After induction of a perivenous liver cell necrosis by CCl4 pretreatment of the rat, ammonia uptake by perfused liver is decreased. This was due to an inhibition of glutamine synthesis from added ammonia, whereas urea synthesis was not affected by CCl4 pretreatment. The data confirm recent findings on hepatocyte heterogeneity in ammonia metabolism and are explained by an impairment of perivenous glutamine synthetase, but not of periportal urea synthesis, by the perivenous liver cell necrosis induced by CCl4. Regarding the pathogenesis of hyperammonemia in acute severe liver disease like CCl4 poisoning, the data point to a role of an impaired glutamine synthesis, but not to an impairment of urea synthesis. 相似文献

Nanopore sequencing promises long read-lengths and single-molecule resolution, but the stochastic motion of the DNA molecule inside the pore is, as of this writing, a barrier to high accuracy reads. We develop a method of statistical inference that explicitly accounts for this error, and demonstrate that high accuracy (>99%) sequence inference is feasible even under highly diffusive motion by using a hidden Markov model to jointly analyze multiple stochastic reads. We address the problem of the possible significance of biological
speciation to the magnitude and pattern of divergence of asexually
transmitted characters in bisexual species. The empirical data for this
report consist of restriction endonuclease site variability in maternally
transmitted mitochondrial DNA (mtDNA) isolated from 82 samples of
Peromyscus polionotus and P. leucopus collected from major portions of the
respective species' ranges. Data are analyzed together with previously
published information on P. maniculatus, a sibling species to polionotus.
Maps of restriction sites indicate that all of the variation observed can
be reasonably attributed to base substitutions leading to loss or gain of
particular recognition sites. Magnitude of mtDNA sequence divergence within
polionotus (maximum approximately equal to 2%) is roughly comparable to
that observed within any of five previously identified mtDNA assemblages in
maniculatus. Sequence divergence within leucopus (maximum approximately
equal to 4%) is somewhat greater than that within polionotus. Consideration
of probable evolutionary links among mtDNA restriction site maps allowed
estimation of matriarchal phylogenies within polionotus and leucopus.
Clustering algorithms and qualitative Wagner procedures were used to
generate phenograms and parsimony networks, respectively, for the
between-species comparisons. Three simple graphical models are presented to
illustrate some conceivable relationships of mtDNA differentiation to
speciation. In theoretical case I, each of two reproductively defined
species (A and B) is monophyletic in matriarchal genealogy; the common
female ancestor of either species can either predate or postdate the
speciation. In case II, neither species is monophyletic in matriarchal
genotype. In case III, species B is monophyletic but forms a subclade
within A which is thus paraphyletic with respect to B. The empirical
results for mtDNA in maniculatus and polionotus appear to conform closely
to case III. These theoretical and empirical considerations raise a number
of questions about the general relationship of the speciation process to
the evolution of uniparentally transmitted traits. Some of these
considerations are presented, and it is suggested that the distribution
patterns of mtDNA sequence variation within and among extant species should
be of considerable relevance to the particular demographies of speciation.

The levels of apparent DNA single-strand breaks (ssb) were measured, following a 0-20 h incubation of normal human skin fibroblasts exposed to the solar uv wavelengths produced by a fluorescent sunlamp. The ssb were determined using the alkaline elution assay, which was performed either without proteinase K (proK) or in its presence, so as to eliminate any DNA-protein crosslinks that may be present in the cells. Cells were irradiated with either 3 kJ/m2 of sunlamp uv greater than 295 nm, 150 kJ/m2 of sunlamp uv greater than 315 nm, or 150 kJ/m2 of sunlamp uv greater than 320 nm. These treatments resulted in the production of 5-10 ssb/10(10) Da. For the two shorter wavelength irradiations, the levels of ssb decreased rapidly upon incubation of the cells. However, when the elutions were performed using proK, the number of ssb increased about twofold following a 2-4 h incubation. In contrast, the levels of ssb decreased in the sunlamp uv greater than 320 nm irradiated cells for elutions performed with or without proK. These results suggest that under certain irradiation conditions, ssb are formed in cells upon incubation, which are hidden by the crosslinking of protein to DNA. 相似文献

DNA-protein crosslinks (DPC) were measured following exposure to the solar UV wavelengths produced by a fluorescent sunlamp in ICR 2A frog cells and two solar UV-sensitive mutants derived from this cell line. Approx. 5-7 DPC per 10(10) dalton were induced in these cells by either 150 kJ/m2 of sunlamp UV greater than 315 nm plus photoreactivating light (PRL) or 10 kJ/m2 of sunlamp UV greater than 295 nm. The irradiated cells were then incubated for 0-24 h and the level of DPC measured using alkaline elution. It was found for the ICR 2A cells exposed to sunlamp UV greater than 315 nm that the level of DPC increased about 3-fold during a 2-h postirradiation incubation and then decreased. The mutant cell lines also showed an enhancement in the level of DPC following irradiation, although it was much less pronounced and the levels decreased much more rapidly. In a similar fashion, the level of DPC increased in ICR 2A cells exposed to sunlamp UV greater than 295 nm with more than a 5-fold enhancement after a 4-h incubation. Once again, the mutant cell lines showed an increase in the level of DPC that was smaller and more transient than the effect in the ICR 2A cells. These results suggests that this enhancement in DPC may be indicative of a process that plays a role in cellular survival following solar UV-irradiation. 相似文献

DNA synthesis was examined in ultraviolet (uv)-irradiated ICR 2A frog cells in which either pyrimidine dimers or nondimer photoproducts represented the major class of DNA lesions. Dimers were induced by exposure of cells to 254 nm uv, while nondimer photoproducts were induced by irradiation of cells with uv produced by a fluorescent sunlamp (FSL) that was filtered through 48A Mylar (removes wavelengths less than 310 nm). The FSL-irradiated cultures were also treated with photoreactivating light (PRL) which removed most of the small number of dimers induced by the irradiation, leaving a relatively pure population of nondimer photoproducts. In addition, cells were exposed to 60Co gamma rays. The cultures were pulse-labeled and the size distribution of the DNA synthesized was estimated using both sucrose gradient sedimentation and alkaline step elution. Using either of these techniques, it was found that the presence of dimers resulted in a reduction principally in the synthesis of high molecular weight (MW) DNA. In contrast, nondimer photoproducts caused a strong inhibition in the synthesis of low MW DNA, as was also observed in gamma-irradiated cells. Hence the induction of pyrimidine dimers in DNA mainly affected the elongation of replicons, whereas nondimer lesions primarily caused an inhibition of replicon initiation. 相似文献
