We conducted field and semi-field experiments to determine the most visually stimulating and acceptable version of a branch-mimicking trap (PVC cylinder topped with an inverted screen funnel) to capture adult plum curculios, Conotrachelus nenuphar (Herbst) (Coleoptera: Curculionidae) foraging within canopies of host trees. Cylinder traps (25 cm tall × 6 cm diam) coated with different colors of gloss or flat latex paint captured similar numbers of feral adults in field tests. Tall cylinder traps (50 cm tall × 6 cm diam) captured significantly more feral adults in field tests in 1999 than cylinder traps of lesser height and/or different diameter, and significantly more released adults arrived at traps with these dimensions compared to cylinder traps of lesser height in semi-field tests. In field and semi-field tests, respectively, significantly more adults were captured by and numerically more adults arrived at tall cylinder traps (50 cm tall × 6 cm diam) attached in an upright, standard position to a horizontal limb than attached at angles of 45°, 90°, 135°, or 180° to horizontal limbs.  相似文献

OBJECTIVE: To demonstrate the utility of fine needle aspiration biopsy (FNAB) in radiofrequency ablation (RFA) of suspected metastatic tumors at various sites. STUDY DESIGN: Eighteen patients referred for RFA underwent 21 aspirations prior to the procedure. A radiologist performed the FNAB and RFA with radiographic guidance. On-site preliminary evaluation of Diff-Quik-stained smears were followed with Papanicolaou staining. A final diagnosis was rendered and compared to the preliminary diagnosis. RESULTS: Liver was aspirated in 17 cases, lung in 3 cases and pubic bone in 1. Fifteen aspirates were deemed on site as positive or suspicious for malignancy. A preliminary, on-site diagnosis of benign was given in one case and adequate with deferment for review of all slides in four others. One FNAB was unsatisfactory. All but one (patient with benign diagnosis) then immediately proceeded to RFA of the lesion. After review of additional slides, the final diagnosis confirmed metastatic adenocarcinoma in 16, hepatocellular carcinoma in 2 and metastatic squamous cell carcinoma in 1. One FNAB yielded benign hepatocytes, and one was unsatisfactory. CONCLUSION: FNAB is an accurate, safe and rapid method of confirming disease in patients just prior to undergoing RFA.  相似文献

Hydrophobic bile acids are toxic to isolated rat hepatocytes by mechanisms involving mitochondrial dysfunction and oxidative stress. In the current study we examined the role of nitric oxide (NO), a potential mediator of apoptosis, during bile acid-induced apoptosis. Freshly isolated rat hepatocytes and hepatic mitochondria generated NO and peroxynitrite (ONOO(-)) in a concentration- and time-dependent manner when exposed to the toxic bile salt glycochenodeoxycholate (GCDC) (25-500 microm), which was prevented by the nitric-oxide synthase (NOS) inhibitors N(G)-monomethyl-N-arginine monoacetate (l-NMMA) and 1400W. Relationships between hepatocyte NO production and apoptosis were examined by comparing the effects of NOS inhibitors with other inhibitors of GCDC-induced apoptosis. Inhibitors of caspases 8 and 9, the mitochondrial permeability transition blocker cyclosporin A, and the antioxidant idebenone reduced NO generation and apoptosis in GCDC-treated hepatocytes. In contrast, NOS inhibitors had no effect on GCDC-induced apoptosis despite marked reduction of NO and ONOO(-). However, treatment with the NO donors S-nitroso-N-acetylpenicillamine and spermine NONOate [N-(-aminoethyl)N-(2-hydroxy-2-nitrohydrazino)-1,2-ethylenediamine) inhibited apoptosis and caspase 3 activity while significantly elevating NO levels above GCDC-stimulated levels. Neither NO donors nor NOS inhibitors affected GCDC-induced mitochondrial permeability transition or cytochrome c release from liver mitochondria or GCDC-induced mitochondrial depolarization from isolated hepatocytes, suggesting that NO inhibits bile acid-induced hepatocyte apoptosis by a non-mitochondrial-dependent pathway. In conclusion, whereas NO produced from GCDC-treated hepatocytes neither mediates nor protects against bile acid-induced apoptosis, higher levels of NO inhibit GCDC-induced hepatocyte apoptosis by caspase-dependent pathways.  相似文献

