The mass exchange characteristics of 50 m3 industrial fermentors with aerodynamic foam suppression and the effect of the specific power input on biosynthesis of penicillin were studied. A change in the specific power input from 1.3 to 1.9 kW/m3 had no effect on the level of the antibiotic accumulation when the medium with 8 per cent of lactose was used. An increase in the aeration rate from 1 to 1.2 m3/m3 X min provided a 1.1-fold increase in the penicillin activity of the fermentation broth. The use of the device for aerodynamic foam suppression with a system of automatic control of the partial pressure of dissolved carbon dioxide allowed decreasing 1.22-fold the oil use and increasing the process productivity by 10 per cent.  相似文献

Agouti-related peptide is expressed in the hypothalamic neurons in humans and animals. Immunohistochemical studies in Wistar rats shows significant changes in the optical density of agouti-related peptide in the neurons of the arcuate hypothalamic nucleus, as well as in their processes in the hypothalamus and nucleus accumbens after 6 h of sleep deprivation (an increase) and after 2 h of post-deprivative sleep (a decrease). Comparison of these findings with the earlier results shows the opposite trends in the changes in the optical density of agouti-related peptide and the speed of the limiting enzyme of dopamine synthesis, tyrosine hydroxylase, in the hypothalamus and in the striatonigral system. An increase in the agouti-related peptide level was accompanied by a decrease in tyrosine hydroxylase, while a decrease in agouti-related peptide, on the contrary, was accompanied by an increase in the tyrosine hydroxylase activity. Our data show the role played by agouti-related peptide as a modulator of the functional activity of the dopaminergic brain neurons. The interrelation between various functions of the body, such as food behavior, sleep, and stress, is considered to be mediated by the participation of the same neurotransmitter systems in their regulation.  相似文献

Controlled burning has been carried out to stimulate moderate ground fire in the Scots pine (Pinus sуlvestris L.) stand (age class I) in the Krasnoyarsk forest steppe at the beginning of the growing season. The structure of annual rings and trophic supply, as well as the cambial zone antioxidant system condition and the content of photosynthetic pigments in the needles of dominant and suppressed trees in the experimental and control plots, were studied 3 h, 5 days, and 2.5 months after the fire. The fire narrowed the annual ring width, since the number and size of early and late tracheids decreased. The primary reaction to pyrogenic heat shock (sharp reduction of chlorophyll content in needles and manifestation of oxidative stress in the stem cambial zone) was an elevated level of hydrogen peroxide and activity of most antioxidant enzymes and a reduction in the nonstructural carbohydrates pool. Restoration of the cambium function after the fire was promoted by an increased activity of amylase, invertase, and peroxidase.  相似文献

The ability of water hyacinth (Eichhornia crassipes) to uptake Ag, Ba, Cd, Mo, and Pb from waters in gold mine tailing area was studied. All experiments were carried out in the field conditions without using of model system. Bioconcentration (BCF) and translocation factors (TF) as well as elements accumulation by plant in different points of tailings-impacted area were evaluated. It has been shown that water hyacinth demonstrates high ability to accumulate Mo, Pb, and Ba with BCF values 24,360 ± 3600, 18,800 ± 2800 and 10,040 ± 1400, respectively and is efficient in translocation of Mo and Cd. The general trend of the plant accumulation ability in relation to the studied elements corresponds to their concentration in the medium. As the distance from tailings increases, concentration of Ag, Ba and Pb in plant decreases more clearly than that of Cd, while the amount of Mo accumulated by plant doesn't drop significantly in accordance with its concentration in water. Under the conditions of the confluence of river Ur and drainage stream Ba and Ag can be considered as potential candidates for phytomining.  相似文献

Exposure to high temperature or other stresses induces a synthesis of heat shock proteins. Many of these proteins are molecular chaperones and some of them help cells to cope with heat-induced denaturation and aggregation of other proteins. In the last decade, chaperones have received increased attention in connection with their role in maintenance and propagation of the Saccharomyces cerevisiae prions, infectious or heritable agents transmitted at the protein level. Recent data suggest that functioning of the chaperones in reactivation of heat-damaged proteins and in propagation of prions is based on the same molecular mechanisms but may lead to different consequences depending on the type of aggregate. In both cases the concerted and balanced action of “chaperones'' team,” including Hsp104, Hsp70, Hsp40 and possibly other proteins, determines whether a misfolded protein is to be incorporated into an aggregate, rescued to the native state or targeted for degradation.Key Words: Amyloid, Hsp40, Hsp70, Hsp104, stress response, yeast  相似文献

In yeast, functions of the endoplasmic reticulum (ER) depend on the Golgi apparatus Ca2+ pool, which is replenished by the medial-Golgi ion pump Pmr1p. Here, to dissect the role of the Golgi Ca2+ pool in protein folding and elimination of unfolded proteins in the ER, the manifestations of the pmr1 mutation in yeast Hansenula polymorpha were studied. The PMR1 gene was disrupted in a H. polymorpha diploid strain. Haploid segregants of this diploid bearing the disruption allele were viable, though they showed a severe growth defect on synthetic medium and rapidly died during storage at low temperature. Disruption of H. polymorpha PMR1 led to defects of the Golgi-hosted protein glycosylation and vacuolar protein sorting. This mutation increased the survival rate of H. polymorpha cells upon treatment with the proapoptotic drug amiodarone. Unlike Saccharomyces cerevisiae, the H. polymorpha pmr1 mutant was not hypersensitive to chemicals that induce the accumulation of unfolded proteins in the ER, indicating that the elimination of unfolded proteins from the ER was not essentially affected. At the same time, the pmr1 mutation improved the secretion of human urokinase and decreased its intracellular aggregation, indicating an influence of the mutation on the protein folding in the ER.  相似文献
