Haemophilus parainfluenzae isolates recovered from patients with respiratory diseases were studied for their ability to undergo genetic transformation by isogenic DNA. Two chromosomal markers, streptomycin resistance and nalidixic acid resistance, were tested for transformation efficiencies in H. parainfluenzae recipients from three biotypes. Most efficient in transformation was biotype II, followed by biotype I, while biotype III was nontransformable. Lack of transformation was not owing to poor donor activity of DNA, but to inability of the cells to develop competence. Strains that formed clumps in liquid media were nontransformable. Since the transformable biotype II is one of the prevalent biotypes world wide, one can speculate that DNA transformation probably plays a major role in the spread of drug resistance in H. parainfluenzae. Mixtures of ruminal bacteria degraded benzo(b)thien-4-yl methylcarbamate (Mobam) to 4-hydroxybenzothiophene, CO(2), and polar product(s). The metabolite, 4-hydroxybenzothiophene, was identified (after acetylation) by comparative infrared and mass spectrometry with an authentic sample. Carbon dioxide and polar product(s) were produced by degradation of the methylcarbamate moiety. Ten previously characterized strains of ruminal bacteria with diverse physiological capabilities did not degrade Mobam. However, three tributyrin-hydrolyzing strains were isolated that did degrade Mobam. Mobam inhibited growth of two of ten strains isolated on Mobam-free glycerol-tributyrin enrichment medium. One of these strains was also sensitive to 2-carbomethoxy-propene-2yl dimethyl phosphate (Phosdrin). Mobam prevented some ruminal bacteria from producing zones of hydrolysis in tributyrin emulsion media and inhibited some ruminal bacteria from degrading 1-naphthyl acetate and fluorescein-3',6'-diacetate.  相似文献

Succinic dehydrogenase (SDH) was localized in the mitochondria of Sarcoma 37 ascites tumor cells by the use of tetranitro-BT (TNBT) and nitro-BT (NBT) in smear preparations. Results with each tetrazolium salt as electron acceptor were evaluated with respect to: (a) size and shape of the formazan precipitate relative to standard mitochondrial morphology; (b) crystallization phenomena of reduced dye; (c) lipid adsorption of formazan. The association of formazan- or iron hematoxylin-stained mitochondria with lipid droplets within the cells was investigated, as was also the influence of formalin fixation, with and without cold acetone pretreatment, on mitochondrial morphology and enzymatic staining. Data from these studies appear to indicate that TNBT is more suitable than NBT for use as a cytochemical reagent in oxidative and/or dehydrogenase enzyme histochemistry and cytochemistry.  相似文献

Fungi are generally easily dispersed, able to colonise a wide variety of substrata and can tolerate diverse environmental conditions. However, despite these abilities, the diversity of fungi in the Atacama Desert is practically unknown. Most of the resident fungi in desert regions are ubiquitous. Some of them, however, seem to display specific adaptations that enable them to survive under the variety of extreme conditions of these regions, such as high temperature, low availability of water, osmotic stress, desiccation, low availability of nutrients, and exposure to high levels of UV radiation. For these reasons, fungal communities living in the Atacama Desert represent an unknown part of global fungal diversity and, consequently, may be source of new species that could be potential sources for new biotechnological products. In this review, we focus on the current knowledge of the diversity, ecology, adaptive strategies, and biotechnological potential of the fungi reported in the different ecosystems of the Atacama Desert.  相似文献

Insect–fungal interactions are an important but understudied aspect of tropical forest ecology. Here we present the first large‐scale study of insect communities feeding on the reproductive structures of macrofungi (basidiomes) in the Neotropics. This trophic interaction is not well characterized in most ecosystems; however, beetle consumption of basidiomes is thought to be affected by fungal factors, via mechanisms analogous to those observed in plant–herbivore interactions and in some interactions with fungi as hosts in the Holarctic region. We investigated how the composition of beetle assemblages varies as a function of fungal taxonomic distance, basidiome consistency, and hyphal systems. We collected 367 basidiomes belonging to the orders Polyporales and Hymenochaetales in the subtropical Araucaria angustifolia forest region of southern Brazil, along with any fauna present or without it. Basidiomes were maintained individually in the laboratory in plastic containers for up to three months to allow beetles to develop to adulthood, at which point the beetles were collected. We found that 207 basidiome specimens representing 40 species were associated with beetles. We recorded 447 occurrences of Coleoptera, representing 90 morphospecies from 20 families. We found that assemblages of fungivorous Coleoptera were more similar among more closely related fungi. Furthermore, the beetle assemblages varied as a function of basidiome toughness, which is influenced by sporocarp consistency and hyphal system type. The associations between beetles and basidiomes resemble those reported previously in temperate zones, suggesting continuity in the structure of such associations across a wide latitudinal range.  相似文献

In the last decades, an increasing fishing effort and a decreasing trend in fish catches have been observed in southern Brazil. Considering that marine mammals and fisheries usually compete for the same resources, it is reasonable to presume that the feeding ecology of these predators is affected by the current scenario. To evaluate this hypothesis, long-term variation in the diet of the South American sea lion (Otaria flavescens) relative to fisheries exploitation was analyzed for two periods (1993–2003 versus 2004–2014). The degree of overlap between the relative biomass of the sea lions’ diet and the target species of six types of local fishery was analyzed. An increase in prey overlap between sea lions and fisheries was observed in the more recent sampling period, along with an increase in prey diversity, richness, and niche breadth of the sea lions’ diet. These results suggest that the overfishing scenario could partly explain the modified feeding ecology of the sea lions. In this context, we recommend a review and better regulation of the current fishing effort in the region, which we believe will be an important step to maintain the fish stocks and minimize the impact of fishing on marine top predators.  相似文献
