Inoculation with Bradyrhizobium sp. strain 127E14 has been shown to cause a dramatic increase in the internode length of lima bean (Phaseolus lunatus L.), when compared to control plants inoculated with strain 127E15. This rhizobial-induced growth also occurs in cowpea (Vigna unguiculata [L.] Walp), an alternate host for the symbiont. Cowpea plants inoculated with strain 127E14 were 23% taller than those inoculated with strain 127E14 after 6 weeks of growth. Petiole length was found to be significantly greater in plants inoculated with strain 127E14. Cowpea plants treated at the apex with exogenous GA3 or GA4/7 responded by increasing internode length when compared to controls. As in lima beans, the rhizobial-induced growth response observed in cowpeas may be in response to an imbalance in the levels of GA-like substances within the plants. Gibberellins A1, A3, A8, A19, A20, A29, and A44have been identified by GC-MS analysis in stems of cowpea, whereas the gibberellins A1, A19, A20, A29, and A44 were found to be present in nodule tissue formed by strain 127E14. The presence of these GAs indicates that the early 13-hydroxylation biosynthetic pathway is operative in cowpea. GAs identified in cowpea nodules are similar to those found in lima bean nodules formed by the same rhizobia. The finding that rhizobial strain 127E14 induces GA-dependent growth responses in two host legumes further supports the hypothesis that the presence of this bacteria alters the GA balance within the plant.  相似文献

A Gibberellin-Deficient Brassica Mutant-rosette   总被引:1,自引:3,他引:1  
A single-gene mutant (rosette [ros/ros]) in which shoot growth and development are inhibited was identified from a rapid cycling line of Brassica rapa (syn campestris). Relative to normal plants, the mutant germinated slowly, had delayed or incomplete floral development, and reduced leaf, petiole, and internode growth. The exogenous application of GA3 by foliar spray or directly to the shoot tip of rosette resulted in rapid flowering, bolting (shoot elongation), and viable seed production. Shoots of rosette contained endogenous levels of total gibberellin (GA)-like substances (`Tan-ginbozu' dwarf rice assay) of about one-tenth of that of the normal rapid-cycling line of B. rapa which consisted almost entirely of a very nonpolar, GA-like substance which yielded GA1 and GA3 upon mild acid hydrolysis. In a normal rapid-cycling B. rapa line, the nonpolar putative GA1 and GA3 conjugates were present, but additionally, free GA1 and GA3 were abundant and identified by gas chromatography-mass spectrometry-selected ion monitoring. The quantities of free GA1 and GA3 in the normal line and in rosette were quantified by GC-MS-SIM using [2H2]GA1 as an internal standard. Fourteen-day-old rosette and normal seedlings contained 5.3 and 23.2 ng GA1 per plant, respectively. At day 21 the rosette plants contained 7.7 and 26.1 nanograms per plant of GA1 and GA3, while normal plants contained 31.1 and 251.5 nanograms per plant, respectively. Thus, normal plants contained from four to ten times higher levels of total GA-like substances, GA1, or GA3, than rosette. The ros allele results in reduced GA level, yielding the rosette phenotype whose delayed germination and flowering, and reduced shoot growth responses indicate a probable role for endogenous GA1 and GA3 in the regulation of these processes in Brassica.  相似文献

Clostridium perfringens is the causative agent of a number of human diseases, such as gas gangrene and food poisoning, and many diseases of animals. Recently significant advances have been made in the development of C. perfringens genetics. Studies on bacteriocin plasmids and conjugative R plasmids have led to the cloning and analysis of many C. perfringens genes and the construction of shuttle plasmids. The relationship of antibiotic resistance genes to similar genes from other bacteria has been elucidated. A detailed physical map of the C. perfringens chromosome has been prepared, and numerous genes have been located on that map. Reproducible transformation methods for the introduction of plasmids into C. perfringens have been developed, and several genes coding for the production of extracellular toxins and enzymes have been cloned. Now that it is possible to freely move genetic information back and forth between C. perfringens and Escherichia coli, it will be possible to apply modern molecular methods to studies on the pathogenesis of C. perfringens infections.  相似文献

Complex Regional Pain Syndrome type one (CRPS I) or formerly Reflex Sympathetic Dystrophy (RSD) is a disabling syndrome, in which a painful limb is accompanied by varying symptoms. Neuropathic pain is a prominent feature of CRPS I, and is often refractory to treatment. Since gabapentin is an anticonvulsant with a proven analgesic effect in various neuropathic pain syndromes, we sought to study the efficacy of the anticonvulsant gabapentin as treatment for pain in patients with CRPS I.  相似文献

Background/Aims

Arthropod-borne pathogens are transmitted into a unique intradermal microenvironment that includes the saliva of their vectors. Immunomodulatory factors in the saliva can enhance infectivity; however, in some cases the immune response that develops to saliva from prior uninfected bites can inhibit infectivity. Most rodent reservoirs of Yersinia pestis experience fleabites regularly, but the effect this has on the dynamics of flea-borne transmission of plague has never been investigated. We examined the innate and acquired immune response of mice to bites of Xenopsylla cheopis and its effects on Y. pestis transmission and disease progression in both naïve mice and mice chronically exposed to flea bites.

