Roots, tubers, and bananas (RTB) are vital staples for food security in the world's poorest nations. A major constraint to current RTB breeding programmes is limited knowledge on the available diversity due to lack of efficient germplasm characterization and structure. In recent years large‐scale efforts have begun to elucidate the genetic and phenotypic diversity of germplasm collections and populations and, yet, biochemical measurements have often been overlooked despite metabolite composition being directly associated with agronomic and consumer traits. Here we present a compound database and concentration range for metabolites detected in the major RTB crops: banana (Musa spp.), cassava (Manihot esculenta), potato (Solanum tuberosum), sweet potato (Ipomoea batatas), and yam (Dioscorea spp.), following metabolomics‐based diversity screening of global collections held within the CGIAR institutes. The dataset including 711 chemical features provides a valuable resource regarding the comparative biochemical composition of each RTB crop and highlights the potential diversity available for incorporation into crop improvement programmes. Particularly, the tropical crops cassava, sweet potato and banana displayed more complex compositional metabolite profiles with representations of up to 22 chemical classes (unknowns excluded) than that of potato, for which only metabolites from 10 chemical classes were detected. Additionally, over 20% of biochemical signatures remained unidentified for every crop analyzed. Integration of metabolomics with the on‐going genomic and phenotypic studies will enhance ’omics‐wide associations of molecular signatures with agronomic and consumer traits via easily quantifiable biochemical markers to aid gene discovery and functional characterization.  相似文献

Several micro-scale chromatography-based procedures for purification of the β-galactosidase from the yeast Kluyveromyces lactis were assayed. Purified enzyme was suitable to be used as antigen to induce polyclonal antibodies production. Specific staining of non-denaturing PAGE gels with chromogenic substrates allowed the determination of the number of subunits forming the native enzyme.  相似文献

In this work, phytosterol-biotransforming strains were selected from Mycobacterium sp., using a high concentration of beta-sitosterol. The selection was made by culturing the strains in a medium enriched with 14 g beta-sitosterol/l as the unique source of carbon. During 2 months, the bacterial cultures were transferred successively. The extraction of the biotransformation products was made with methanol and ethyl acetate. The qualitative and quantitative analysis was made by means of thin-layer chromatography, gas-liquid chromatography (GLC) and GLC-mass spectrometry. Under these conditions, it was observed that after seven transfers, the strains MYcobacterium sp. MB-3683 and the Mycobacterium fortuitum B-11045 increased their biotransformation capacity from 20% to 64% and from 34% to 55%, respectively. The products in the highest proportion identified for each trial were androstenedione and androstadienedione. The results suggest that the high substrate concentration could be a selective mechanism to obtain strains more efficient in the biotransformation of beta-sitosterol into steroidal bases.  相似文献

A case of pure squamous carcinoma of the breast diagnosed by needle aspiration cytology is presented. The three conditions required for that diagnosis were fulfilled: (1) an absence of elements other than malignant epidermoid cells, (2) the independence of the lesion from adjacent dermal structures, and (3) the exclusion of other squamous carcinomas in the patient. Teleost fin rays are able to regenerate, when they are cut, restoring the whole structure in a few weeks. Following the formation and growth of an apical blastema, deposition of lepidotrichial matrix occurs. We have histo and immunochemically analyzed the maturation process of the lepidotrichial hemisegment, pointing out the interactions between their components and likewise the temporal and spatial distribution of some extracellular matrix components during regeneration. Lepidotrichial matrix is rich in sulfated glycosaminoglycans (GAGs), most of which are forming proteoglycans. Collagen is abundant and it strongly interacts with GAGs, as the tissue differentiates. The use of specific digestions with papain and collagenase suggests that some mannose rich glycoproteins may be also implicated in lepidotrichial maturation before mineralization. In each hemisegment a central band (CB) can be observed. In spite of the histochemical similarities between the CB and the subepidermical basement membrane, neither collagen IV nor laminin are present. This CB could be the result of a transient transdifferentiation of the outer lepidotrichial synthesizing cells.  相似文献

