Behavioural investigations into the transmission of bovine tuberculosis (Mycobacterium bovis) between badgers and cattle suggest that badger activity in farm buildings may incur a significant risk of cross-infection. However, measures to exclude badgers from buildings have not been systematically field-tested. In the present study, remote surveillance and radio-tracking were used to monitor the effect of electric fencing manipulations on the frequency of badger incursions into feed stores and cattle housing, and on badger ranging behaviour. Electric fencing was effective in preventing access to the farm buildings where it was installed and also significantly reduced incursions into unfenced buildings. Badger home range and core activity areas tended to increase in size when the fencing was installed, although they did not extend beyond the boundaries of the relevant social group territories. We discuss the logistical constraints of using electric fencing in this context and conclude that it is a potentially useful method of reducing contact between badgers and cattle, within farm buildings and yards.  相似文献

Poorly known species may be cryptically endangered, especially when they inhabit fragmented and threatened habitats. Heart‐tongued frogs (genus Phyllodytes, family Hylidae, Lophyohylinae) comprise 17 species of poorly known frogs that have obligatory associations with tank bromeliads. The distributions of all species are restricted to a small, extremely fragmented, region of Atlantic Forest in eastern Brazil. We model climate and tank bromeliad distributions to better understand frog distribution limits. Using records from several sources for frogs and bromeliads with climate data from WorldClim, we modelled the distribution of Phyllodytes using maximum entropy. We compared climate and altitude within the distribution and nearby to test how climate may limit distribution. Climate together with bromeliad distributions provided the best model and predicted the smallest suitable area for Phyllodytes that was larger than that occupied, from the state of Paraíba in the north to Rio Grande do Sul in the south. Phyllodytes occurs in lower elevations that are warmer, wetter and less variable than the surrounding regions where it does not occur, and dispersal is apparently limited by the surrounding, inhospitable, region. Dispersal limitation and habitat fragmentation have relegated Phyllodytes to many very small habitat fragments. With many species in this genus being known from a single or few samples, this unfortunate combination of limitation and fragmentation suggests that some or all species of Phyllodytes may be threatened with extinction, especially if habitat fragmentation continues at its present pace in eastern Brazil.  相似文献

Phospholipase D (PLD) genes are members of a superfamily that is defined by several highly conserved motifs. PLD in mammals has been proposed to play a role in membrane vesicular trafficking and signal transduction. Using site-directed mutagenesis, 25 point mutants have been made in human PLD1 (hPLD1) and characterized. We find that a motif (HxKxxxxD) and a serine/threonine conserved in all members of the PLD superfamily are critical for PLD biochemical activity, suggesting a possible catalytic mechanism. Functional analysis of catalytically inactive point mutants for yeast PLD demonstrates that the meiotic phenotype ensuing from PLD deficiency in yeast derives from a loss of enzymatic activity. Finally, mutation of an HxKxxxxD motif found in a vaccinia viral protein expressed in the Golgi complex results in loss of efficient vaccinia virus cell-to-cell spreading, implicating the viral protein as a member of the superfamily and suggesting that it encodes a lipid modifying or binding activity. The results suggest that vaccinia virus and hPLD1 may act through analogous mechanisms to effect viral cellular egress and vesicular trafficking, respectively.  相似文献