Methods/Principal Findings

The immune response of C57BL/6 mice to uninfected flea bites was characterized by flow cytometry, histology, and antibody detection methods. In naïve mice, flea bites induced mild inflammation with limited recruitment of neutrophils and macrophages to the bite site. Infectivity and host response in naïve mice exposed to flea bites followed immediately by intradermal injection of Y. pestis did not differ from that of mice infected with Y. pestis without prior flea feeding. With prolonged exposure, an IgG1 antibody response primarily directed to the predominant component of flea saliva, a family of 36–45 kDa phosphatase-like proteins, occurred in both laboratory mice and wild rats naturally exposed to X. cheopis, but a hypersensitivity response never developed. The incidence and progression of terminal plague following challenge by infective blocked fleas were equivalent in naïve mice and mice sensitized to flea saliva by repeated exposure to flea bites over a 10-week period.

Conclusions

Unlike what is observed with many other blood-feeding arthropods, the murine immune response to X. cheopis saliva is mild and continued exposure to flea bites leads more to tolerance than to hypersensitivity. The immune response to flea saliva had no detectable effect on Y. pestis transmission or plague pathogenesis in mice.  相似文献

These studies represent the first systematic survey of the incidence of conjugative antibiotic resistance in Clostridium perfringens. Ninety-two antibiotic-resistant porcine strains were examined to see if they could donate their antibiotic-resistance determinants to sensitive recipient strains. Fifteen of the 89 tetracycline-resistant strains transferred their tetracycline resistance in mixed-plate mating experiments but no transfer of macrolide-lincosamide resistance was detected. The efficiencies of transfer of tetracycline resistance varied from 1.3 X 10(-3) to 1.9 X 10(-6) transconjugants per donor cell. Significantly higher transfer efficiencies were observed when both the donor and recipient strains were derivatives of strain CW 362. These values ranged from 3.7 X 10(-1) to 4.6 X 10(-2) transconjugants per donor cell. This high frequency transfer system should prove invaluable for further genetic studies on this microorganism.  相似文献

The correlation between gibberellin (GA) metabolism and growth rate was investigated using two Sorghum bicolor inbred lines, Hegari and AT×623, and their heterotic F1 hybrid. Previous studies have demonstrated that this hybrid is taller and has substantially greater shoot dry weights and leaf areas than either parental inbred. [3H]GA20 was applied to the leaf whorl of seedlings and after 24 hours, plants were harvested and separated into roots, shoot cylinders containing the apical meristems, and leaf blades. Chromatographic analyses of metabolites indicated the conversions of [3H]GA20 to [3H]GA1,8 and 29. The conversion of [2H]GA20 to [2H]GA1 was demonstrated by gas chromatography-selected ion monitoring (GC-SIM). Putative glucosyl conjugates of all of the [3H]GAs were also produced and GA8 was identified by GC-SIM following enzymic cleavage of the putative [3H]GA8 glucosyl conjugate fraction. Comparing the genotypes, [3H]GA20 metabolism was more rapid in the shoot cylinders of the hybrid than in the shoot cylinders from inbreds. In the hybrid samples, there was a three-fold increase in the putative conjugate(s) of [3H]GA1 which was the principal metabolite, and increased production of [3H]GA8 and the putative conjugates of [3H]GA29 and [3H]GA8. Conversely, levels of the remaining precursor, [3H]GA20, and its putative conjugate(s) were reduced in the hybrid. The rate of GA20 metabolism was thus positively correlated with growth rate across these sorghum genotypes. This correlation supports a promotive role of GA in the regulation of shoot growth and in the expression of heterosis (hybrid vigor) in sorghum.  相似文献

Under controlled environment and/or field conditions, vegetative growth (height, internode length, leaf area, shoot dry weight, grain yield) was greater in an F1 maize hybrid than in either parental inbred. Endogenous gibberellin (GA)-like substances in apical meristem cylinders were also higher in the hybrid than in either inbred, both on a per plant and per gram dry weight basis. There were no apparent qualitative differences in GA-like substances, however. Levels of GA-like substances in all genotypes were highest prior to tassel initiation. Chromatographic comparisons of the GA-like substances and authentic standards of GA native to maize on gradient-eluted SiO2 partition and reverse-phase C18 high-pressure liquid chromatography columns are described. No consistent differences in abscisic acid levels of the three genotypes were observed. This correlation of heterosis for endogenous GA-like substances with heterosis for growth suggests that amounts of endogenous GA may be related to hybrid vigor in maize.  相似文献
