On Wistar rats in view of electrophysiological parameters after sleep deprivation (SD; awake by gentle handling method) and the subsequent postdeprivative sleep (PDS) immunohistochemical investigation of Bcl-2 and p53 peptides optical density levels in neurons of paraventricular (PVN), supraoptic (SON) and median (MnPN) hypothalamus nuclei was carried out. The Bcl-2 was increased in all nuclei both after SD and PDS. The level of p53 was increased in PVN and SON after SD and PDS, but in MnPN only on PDS. Any morphological attributes of apoptosis in the nuclei was not revealed. Obtained data testify an active role of p53 and Bcl-2 peptides in regulation of neuronal activity in hypothalamus at change of a cycle wakefulness-sleep.  相似文献

The identification and studying the molecular bases of functioning of new cytotoxic agents finds an important implication in developing drugs for fighting with tumors. While investigating the cytotoxic action of protein complex Tag7-Hsp70 which was opened in our laboratory previously we found that Tag7-Hsp70 demonstrated the same specificity in regard to different tumor target cells as it was for classical cytokine TNF-α. L-929 cells and Jurkat cells appeared to be good targets representing up to 30% of dead cells within a population and HeLa cells-bad targets representing less than 5% of dead cells after 20 h of incubation with either of the cytotoxic agents. While investigating the action of either TNF-α or Tag7-Hsp70 on L-929 cells we detected two peaks of death: after 3 h and after 20 h. For both cytotoxic agents we observed the first, smaller (13–15%), peak to be eliminated after the addition of caspase inhibitor YVAD-CHO and the second, greater (25–30%), peak to become even bigger in presence of caspase inhibitor. Probably, protein complex Tag7-Hsp70 interacts like TNF-α with a receptor on the surface of tumor cells that results in triggering two alternative mechanisms of programmed cell death: apoptosis and necroptosis.  相似文献

Chemical syntheses of 2'-O-(allyloxycarbonyl)methyladenosine, 2'-O-(methoxycarbonyl)methyladenosine and 2'-O-(2,3-dibenzoyloxy)propyluridine 3'-2-cyanoethyl-N,N-diisopropyl phosphoramidite building blocks are described. These monomers were used successfully to incorporate carboxylic acid, 1,2-diol and aldehyde functionalities into synthetic oligonucleotides.  相似文献

All DNA (cytosine-5)-methyltransferases contain a single conserved cysteine. It has been proposed that this cysteine initiates catalysis by attacking the C6 of cytosine and thereby activating the normally inert C5 position. We show here that substitutions of this cysteine in the E. coli methylase M. EcoRII with either serine or tryptophan results in a complete loss of ability to transfer methyl groups to DNA. Interestingly, mutants with either serine or glycine substitution bind tightly to substrate DNA. These mutants resemble the wild-type enzyme in that their binding to substrate is not eliminated by the presence of non-specific DNA in the reaction, it is sensitive to methylation status of the substrate and is stimulated by an analog of the methyl donor. Hence the conserved cysteine is not essential for the specific stable binding of the enzyme to its substrate. However, substitution of the cysteine with the bulkier tryptophan does reduce DNA binding. We also report here a novel procedure for the synthesis of DNA containing 5-fluorocytosine. Further, we show that a DNA substrate for M. EcoRII in which the target cytosine is replaced by 5-fluorocytosine is a mechanism-based inhibitor of the enzyme and that it forms an irreversible complex with the enzyme. As expected, this modified substrate does not form irreversible complexes with the mutants.  相似文献

Conditions for efficient regeneration in mutant strains of the doxorubicin-producing organism Str. peucetius var. caesius were developed. The effect of the protoplast regeneration on changes in the proportion of the components of the anthracycline complex produced by these strains was shown. Variants with doxorubicin productivity 2 times higher than that of the parent strain were isolated.  相似文献

The plasmid pSS120, determining the synthesis of species specific I phase antigen of Shigella sonnei is mobilized for genetic transfer into E. coli K12 recipient cells with the frequency 12-41%. The frequency depends on the type of mobilized plasmid and recipient strain. The I phase antigen is normally expressed in II phase recipient cells and in E. coli cells. During mobilization pSS120 forms cointegrates representing a recombinant of mobilizing and mobilized plasmids DNA. The study of pSS120 inheritance stability has shown the plasmid to be unstable during culturing of bacteria and to be partially lost from the parent Shigella sonnei strains as well as from the "hybrid" transconjugants obtained. The 60 Md plasmid present in the donor strains of Shigella sonnei is prone to structural fragmentation particularly expressed in Shigella sonnei/E. coli hybrids.  相似文献

In experiments with rats a study was made of a number of factors influencing the resorption of 241Am from the gastrointestinal tract (GIT). The resorption of 241Am from GIT was found to be 120-245 times more intensive in neonatal rats, during the first 21 days after birth (a milk diet), than in adult animals. A milk diet for adult rats produced a 5-fold increase in the resorption of 241Am from GIT. The additional administration of digestive enzymes, as a homogenate from pancreas and small intestine, produced a 7--9-fold increase in the rate of 241Am resorption from GIT.  相似文献

A comparative analysis of the temperature effect on the thrombin- and trypsin-catalyzed hydrolysis of synthetic substrates which are derivatives of A(alpha)-chain of fibrinogen has been carried out. The substrates have general formula: Tos-P2-Arg-OCH3 and Tos-P3-P2-Arg-OCH3, where P2-Gly, Val, MeVal; P3 = Gly, Sar. The activation parameters delta G not equal to, delta H not equal to and delta S not equal to are determined. Thrombin is shown to split the most effectively tripeptide containing amino acids sequence Gly-Gly at the subsites of P3 and P2. It is suggested to be caused by an ability of the above compound to take a bent conformation at the active site of thrombin.  相似文献

Changes of the total number of leucocytes and the leucocytic formula of peripheral blood were studied and stability of development was evaluated by the indicator of fluctuating asymmetry of populations of green frogs Rana lessonae Cam. and Rana ridibunda Pall. inhabiting city reservoirs of Nizhni Novgorod and the Nizhegorodskaya Province. The revealed changes in the amphibians white blood depending on the degree of anthropogenic effects in the studied reservoirs can be divided into two types. In the first type (the level of stability of development of populations is 2 scores according to Zakharov) there were revealed neutropenia, increased content of eosinophils, and total leucopenia. In the second type, a more intensive anthropogenic effect and deviation of the organism state from normal conditions, estimated by 3 scores (Zakharov), the leucograms of frogs were characterized by more pronounced negative alterations. Leucocytosis accompanied by neutropenia, eosinophilia, basophilia, and a decrease of the total number of lymphocytes has been established. In the absence of deviations of the stability of development from the conditionally normal state, there was revealed the balance of leucogram indicators, close to norm. An increase of the number of lymphocytes and young forms of neutrophils in the amphibian peripheral blood indicates stimulation of the processes providing the organism protective reaction. Cross-correlation between the integral indicator of stability of development, the content of eosinophils and lymphocytes has been established in individuals from populations of green frogs inhabiting the studied reservoirs.  相似文献