We investigated song development in the pre‐independent zebra finch (aged 15–35 d), a period when neural pathways for song learning and production are forming and social influences outside the family are limited. Expt 1 investigated the onset and the minimum duration of tutoring needed for song learning in fledglings. We found most begin to learn song from 25 d of age and need about 10 d contact with the father tutor to make accurate copies. This onset corresponds with major developments in the formation of the neural circuitry implicated in song acquisition. Subsong also begins on day 25 suggesting that the sensory and motor phases of song learning fully overlap in the zebra finch. Our findings support the hypothesis that the song circuitry is fully functional by 35 d of age and the sensitive phase for zebra finches extends therefore from about days 25–65. However, only the first 10 d of this period are necessary to learn a tutor's song with fair accuracy. Expt 2 investigated the role of the paternal bond, spatial proximity and mating status in a fledgling's choice of song tutor where the father was the sole parent. Young chose the father over single unrelated males (expt 2a) or unrelated males in company with their female partners (expt 2b). Given the close spatial proximity of both potential tutors to the fledglings it is probably the filial bond, established via paternal care that is the cause of this preference. Zebra finches sing the same song phrase in two contrasting contexts: female‐directed song during pre‐copulatory courtship and undirected song where no female or display is involved. In expt 3 we tested the song learning preference of pre‐independent young for two categories of non‐paternal tutors: those singing predominantly female‐directed song and those singing exclusively undirected song. There was a small, but significant, preference for fledgling zebra finches to copy songs from males that sang female‐directed song. This preference is consistent with the hypothesis that young males not only learn the acoustic features of their tutor's song but also the visual and dynamic movements that constitute the courtship display.  相似文献

The vaccinia virus (VV) A33R gene encodes a highly conserved 23- to 28-kDa glycoprotein that is specifically incorporated into the viral outer envelope. The protein is expressed early and late after infection, consistent with putative early and late promoter sequences. To determine the role of the protein, two inducible A33R mutants were constructed, one with the late promoter and one with the early and late A33R promoter elements. Decreased A33R expression was associated with small plaques that formed comets in liquid medium. Using both an antibiotic resistance gene and a color marker, an A33R deletion mutant, vA33Δ, was isolated, indicating that the A33R gene is not essential for VV replication. The plaques formed by vA33Δ, however, were tiny, indicating that the A33R protein is necessary for efficient cell-to-cell spread. Rescue of the large-plaque phenotype was achieved by inserting a new copy of the A33R gene into the thymidine kinase locus, confirming the specific genetic basis of the phenotype. Although there was a reduction in intracellular virus formed in cells infected with vA33Δ, the amount of infectious virus in the medium was increased. The virus particles in the medium had the buoyant density of extracellular enveloped viruses (EEV). Additionally, amounts of vA33Δ cell-associated extracellular enveloped viruses (CEV) were found to be normal. Immunogold electron microscopy of cells infected with vA33Δ demonstrated the presence of the expected F13L and B5R proteins in wrapping membranes and EEV; however, fully wrapped vA33Δ intracellular enveloped viruses (IEV) were rare compared to partially wrapped particles. Specialized actin tails that propel IEV particles to the periphery and virus-tipped microvilli (both common in wild-type-infected cells) were absent in cells infected with vA33Δ. This is the first deletion mutant in a VV envelope gene that produces at least normal amounts of fully infectious EEV and CEV and yet has a small-plaque phenotype. These data support a new model for VV spread, emphasizing the importance of virus-tipped actin tails.  相似文献

This paper studies dissection propagation subject to internal pressure in a residually-stressed two-layer arterial model. The artery is assumed to be infinitely long, and the resultant plane strain problem is solved using the extended finite element method. The arterial layers are modelled using the anisotropic hyperelastic Holzapfel–Gasser–Ogden model, and the tissue damage due to tear propagation is described using a linear cohesive traction–separation law. Residual stress in the arterial wall is determined by an opening angle \(\alpha \) in a stress-free configuration. An initial tear is introduced within the artery which is subject to internal pressure. Quasi-static solutions are computed to determine the critical value of the pressure, at which the dissection starts to propagate. Our model shows that the dissection tends to propagate radially outwards. Interestingly, the critical pressure is higher for both very short and very long tears. The simulations also reveal that the inner wall buckles for longer tears, which is supported by clinical CT scans. In all simulated cases, the critical pressure is found to increase with the opening angle. In other words, residual stress acts to protect the artery against tear propagation. The effect of residual stress is more prominent when a tear is of intermediate length (\(\simeq \)90\(^\circ \) arc length). There is an intricate balance between tear length, wall buckling, fibre orientation, and residual stress that determines the tear propagation.  相似文献
