Oscillations and efficiency in glycolysis 总被引:6,自引:0,他引:6
We suggest that temporal oscillations of concentrations of intermediates in biochemical reaction systems may enhance the efficiency of free energy conversion (reduce dissipation) in those reactions. Experiments on glycolysis are used to estimate the Gibbs free energy changes along the glycolysis mechanism, and to postulate a construct for the glycolysis "machine" which involves: the PFK reaction as the primary oscillophor; the GAPDH reaction as a phase-shifting device; and the PK reaction with the property of intrinsic oscillatory response at resonance with the driving frequency. Analysis of a simple reaction mechanism with these postulated properties shows that the conversion of free energy from reactants to products is more efficient in an oscillatory than a steady state operation. The efficiency of free energy conversion in glycolysis from glucose + ADP to products + ATP is estimated to be increased by 5--10% due to oscillations. This may have been relevant for the evolutionary development of oscillations such as in glycolysis, especially in anaerobic cells. 相似文献

The binding of NAD+, NADH, and ADP-ribose to horse liver alcohol dehydrogenase has been studied calorimetrically as a function of pH at 25 degrees C. The enthalpy of NADH binding is 0 +/- 0.5 kcal mol-1 in the pH range 6 to 8.6. The enthalpy of NAD+ binding, however, varies with pH in a sigmoidal fashion and is -4.0 kcal mol(NAD)-1 at pH 6.0 and +4.5 kcal mol(NAD)-1 at pH 8.6 with an apparent pKa of 7.6 +/- 0.2. The enthalpy of proton ionization of the group on the enzyme is calculated to be in the range 8.8 to 9.8 kcal mol(H+)-1. In conjunction with the available thermodynamic data on the ionization of zinc-bound water in model compounds, it is concluded that the group with a pKa of 9.8 in the free enzyme and 7.6 in the enzyme . NAD+ binary complex is, most likely, the zinc-bound water molecule. Our studies with zinc-free enzyme provide further evidence for this conclusion. Therefore, the processes involving a conformational change of the enzyme upon NAD+ binding and the suggested mechanism of subsequent quenching of the fluorescence of Trp-314 implicating the participation of an ionized tyrosine group must be re-evaluated in the light of this thermodynamic study. 相似文献

A field study was made to test whether the population size of a diurnal reef fish, the wrasse Thalassoma bifasciatum (Bloch), was limited by inter- or intraspecific competition for sleeping shelter. T. bifasciatum is often attacked at dusk by two small territorial damselfishes, Eupomacentrus dorsopunicans (Poey) and E. planifrons (Cuvier). Although these three species sleep in the same general habitats, there are qualitative differences in the types of holes they use and how they use them. Wrasse holes are usually in these damselfishes' territories, but damselfish attacks do not prevent wrasses entering holes. Wrasses infrequently defend their holes intraspecifically. They regularly change their holes, with little intra- or interspecific aggressive interaction. When its hole is removed, a wrasse is late in retiring but finds a hole near its old one with little aggressive interaction, and does not have a higher mortality rate. Empty wrasse holes are rarely refilled, and then only by conspecifics. Wrasses added to reefs find unoccupied holes and do not usurp other fishes' holes. Damselfish defend their eggs and food against the wrasse, but not their sleeping shelter, nor living space per se. Sleeping sites are not limiting the wrasse, but are present in a surplus. Intraspecific hole defense by a wrasse prevents a delay in its retiring that would increase the risk of crepuscular predation on it. 相似文献

Using lettuce (Lactuca sativa L., cv. Grand Rapids) embryos in osmotica, we have demonstrated that when the growth rates of the embryonic axes of seeds treated with red (R) or far-red (FR) light are equalized, the axes of R-treated seeds develop a 3.4-bar decrease in water potential (paper No. III).As axial growth begins, reserve protein and phytin decrease rapidly, concomitant with increases in reducing sugars, -amino nitrogen, and inorganic and esterified soluble phosphates. However, no differences between the axes of R-and FR-treated seeds are found with respect to the changes in these compounds, indicating that these changes arise as a result of growth and are not under immediate phytochrome control. Little change in the total lipid content is found in either treatment. The axes of FR-treated seeds hydrolyze endogenous sucrose at a greater rate thant those of R-treated seeds. Axes of R-treated seeds accumulate K+ and Na+ to a greater extent than those of FR-treated seeds. When potassium salts are added to the incubation medium, R induces increased K+ uptake by the axis and greater medium acidification by the axis. Malate and other organic acids and acidic amino acids increase at equal rates in both treatments, indicating that inorganic anions may also be taken up to balance the ionic charges. The results are compatible with the assumption that changes in the osmotic and pressure potentials of the embryonic axes of R-treated seeds are the result of a phytochrome-stimulated proton pump which, in whole dormant seeds, would initiate water-potential changes allowing the embryos to overcome the mechanical restraint of the surrounding seed layers, resulting in germination.Abbreviations FR