In this study we investigated the capacity of Andean alder (Alnus acuminate Kunth), inoculated withFrankia and two ectomycorrhizal fungi (Alpova austroalnicola Dominguez andAlpova diplophloeus ([Zeller and Dodge] Trappe and Smith), for nodulation and growth in pots of a soilless medium that contained vermiculite or a mixture of ground basalt rock and vermiculite. The seedlings were inoculated withFrankia suspensions prepared from root nodules ofA. Acuminate, followed by inoculation with spores of either one of the twoAlpova species. The seedlings were grown in a greenhouse for 12 months. The seedlings grown in the vermiculite-based growth medium containing large (1-3 mm) basalt particles andAlpova austroalnicola or medium-sized (0.5-1 mm) basalt particles andA. Diplophloeus had the heaviest shoot and root nodule dry weights and abundant ectomycorrhizal colonization. Ectomycorrhizas formed byA. Acuminate withAlpova austroalnicola is described here for the first time. Growth ofAlnus acuminate inoculated with ectomycorrhizal fungi andFrankia in the soilless primary minerals indicates that Andean alder can alter resource supply by tapping an otherwise unavailable nutrient source.  相似文献

The gross development of the trout inner ear between embryonic and juvenile stages was studied by light microscopy. The otocyst has already formed in 3–4 mm embryos. The semicircular canals begin to separate from the utriculo-saccular cavity in 6 mm embryos, the anterior canal first, then the posterior and the horizontal canal later. The formation of the saccular cavity begins in 7 mm embryos, whereas that of the lagena occurs in 18 mm fry. The first macular primordia appear before the separation of cavities. The anterior and horizontal crests arise from the primordium of the utricular macula, and the posterior crest, macula lagena, and macula neglecta arise from that of the saccular macula. The macula lagena and macula neglecta appear later. The sensory areas of the labyrinth and the number of receptor cells grow continuously between the embryonic and juvenile stages. © 1993 Wiley-Liss, Inc.  相似文献

Abstract: Pigment epithelium-derived factor (PEDF), purified from human fetal retinal pigment epithelium cell culture medium, was shown to potentiate the differentiation of human Y-79 retinoblastoma cells. To investigate potential neurotrophic effects of PEDF on neurons other than those of retinal derivation, we used cultures of cerebellar granule cells. The number of cerebellar granule cells was significantly larger in the presence of PEDF, as demonstrated by an assay for viable cells that uses 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2 H -tetrazolium, inner salt, conversion, by cell count, and by immunocytochemistry. The effect of PEDF showed a dose-response relationship, with a larger effect in chemically defined medium than in serum-containing medium [ED50 = 30 ng/ml (0.70 n M ) in chemically defined medium and 100 ng/ml (2.3 n M ) in serum-containing medium]. PEDF had no effect on incorporation of bromodeoxyuridine (cell proliferation) or on neurofilament content (neurite outgrowth) measured by an enzyme-linked immunoadsorbent assay. These results demonstrate that PEDF has a neurotrophic survival effect on cerebellar granule cells in culture and suggest the possibility that it may affect other CNS neurons as well.  相似文献

Background  

Archaeosomes (ARC), vesicles prepared from total polar lipids (TPL) extracted from selected genera and species from the Archaea domain, elicit both antibody and cell-mediated immunity to the entrapped antigen, as well as efficient cross priming of exogenous antigens, evoking a profound memory response. Screening for unexplored Archaea genus as new sources of adjuvancy, here we report the presence of two new Halorubrum tebenquichense strains isolated from grey crystals (GC) and black mood (BM) strata from a littoral Argentinean Patagonia salt flat. Cytotoxicity, intracellular transit and immune response induced by two subcutaneous (sc) administrations (days 0 and 21) with BSA entrapped in ARC made of TPL either form BM (ARC-BM) and from GC (ARC-GC) at 2% w/w (BSA/lipids), to C3H/HeN mice (25 μg BSA, 1.3 mg of archaeal lipids per mouse) and boosted on day 180 with 25 μg of bare BSA, were determined.  相似文献
