Aims Understanding the role of genetics in biological invasions has become an important aspect for modern plant ecology. Many studies suggest that increased ploidy level benefits the success of an invasive species, but the basis for this phenomenon is not fully understood. In its native, North American range, Solidago gigantea has three geo-cytotypes comprising di-, tetra- and hexaploid populations, while in Europe, where it is highly invasive, S. gigantea stands are composed primarily of tetraploid individuals. Our study investigates whether North American hexaploids can induce a greater risk of invasion, due to their higher performance in a non-native range, as compared to the existing tetraploids of that range.Methods We performed greenhouse and common garden experiments along with microsatellite analyses to test whether differences in chromosome number and origin of the species mean superior fitness in the introduced range.Important findings Genetic diversity was significantly higher in the native hexaploid populations (A R = 6.04; H e = 0.7794), rather than the non-native tetraploid populations (A R = 4.83; H e = 0.6869). Furthermore, differentiation between geo-cytotypes was moderate (ρ ST = 0.1838), which was also confirmed by their clear segregation in principal component analysis and structure analyses, proving their different genetic structure. In contrast to genetic diversity, the non-native tetraploid geo-cytotype performed better in the common garden experiment, implying that higher genetic diversity does not always mean better success. Our results suggest that native hexaploids do not present a greater risk, as assessed by their performance in the introduced range, when compared to the non-native tetraploids, as was suggested by previous studies. Nevertheless, their introduction is still undesirable due to their different genetic structure, which, through hybridization, could give a new drive to the invasion of S. gigantea .  相似文献

  • Environmental gradients, and particularly climatic variables, exert a strong influence on plant distribution and, potentially, population genetic diversity and differentiation. Differences in water availability can cause among‐population variation in ecological processes and can thus interrupt populations’ connectivity and isolate them environmentally. The present study examines the effect of environmental heterogeneity on plant populations due to environmental isolation unrelated to geographic distance.
  • Using AFLP markers, we analyzed genetic diversity and differentiation among 12 Salvia spinosa populations and 13 Salvia syriaca populations from three phytogeographical regions (Mediterranean, Irano‐Turanian and Saharo‐Arabian) representing the extent of the species’ geographic range in Jordan. Differences in geographic location and climate were considered in the analyses.
  • For both species, flowering phenology varied among populations and regions. Irano‐Turanian and Saharo‐Arabian populations had higher genetic diversity than Mediterranean populations, and genetic diversity increased significantly with increasing temperature. Genetic diversity in Salvia syriaca was affected by population size, while genetic diversity responded to drought in S. spinosa. For both species, high levels of genetic differentiation were found as well as two well‐supported phytogeographical groups of populations, with Mediterranean populations clustering in one group and the Irano‐Turanian and Saharo‐Arabian populations in another. Genetic distance was significantly correlated to environmental distance, but not to geographic distance.
  • Our data indicate that populations from moist vs. arid environments are environmentally isolated, where environmental gradients affect their flowering phenology, limit gene flow and shape their genetic structure. We conclude that environmental heterogeneity may act as driver for the observed variation in genetic diversity.

Based on experimental data, a kinetic model for the deactivation of partially purified pyruvate decarboxylase (PDC) by benzaldehyde (0-200 mM) in MOPS buffer (2.5 M) has been developed. An initial lag period prior to deactivation was found to occur. With first order dependencies of PDC deactivation on exposure time and on benzaldehyde concentration, a reaction time deactivation constant of 2.64×10-3 h-1 and a benzaldehyde deactivation coefficient of 1.98×10-4 mM-1 h-1 were determined for benzaldehyde concentrations up to 200 mM. The PDC deactivation kinetic equations established in this study are an essential component in an overall model being developed to describe the enzymatic biotransformation of benzaldehyde and pyruvate to produce the pharmaceutical intermediate (R)-phenylacetylcarbinol (R-PAC).  相似文献

Extracts of 14 filamentous fungi were examined regarding their potential for production of (R)-phenylacetylcarbinol [(R)-PAC], which is the chiral precursor in the manufacture of the pharmaceuticals ephedrine and pseudoephedrine. Benzaldehyde and pyruvate were transformed at a scale of 1.2 ml into PAC by cell-free extracts of all selected strains, covering the broad taxonomic spectrum of Ascomycota, Zygomycota and Basidiomycota. Highest final PAC concentrations were obtained with the extracts of Rhizopus javanicus and Fusarium sp. [78-84 mM (11.7-12.6 g/l) PAC within 20 h from initial substrate concentrations of 100 mM benzaldehyde and 150 mM pyruvate]. (R)-PAC was in about 90-93% enantiomeric excess. Rhizopus javanicus had the advantage of faster growth than Fusarium sp. Rhizopus javanicus mycelia were used as an example in a biotransformation process based on whole cells and benzaldehyde and glucose as substrates. The substrate pyruvate was generated through the fungal fermentation of glucose. Only 19 mM PAC (2.9 g/l) were produced within 8 h from 80 mM benzaldehyde. with evidence of significant benzyl alcohol production.  相似文献

