Serum insulin-like growth factor (IGF)-I and IGF-binding protein-3 levels were measured in two groups of postmenopausal women with advanced breast cancer, who received the aromatase inhibitor letrozole 0.5 or 2.5 mg p.o. once daily. Blood samples were obtained from 15 patients in each dose group at baseline, and one and three months after starting therapy. Circulating IGF-I and IGFBP-3 concentrations were determined by means of radioimmunoassay. In both dosage groups a statistically significant increase in the IGF-I levels was observed during three months of letrozole treatment (P = 0.003). In addition, the multiple testing procedure yielded in the whole patient population a significant result in the comparison between mean IGF-I values after three months of therapy and those observed at baseline (P = 0.004), the estimated average increase being of 24%. No significant result was obtained in the analysis for the dose effect (P = 0.077) and for the time × dose interaction (P = 0.208). Circulating IGFBP-3 levels did not appear to be affected by letrozole treatment in either of the dose groups. This is the first report concerning the short-term effects of letrozole on components of the IGF system in breast cancer patients; further investigations are warranted in order to confirm these preliminary data.  相似文献

We have previously shown that clusters of guanine quadruplex (G4) structures can form in the human herpes simplex-1 (HSV-1) genome. Here we used immunofluorescence and immune-electron microscopy with a G4-specific monoclonal antibody to visualize G4 structures in HSV-1 infected cells. We found that G4 formation and localization within the cells was virus cycle dependent: viral G4s peaked at the time of viral DNA replication in the cell nucleus, moved to the nuclear membrane at the time of virus nuclear egress and were later found in HSV-1 immature virions released from the cell nucleus. Colocalization of G4s with ICP8, a viral DNA processing protein, was observed in viral replication compartments. G4s were lost upon treatment with DNAse and inhibitors of HSV-1 DNA replication. The notable increase in G4s upon HSV-1 infection suggests a key role of these structures in the HSV-1 biology and indicates new targets to control both the lytic and latent infection.  相似文献

Cell union in mating pairs in the ciliate Euplotes raikovi is controlled by a system of multiple mating types which are inherited with alleles codominant at the genetic locus mat and expressed via diffusible mating pheromones. The mating pheromones Er-2, Er-3, and Er-11 were purified from cells homozygous for the mat-2, mat-3, and mat-11 alleles, respectively. These pheromones are proteins of similar Mr (11,000-12,000) and acidity (pI 3.7-4.0) and are active at a concentration that varies from 2.9 X 10(-12) to 1.2 X 10(-11) M. Data on amino acid composition revealed that an unusually high amount of cysteine (12-15.7%) and poor contents of basic amino acids are common to every pheromone. On the basis of this uniformity in the main biochemical traits, which also holds for the previously purified pheromone Er-1, it was concluded that E. raikovi mating pheromones are members of a family of proteins structurally diversified from each other to varying extents.  相似文献

Summary The role and fate of male germ cells in planarian regeneration was studied in a population ofDugesia lugubris s.1. which provided a suitable karyological marker to distinguish diploid male germ cells from triploid embryonic and somatic cells. The nuclear Feulgen-DNA content in non-replicating triploid muscle cells of the pharynx and in non-replicating male gonia of testes from intact animals were measured by the cytophotometric technique. The pharynx was then removed by transection and each anterior regenerant was allowed to completely regenerate this organ. Measurements of the Feulgen-DNA content in muscle cells of the regenerated pharynx showed that most of these cells (95%) have a DNA content typical of triploid cells; however, some muscle cells (5%) with a nuclear DNA content typical of male gonia alone were observed.These results were interpreted in the following way. After transection, young male germ cells move from the testes to the wound where they participate in blastema formation along with reserve and/or somatic dedifferentiated cells. During regeneration some of these cells of male origin differentiate into pharyngeal muscle cells. Our findings are discussed in relation to the occurrence of mataplasia in planarians.  相似文献

Anacamptis pyramidalis is a variable and wide-spread European-Mediterranean taxon. Beside a dominant cytotype with 2n = 36 it includes cytotypes with 2n = 54 and 63 in northern Tuscany (and the Eastern Pyrenees) and one with 2n = 72 on Malta. In contrast,A. urvilleana, formerly often misidentified and included inA. pyramidalis, is a monomorphic and distinct species, endemic to the Maltese Islands. It has 2n = 36, can be clearly separated by morphological and anatomical features and is isolated from partly sympatric populations ofA. pyramidalis with 2n = 72 by differences in chromosome number, flowering time and habitat preference.  相似文献

A novel role of peroxin PEX6: suppression of aging defects in mitochondria   总被引:1,自引:0,他引:1  
Yeast cells become older with each division, but their daughters are born young. Mutational analysis shows that maintenance of this age asymmetry requires segregation of a complement of active mitochondria to daughters and that this process breaks down in older mother cells. This decline has implications for stem cell aging in higher organisms. PEX6, a peroxisome biogenesis gene, has been isolated as a multicopy suppressor of an atp2 age asymmetry mutant. Suppression depended on the presence of particular amino acid residues in Atp2p, and required adenosine triphosphate (ATP) binding and/or ATP hydrolysis activity of Pex6p. Extra copies of PEX6 corrected the deficit in Atp2p in mitochondria in the mutant by improving its import kinetics, resulting in near normal mitochondrial inheritance by daughter cells. The novel function of Pex6p described here may provide insights into peroxisomal and mitochondrial disorders and into metabolic diseases in general.  相似文献
