nuclease.Abbreviations GW groundwater GWA groundwater aquifer  相似文献

This study examined the superoxide dismutase 3 (SOD3) R231G polymorphism in relation to the severity of coronary artery disease (CAD) and the risk of myocardial infarction (MI) in 3211 individuals; 94.4% of study participants were homozygous for SOD3 231RR and 5.5% were heterozygous for SOD3 231RG. The odds ratios of the RG and GG genotype (adjusted for age, gender and for conventional cardiovascular risk factors) were 2.02 (95% CI, 1.23–3.33, p=0.005) for the highest vs the lowest Friesinger coronary score and 1.40 (95% CI, 1.02–1.92, p=0.037) for MI, respectively. Further the SOD3 RG and GG genotype was associated with lower alpha-tocopherol levels than the wild type SOD3 RR genotype. It is concluded that the SOD3 231RG and GG genotype is associated with lower alpha-tocopherol levels and the severity of CAD and the risk of MI.  相似文献

Background and Aims

Cold neutron radiography was applied to directly observe embolism in conduits of liana stems with the aim to evaluate the suitability of this method for studying embolism formation and repair. Potential advantages of this method are a principally non-invasive imaging approach with low energy dose compared with synchrotron X-ray radiation, a good spatial and temporal resolution, and the possibility to observe the entire volume of stem portions with a length of several centimetres at one time.

Methods

Complete and cut stems of Adenia lobata, Aristolochia macrophylla and Parthenocissus tricuspidata were radiographed at the neutron imaging facility CONRAD at the Helmholtz-Zentrum Berlin für Materialien und Energie, with each measurement cycle lasting several hours. Low attenuation gas spaces were separated from the high attenuation (water-containing) plant tissue using image processing.

Key results

Severe cuts into the stem were necessary to induce embolism. The formation and temporal course of an embolism event could then be successfully observed in individual conduits. It was found that complete emptying of a vessel with a diameter of 100 µm required a time interval of 4 min. Furthermore, dehydration of the whole stem section could be monitored via decreasing attenuation of the neutrons.

Conclusions

The results suggest that cold neutron radiography represents a useful tool for studying water relations in plant stems that has the potential to complement other non-invasive methods.  相似文献

Glycogen synthase kinase-3 (GSK-3) is a key player in various important signaling pathways in animals. The activity of GSK-3 is known to be modulated by protein phosphorylation and differential complex formation. However, little information is available regarding the function and regulation of plant GSK-3/shaggy-like kinases (GSKs). Analysis of the in vivo kinase activity of MsK1, a GSK from Medicago sativa, revealed that MsK1 is active in healthy plants and that MsK1 activity is down-regulated by the elicitor cellulase in a time- and dose-dependent manner. Surprisingly, cellulase treatment triggered the degradation of the MsK1 protein in a proteasome-dependent manner suggesting a novel mechanism of GSK-3 regulation. Inhibition of MsK1 kinase activity and degradation of the protein were two successive processes that could be uncoupled. In a transgenic approach, stimulus-induced inhibition of MsK1 was impeded by constant replenishment of MsK1 by a strong constitutive promoter. MsK1 overexpressing plants exhibited enhanced disease susceptibility to the virulent bacterial pathogen Pseudomonas syringae. MAP kinase activation in response to pathogen infection was compromised in plants with elevated MsK1 levels. These data strongly suggest that tight regulation of the plant GSK-3, MsK1, may be important for innate immunity to limit the severity of virulent bacterial infection.  相似文献

Approximately 13% of the human genome can fold into non-canonical (non-B) DNA structures (e.g. G-quadruplexes, Z-DNA, etc.), which have been implicated in vital cellular processes. Non-B DNA also hinders replication, increasing errors and facilitating mutagenesis, yet its contribution to genome-wide variation in mutation rates remains unexplored. Here, we conducted a comprehensive analysis of nucleotide substitution frequencies at non-B DNA loci within noncoding, non-repetitive genome regions, their ±2 kb flanking regions, and 1-Megabase windows, using human-orangutan divergence and human single-nucleotide polymorphisms. Functional data analysis at single-base resolution demonstrated that substitution frequencies are usually elevated at non-B DNA, with patterns specific to each non-B DNA type. Mirror, direct and inverted repeats have higher substitution frequencies in spacers than in repeat arms, whereas G-quadruplexes, particularly stable ones, have higher substitution frequencies in loops than in stems. Several non-B DNA types also affect substitution frequencies in their flanking regions. Finally, non-B DNA explains more variation than any other predictor in multiple regression models for diversity or divergence at 1-Megabase scale. Thus, non-B DNA substantially contributes to variation in substitution frequencies at small and large scales. Our results highlight the role of non-B DNA in germline mutagenesis with implications to evolution and genetic diseases.  相似文献

Research in autophagy continues to accelerate,(1) and as a result many new scientists are entering the field. Accordingly, it is important to establish a standard set of criteria for monitoring macroautophagy in different organisms. Recent reviews have described the range of assays that have been used for this purpose.(2,3) There are many useful and convenient methods that can be used to monitor macroautophagy in yeast, but relatively few in other model systems, and there is much confusion regarding acceptable methods to measure macroautophagy in higher eukaryotes. A key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers of autophagosomes versus those that measure flux through the autophagy pathway; thus, a block in macroautophagy that results in autophagosome accumulation needs to be differentiated from fully functional autophagy that includes delivery to, and degradation within, lysosomes (in most higher eukaryotes) or the vacuole (in plants and fungi). Here, we present a set of guidelines for the selection and interpretation of the methods that can be used by investigators who are attempting to examine macroautophagy and related processes, as well as by reviewers who need to provide realistic and reasonable critiques of papers that investigate these processes. This set of guidelines is not meant to be a formulaic set of rules, because the appropriate assays depend in part on the question being asked and the system being used. In addition, we emphasize that no individual assay is guaranteed to be the most appropriate one in every situation, and we strongly recommend the use of multiple assays to verify an autophagic response.  相似文献

We examined the effect of cadmium (Cd) additions on a GDH1-null line of maize and its wild-type isogenic sibling. Addition of Cd increases the synthesis of metallothioneines which are glutamate- and cysteine-rich peptides. We predicted a reduced synthesis of γ-glutamylcysteine (γEC) peptides in the mutant relative to the wild type if glutamate dehydrogenase (GDH) was limiting the drainage of carbon from the tricar-boxylic acid cycle (TCAC). In our experiments there were similar increases in levels of γEC peptides in both mutant and wild-type seedlings in response to Cd. There was a marked increase in the phosphoenolpyruvate carboxylase (PEPcase) polypeptide and in one of the polypeptide bands of glutamine synthetase in both mutant and wild-type seedlings. However, no change was seen in the polypeptide levels of GDH or glutamate synthase (GOGAT). Thus, in contrast to PEPcase, an enhanced carbon drain from the TCAC in response to Cd exposure does not require enhanced levels of either GDH or GOGAT polypeptides.  相似文献