Despite the acknowledged importance of prescribed fire in creating northern bobwhite (Colinus virginianus) breeding cover, little research has investigated bobwhite breeding season habitat selection relative to time since fire. In 2016 and 2017, we monitored radio-tagged bobwhite on a 17,000-ha portion of a military installation managed with frequent (every ~3 years) prescribed fires, applied during the growing and dormant seasons. We monitored bobwhite to determine which vegetation characteristics associated with prescribed burning were important to bobwhite breeding season habitat selection at the microsite (i.e., telemetry location compared to nearby random location) and the macrosite scale (i.e., the burn-unit containing the location compared to study area availability). During 2 breeding seasons, we collected 2,315 bobwhite locations and compared percent cover of vegetation, days since burn, basal area, and distance to key landscape features (e.g., stream, wildlife opening) at a subset of microsite locations (301 locations during 2016 and 890 locations during 2017) to paired random locations. At the microsite scale, bobwhite selected lower basal area of hardwoods, greater woody understory cover, greater other (not wiregrass [Aristida stricta]) grass cover, and greater forb cover than at random points. At the macrosite scale, bobwhite selected units with <4.6 m2/ha basal area (combined hardwoods and pines) in 2016 and units with <9.2 m2/ha basal area in 2017. At the macrosite scale, bobwhite selected for areas burned in the dormant season of the same year, avoided areas burned in the growing season of the same year, and used other times since last burn categories proportionate to their availability. The selection for a low basal area at both scales indicates prescribed fire effects would be limited by shading from dense overstory, and the shrubs, grasses, and forbs that provide essential cover for bobwhite during the breeding season will not develop. In lower productivity soil regions similar to our study area, we advise that thinning operations set target basal areas below 10 m2/ha to create and maintain breeding season habitat for northern bobwhite. © 2019 The Wildlife Society  相似文献

The mutation G1691A (R506Q) in the human factor V gene is associated with the resistance to activated protein C (APC) that represents a major risk of development of venous thrombosis. A population study of 180 unrelated individuals from south Germany was performed. Examination of the allelic frequencies revealed a high prevalence of this disease-related mutation (Q506, FV Leiden). The heterozygosity rate was 7.8% with a confidence interval between 4% and 11%.  相似文献

Objective: National surveys have pointed to a particularly high risk of pediatric overweight among U.S. Hispanics. However, the data have been primarily from the Mexican‐American community. We studied the prevalence of overweight and clinical comorbidities in children and youth of predominantly El Salvadoran ancestry. Research Methods and Procedures: A sample of 309 Hispanic youth, 6–18 years was surveyed from two inner city Washington, DC, clinics. BMI; triceps skinfold (TSF) and subscapular skinfold thickness (SSSF); bioelectrical impedance analysis (BIA); and blood pressure measures were obtained, along with information regarding physical activity, sedentary behavior, dietary history, family, and personal medical history. Results: Thirty‐eight percent were overweight (BMI ≥ 95th percentile) and 22% at risk for overweight (BMI 85–94th percentile). Thirty‐four percent had TSF ≥ 90th percentile and 29% had SSSF ≥ 90th percentile. Fifty‐one percent of males and 70% of females had body fat > 30%. Compared to their nonoverweight counterparts, overweight youth had significantly higher systolic blood pressure (111.4 ± 1.3 vs. 104.5 ± 0.9 mm Hg, p < 0.0001). Among children younger than 11 years, overweight was associated with onset of adrenarche (23% vs. 10%, p = 0.01). Participation in one or more sports teams was negatively correlated with overweight) p = 0.04). Discussion: The prevalence of overweight and at risk for overweight in this sample was twice the national average for U.S. children and 1.7 times greater than that of Mexican‐American children in national surveys. Overweight was associated with advanced pubertal development, high body fat, elevated blood pressure, and decreased sports participation.  相似文献

The stability of CTG triplet repeats was analyzed in Escherichia coli to identify processes responsible for their genetic instability. Using a biochemical assay for stability, we show that the absence of single-stranded-DNA-binding protein leads to an increase in the frequency of large deletions within the triplet repeats.  相似文献