The influence of several plant growth regulators on the growth of the embryonic axes from red- and far-red-(R- and FR-)treated lettuce (Lactuca sativa L., cv. Grand Rapids) seeds was examined; as shown previously, the water potential of the axes from R-treated seeds has been lowered by 3.5–5.6 bars compared to that in axes from FR-treated ones. Kinetin and abscisic acid (ABA), when included in the incubation medium, reduced the elongation of the axes whereas fusicoccin stimulated it; however, these effects were the same in axes of both R- and FR-treated seeds. In contrast, elongation of axes from FR-treated seeds was stimulated by gibberellic acid (GA3, but elongation of axes from R-treated ones was not affected by this hormone. This latter result indicates that gibberellins may be involved in the phytochrome-mediated growth responses in lettuce axes.When the root caps of the embryos were removed prior to light treatment, R was still able to induce a water-potential decrease in the embryonic axes, indicating that at least a portion of the active Pfr resides in the axis and not the root cap.Abbreviations ABA

The volume of cells grown in tissue culture following exposure to a wide variety of cytotoxic drugs or x-rays increases at a rate of 1 to 10% of cell mass per hour. The same phenomenon is seen in animal neoplasias and human leukemias. This increase in cell volume is the result of unbalanced cell growth with a resulting disproportionate synthesis of proteins and possibly other macromolecules in the cytoplasm and nucleus. The dose response curve for a decrease in cell survival as measured by cloning efficiency in tissue culture is quantitatively correlated with the dose response curve for inducing an increase in cell volume. This quantitative relationship makes feasible the use of the phenomenon of unbalanced cell growth as a measure of cell death in screening for cytotoxic drugs or in monitoring response to therapy. An hypothesis to explain this increase in cell volume following chemotherapy is that cells are by the action of these drugs induced into an abortive or unbalanced pseudo-cycle which is characterized by synthesis of substantial amounts of protein without other preparative steps for cell division. 相似文献

Summary Detailed growth analyses of cultured skin fibroblasts from two patients with Huntington's Disease (HD) were compared with those from controls matched for age and sex. In contrast to control cells, HD fibroblasts plated more efficiently at the low seeding densities used. Subsequent exponential growth of HD cultures was more stable towards routine trypsinisation than that of controls. However, the most striking feature of HD cultures was their ability to grow to significantly higher cell saturation densities. Inoculated, irradiated pork (2,300 cans) and chicken (2,000 cans) pack studies were performed to establish the 12D dose for these foods. Each can was inoculated with a mixture of 10(6) spores of each of 10 strains of Clostridium botulinum (five type A and five type B), or a total of 10(7) spores. The cans received a series of increasing doses of gamma rays (60Co) at -30 +/- 10 degrees C; they were incubated for 6 months at 30 +/- 2 degrees C and examined for swelling, toxicity, and recoverable botulinal cells. The highest rate of swelling for both foods occurred within the first week of incubation, and maximum swelling was observed within 4 to 5 weeks. The minimal experimental sterilizing dose (ESD) based on flat, nontoxic sterile cans was 3.0 less than ESD less than or equal to 3.2 Mrad for pork and 4.0 less than ESD less than or equal to 4.2 Mrad for chicken. An analysis of the partial spoilage data by extreme-value statistics indicated with 90% confidence that the rate of spore death in the two foods was not a normal distribution, but appeared to favor a shifted exponential function. Based on the latter distribution, and assuming one most resistant strain in the mixture of 10 used, the 12D dose computed to 4.37 Mrad, with a shoulder of 0.11 Mrad, for pork and to 4.27 Mrad, with a shoulder of 0.51 Mrad, for chicken. An assumption that there were two or more most resistant strains in the inoculum progressively lowered the 12D dose. There was an apparent antagonism between the irradiated type A and B viable strains in the two foods. Cans with type B cells and toxin predominated over cans with type A cells and toxin, but cans with a mixture of type A and B toxins predominated over cans with a mixture of Type A and B cells. At the highest sublethal doses, only type A cells survived in pork, but in chicken there was a least one type B strain that was at least as resistant as type A strains. 相似文献
