Dichelobacter nodosus is the essential causative agent of footrot in sheep. The major D. nodosus-encoded virulence factors that have been implicated in the disease are type IV fimbriae and extracellular proteases. To examine the role of the fimbriae in virulence, allelic exchange was used to insertionally inactivate the fimA gene, which encodes the fimbrial subunit protein, from the virulent type G D. nodosus strain VCS1703A. Detailed analysis of two independently derived fimA mutants revealed that they no longer produced the fimbrial subunit protein or intact fimbriae and did not exhibit twitching motility. In addition, these mutants were no longer capable of undergoing natural transformation and did not secrete wild-type levels of extracellular proteases. These effects were not due to polar effects on the downstream fimB gene because insertionally inactivated fimB mutants were not defective in any of these phenotypic tests. Virulence testing of the mutants in a sheep pen trial conducted under controlled environmental conditions showed that the fimA mutants were avirulent, providing evidence that the fimA gene is an essential D. nodosus virulence gene. These studies represent the first time that molecular genetics has been used to determine the role of virulence genes in this slow growing anaerobic bacterium. 相似文献

The genera Cimex Linnaeus and Oeciacus Stål (Heteroptera: Cimicidae) are common haematophagous ectoparasites of bats or birds in the Holarctic region. Both their phylogenetic relationship and the systematics of the entire family previously were based on data from morphology and host relationships. Relationships among nine species of the genus Cimex and three species of the genus Oeciacus were analysed here using two mitochondrial and three nuclear genes. Cimex was shown to be paraphyletic with respect to Oeciacus. Oeciacus is thus proposed as a synonym of Cimex. The characteristic phenotype of Oeciacus results from the specific host association with different species of swallows (Hirundinidae). The morphological characters that have been used as diagnostic for the genera were shown to be valid and can be further used for determination at species level. The present analyses recovered the four traditional morphologically defined species groups of the genus Cimex. However, their relationships were poorly resolved – only the C. hemipterus group showed a well‐supported relationship to the C. pipistrelli group. The molecular differentiation within the Palaearctic C. pipistrelli and the Nearctic C. pilosellus species groups correlates with their karyotype differentiation. Furthermore, the analyses suggest poly‐ or paraphyly of the former genus Oeciacus. Either way this indicates there is a large amount of host‐associated phenotypic convergence in either bat‐ or bird‐associated groups of species. The probability of host choice and subsequent switch in Cimicidae are discussed and possible scenarios of the evolution of host association in species of Cimex are suggested. 相似文献

If future HIV vaccine design strategies are to succeed, improved understanding of the mechanisms underlying protection from infection or immune control over HIV replication remains essential. Increased cytotoxic capacity of HIV-specific CD8+ T-cells associated with efficient elimination of HIV-infected CD4+ T-cell targets has been shown to distinguish long-term nonprogressors (LTNP), patients with durable control over HIV replication, from those experiencing progressive disease. Here, measurements of granzyme B target cell activity and HIV-1-infected CD4+ T-cell elimination were applied for the first time to identify antiviral activities in recipients of a replication incompetent adenovirus serotype 5 (Ad5) HIV-1 recombinant vaccine and were compared with HIV-negative individuals and chronically infected patients, including a group of LTNP. We observed readily detectable HIV-specific CD8+ T-cell recall cytotoxic responses in vaccinees at a median of 331 days following the last immunization. The magnitude of these responses was not related to the number of vaccinations, nor did it correlate with the percentages of cytokine-secreting T-cells determined by ICS assays. Although the recall cytotoxic capacity of the CD8+ T-cells of the vaccinee group was significantly less than that of LTNP and overlapped with that of progressors, we observed significantly higher cytotoxic responses in vaccine recipients carrying the HLA class I alleles B*27, B*57 or B*58, which have been associated with immune control over HIV replication in chronic infection. These findings suggest protective HLA class I alleles might lead to better outcomes in both chronic infection and following immunization due to more efficient priming of HIV-specific CD8+ T-cell cytotoxic responses. 相似文献

Endogenous gibberellins (GAs) were extracted from safflower (Carthamus tinctorius L.) stems and detected by capillary gas chromatography-mass spectrometry from which GA1, GA3, GA19,, GA20, GA29, and probably, GA44 were detected. The detection of these GAs suggests that the early 13-OH biosynthetic pathway is prevalent in safflower shoots. Deuterated GAs were used as internal standards and GA concentrations were determined in stems harvested at weekly intervals. GA1 and GA19 levels per stem increased but concentrations per gram dry weight decreased over time. GA20 was only detected in young stem tissue.Gibberellic acid (GA3) was also applied in field trials and both GA3 and the GA biosynthetic inhibitor, paclobutrazol, were applied in growth chamber tests. GA3 increased epidermal cell size, internode length, and increased internode cell number causing stem elongation. Conversely, paclobutrazol reduced stem height, internode and cell size, cell number and overall shoot weight. In field tests, GA3 increased total stem weight, but decreased leaf weight, flower bud number and seed yield. Thus, GA3 promoted vegetative growth at the expense of reproductive commitment. These studies collectively indicate a promotory role of GAs in the control of shoot growth in safflower, and are generally consistent with gibberellin studies of related crop plants.

