Histochemical methods, especially azo dye methods for detecting acid phosphomonoesterase activity were applied to normal, regenerating and denervated, amputated limbs from larval Amblystoma maculatum. Efforts were made to control inactivation of enzymic activity and diffusion of both enzyme and reaction product. “Base-line” values for enzymic activity were determined for normal limbs. Activity appeared most intense in macrophages, less intense in epidermis and cartilage matrix. Some activity was detected in Schwann cells, peri- and endoneurium and muscle fibers form normal limbs. Enzymic activity in regenerating limbs was strongest within macrophages which appeared in increased numbers especially in early stages. Wound tissue showed little increased activity. As the blastema formed, increased enzymic activity was detected in epidermis and within increased numbers of macrophages. Chondrocytes showed increased activity especially during cartilage matrix deposition. Amputated, denervated limbs showed large numbers of active macrophages beneath and within epidermis and along muscle. As regression commenced, areas of cartilage matrix breakdown showed increased enzymic activity but, in general, greatest activity was in macrophages. The various possible roles of acid phosphomonoesterase activity in the specific biological situations dealt with are considered in light of such observations. 相似文献

Pseudomonas aeruginosa produces phenazine-1-carboxylic acid (PCA) and pyocyanin (PYO), which aid its anaerobic survival by mediating electron transfer to distant oxygen. These natural secondary metabolites are being explored in biotechnology to mediate electron transfer to the anode of bioelectrochemical systems. A major challenge is that only a small fraction of electrons from microbial substrate conversion is recovered. It remained unclear whether phenazines can re-enter the cell and thus, if the electrons accessed by the phenazines arise mainly from cytoplasmic or periplasmic pathways. Here, we prove that the periplasmic glucose dehydrogenase (Gcd) of P. aeruginosa and P. putida is involved in the reduction of natural phenazines. PYO displayed a 60-fold faster enzymatic reduction than PCA; PCA was, however, more stable for long-term electron shuttling to the anode. Evaluation of a Gcd knockout and overexpression strain showed that up to 9% of the anodic current can be designated to this enzymatic reaction. We further assessed phenazine uptake with the aid of two molecular biosensors, which experimentally confirm the phenazines’ ability to re-enter the cytoplasm. The biology of fracture healing is better understood than ever before, with advancements such as the locking screw leading to more predictable and less eventful osseous healing. However, at times one’s intrinsic biological response, and even concurrent surgical stabilization, is inadequate. Fifty consecutive patients admitted for replantation surgery received a psychiatric evaluation. In 28 percent of the patients there was evidence of preaccident psychopathology. Fifty-four percent of the patients reported a recent stressful life event predating the accident. Fifty-four percent of the patients were assessed as having an adverse postoperative emotional reaction. The presence of preaccident psychopathology, evidence of family dysfunction, and a history of a recent stressful life event were positively linked with an adverse postoperative emotional reaction. In addition, patients with hand or arm replants were significantly less likely to require a psychiatric intervention than patients with leg replants. The psychological processes pertaining to the replantation journey are described in five stages: the preaccident period, the initial response, the stage of uncertainty, the recognition of loss, and acceptance and reintegration. Finally, the role of a liaison psychiatrist on a replantation unit is outlined and further research is strongly urged. 相似文献