Herpes simplex virus type 1 utilizes cell surface heparan sulfate as receptors to infect target cells. The unique heparan sulfate saccharide sequence offers the binding site for viral envelope proteins and plays critical roles in assisting viral infections. A specific 3-O-sulfated heparan sulfate is known to facilitate the entry of herpes simplex virus 1 into cells. The 3-O-sulfated heparan sulfate is generated by the heparan sulfate d-glucosaminyl-3-O-sulfotransferase isoform 3 (3-OST-3), and it provides binding sites for viral glycoprotein D (gD). Here, we report the purification and structural characterization of an oligosaccharide that binds to gD. The isolated gD-binding site is an octasaccharide, and has a binding affinity to gD around 18 microm, as determined by affinity coelectrophoresis. The octasaccharide was prepared and purified from a heparan sulfate oligosaccharide library that was modified by purified 3-OST-3 enzyme. The molecular mass of the isolated octasaccharide was determined using both nanoelectrospray ionization mass spectrometry and matrix-assisted laser desorption/ionization mass spectrometry. The results from the sequence analysis suggest that the structure of the octasaccharide is a heptasulfated octasaccharide. The proposed structure of the octasaccharide is DeltaUA-GlcNS-IdoUA2S-GlcNAc-UA2S-GlcNS-IdoUA2S-GlcNH(2)3S6S. Given that the binding of 3-O-sulfated heparan sulfate to gD can mediate viral entry, our results provide structural information about heparan sulfate-assisted viral entry.  相似文献

The African crowned eagle (Stepahnoaetus coronatus) is the primary predator for arboreal primates throughout sub-Saharan forests. Monkeys typically respond with alarm calls when they are aware of the presence of crowned eagles and such calls can be considered a corollary of predation risk within primate groups. Alarm calls from six species of monkeys were recorded across the home range of an eagle pair in Taï National Park, Côte d''Ivoire. Spatial and temporal variation in primate alarm calling was found to be related to eagle ranging behaviour according to the predictions of central-place foraging models. Radio-tracking data indicate that eagle activity is higher in the centre of their home range and monkey alarm-calling rates are correspondingly elevated near eagle nests as opposed to farther away. Alarm-calling rates are also temporally coupled with measures of eagle activity. There were considerable differences between the species in both rates and spatial patterns of alarm calling. The variation we measure in predation risk is expected to have consequences at the behavioural and population level.  相似文献

The transport and attachment behaviors of Spumella guttula (Kent), a nanoflagellate (protist) found in contaminated and uncontaminated aquifer sediments in Cape Cod, Mass., were assessed in flowthrough and static columns and in a field injection-and-recovery transport experiment involving an array of multilevel samplers. Transport of S. guttula harvested from low-nutrient (10 mg of dissolved organic carbon per liter), slightly acidic, granular (porous) growth media was compared to earlier observations involving nanoflagellates grown in a traditional high-nutrient liquid broth. In contrast to the highly retarded (retardation factor of approximately 3) subsurface transport previously reported for S. guttula, the peak concentration of porous-medium-grown S. guttula traveled concomitantly with that of a conservative (bromide) tracer. About one-third of the porous-medium-grown nanoflagellates added to the aquifer were transported at least 2.8 m downgradient, compared to only approximately 2% of the broth-grown nanoflagellates. Flowthrough column studies revealed that a vital (hydroethidine [HE]) staining procedure resulted in considerably less attachment (more transport) of S. guttula in aquifer sediments than did a staining-and-fixation procedure involving 4',6'-diamidino-2-phenylindole (DAPI) and glutaraldehyde. The calculated collision efficiency (approximately 10(-2) for porous-medium-grown, DAPI-stained nanoflagellates) was comparable to that observed earlier for the indigenous community of unattached groundwater bacteria that serve as prey. The attachment of HE-labeled S. guttula onto aquifer sediment grains was independent of pH (over the range from pH 3 to 9) suggesting a primary attachment mechanism that may be fundamentally different from that of their prey bacteria, which exhibit sharp decreases in fractional attachment with increasing pH. The high degree of mobility of S. guttula in the aquifer sediments has important ecological implications for the protistan community within the temporally changing plume of organic contaminants in the Cape Cod aquifer.  相似文献

Nutritional regulatory responses of the generalist caterpillar Spodoptera littoralis (Boisduval) were explored, in choice and no-choice experiments, using the Geometric Framework. In the choice experiment, newly moulted final instar larvae were provided with one of three protein-biased foods (PB-food: p35:c7, p28:c5.6 or p21:c4.2) and one of three equal protein-carbohydrate ratio foods (ER-food: p21:c21, p16.8:c16.8 or p12.6:c12.6). On five of the nine treatments, caterpillars independently regulated protein and carbohydrate intake to a mixture of 57 and 43%, respectively. However, when the concentration of the ER-food decreased and that of the paired PB-food increased, caterpillars progressively abandoned regulation and ate more of the PB-food. Despite these regulatory differences, performance (survivorship, growth and development) was similar across all nine treatments. In the no-choice experiment, caterpillars were given one of five foods (p35:c7, p28:c14, p21:c21, p14:c28 and p7:c35). Results indicated that caterpillars moved to a point in protein-carbohydrate space that was consistent with the Equal Distance Rule of compromise, a pattern previously predicted for generalist feeders. The insects on the two extreme foods, p35:c7 and p7:c35, showed reduced pupal mass and longer development, respectively. There was also strong evidence for post-ingestive regulation of nutrient utilisation, notably for protein.  相似文献