An analysis of the genetic traits of human minor histocompatibility (mH) antigens is, unlike with inbred mice, rather complicated. Moreover, the fact that mH antigens are recognized in the context of MHC molecules creates an additional complication for reliable segregation analysis. To gain insight into the mode of inheritance of the mH antigens, we relied upon a series of HLA-A2-restricted cytotoxic T-cell (CTL) clones specific for four mH antigens. To perform segregation analysis independent of HLA-A2 gene: we transfected HLA-A2-negative cells with the HLA-A2 gene: this results in the cell surface expression of the HLA-A2 gene product and, if present, mH antigen recognition. The mode of inheritance of the HLA-A2-restricted mH antigens HA-1, -2, -4, and -5 was analysed in 25 families whoese members either naturally expressed positive. Analysis of distribution of the mH antigens in the parent population among the mating types, together with their inheritance patterns in the families, demonstrated that the four mH antigens behaved as Mendelian traits, whereby each can be considered a product of a gene with two alleles, one expressing and one not expressing the detected specificity. We also showed that the loci encoding the HA-1 and HA-2 antigens are not closely linked to HLa (lod scores Z (0 = 0.05) <–4.0). Some indication was obtained that the HA-4- and HA-5-encoding loci may be losely linked to HLA. While we are aware of the limited results of this nonetheless comprehensive study, we feel the similarity in immunogenetic traits between human and mouse mH antigens is at least striking. 相似文献

A newly defined leukocyte antigen system, the secondary B-cell (SB) system, was shown to be linked to HLA. The SB marker has been investigated in lymphocyte donors presumed to be genetically homozygous for HLA-A through HLA-D/DR by virtue of descent from a first-cousin marriage and of phenotypic homozygosity for these HLA markers. Of 19 donors, 3 were found to be heterozygous for SB. Studies of the families of these three donors could not distinguish with certainty whether the heterozygosity resulted from SB/DR recombination or from “pseudohomozygosity” for HLA-A through -D/DR by inheritance of two genetically unrelated but similar haplotypes. However, our data favored the occurrence of SB/DR recombination with a meiotic distance perhaps as large as 3.3 cMorgan. Recombinations were identified which mapped SB between HLA-B and GL01. These studies demonstrate the usefulness of cells from first-cousin marriage offspring in mapping a polymorphic genetic system. 相似文献

Greenhouse-grown oilseed rape (Brassica napus, annual Canola variety `Westar') plants were harvested at six dates from the vegetative phase until the early pod (silique)-fill/late flowering stage. Endogenous gibberellin (GA)-like substances were extracted from stems, purified, and chromatographed on silica gel partition columns prior to bioassay in serial dilution using the `Tan-ginbozu' dwarf rice microdrop assay. The concentrations of total endogenous GA-like substances were low during vegetative stages (1 nanogram GA3 equivalents/gram dry weight), and rose 300-fold by the time of floral initiation. After floral initiation the concentration of GA-like substances fell, then rose again during bolting to maximal levels during the early pod-fill stage (940 nanograms per gram dry weight). The qualitative profiles of GA-like substances varied across harvests, with higher proportions of a GA1-like substance at the early pod-fill stage. In a second study stems were similarly harvested at eight dates and the concentrations of endogenous GA1, the principal bioactive native GA of oilseed rape, were determined by gas chromatography-selected ion monitoring using [17,17-2H]GA1 as a quantitative internal standard. The concentration of GA1 increased at about the time of floral initiation and then subsequently fell, thus confirming the pattern noted above for total GA-like substances. The exogenous application of paclobutrazol (PP333), a persistent triazole plant growth regulator (PGR) which blocks GA biosynthesis, or another triazole, triapenthenol (RSW0411), prevented flowering as well as bolting; plants remained at the vegetative rosette stage. These results imply a causal role for endogenous GA, in the control of bolting, which normally precedes anthesis. Further, the rise in the concentration of total endogenous GA-like substances, including GA1, which was associated with floral initiation, and the prevention of visable floral development by the triazole PGRs, also indicates a role for endogenous GAs in the regulation of flowering in B. napus. 相似文献

Individuals of known HLA phenotype were used to examine the specificity of secondary proliferative (restimulation) responses in lymphocyte populations which had been primed previously in long-term, one-way mixed lymphocyte reactions (MLR). The objective was to identify the genetic region responsible for this secondary mitogenic effect. Restimulation responses between unrelated and related individuals from a family with a recombinant betweenHLA-B andD produced similar results. In both cases responses were grouped into zero, intermediate, and positive. Restimulation was by three types of cell: those with HLA-D types compatible with the responder; those with HLA-D types specific to the primary stimulator; and those with HLA-D types of a third-party cell. Compatible responses fell into the zero category and the intermediate category, third-party responses fell into the intermediate category, and specific responses fell into the positive category. Our conclusion was that HLA-D products evoked secondary mitogenic responses, while structures coded for by the HLA-A and B regions appeared to have no effect. 相似文献
