The en face organization of human aortic endothelium in zones of low (LP) and high (HP) probability of sudanophilia was examined in preparation impregnated with silver nitrate. It is found that the heterogeneity of endothelial cells (EC) by area is "random" or "clusterized". In the latter case the major part of small and medium-sized EC (less than 800 microns 2) is associated in distinct groups ("clusters") and form foci with high monolayer density. The luminal surface outside the clusters was formed by preferentially large and giant EC. Clusterized endothelium was found with statistically significant higher frequency in HP zones of both "normal" and "atherosclerotic" vessels. The maximum clusterization of EC was revealed on the shoulder region and on the periphery of atherosclerotic plaques which was speculated to be a growth zone of the lesion. It is suggested that the appearance of clusterized endothelium is associated with the active development of atherosclerosis. 相似文献

The arrangement of proteins into complexes is a key organizational principle for many cellular functions. Although the topology of many complexes has been systematically analyzed in isolation, their molecular sociology in situ remains elusive. Here, we show that crude cellular extracts of a eukaryotic thermophile, Chaetomium thermophilum, retain basic principles of cellular organization. Using a structural proteomics approach, we simultaneously characterized the abundance, interactions, and structure of a third of the C. thermophilum proteome within these extracts. We identified 27 distinct protein communities that include 108 interconnected complexes, which dynamically associate with each other and functionally benefit from being in close proximity in the cell. Furthermore, we investigated the structure of fatty acid synthase within these extracts by cryoEM and this revealed multiple, flexible states of the enzyme in adaptation to its association with other complexes, thus exemplifying the need for in situ studies. As the components of the captured protein communities are known—at both the protein and complex levels—this study constitutes another step forward toward a molecular understanding of subcellular organization. 相似文献

A general method is developed for a numerical analysis of the frequency spectra of internal, internal-surface, and surface slow waves in a waveguide with transverse plasma density variations. For waveguides with a piecewise constant plasma filling, the spectra of slow waves are thoroughly examined in the limits of an infinitely weak and an infinitely strong external magnetic field. For a smooth plasma density profile, the frequency spectrum of long-wavelength surface waves remains unchanged, but a slow damping rate appears that is caused by the conversion of the surface waves into internal plasma waves at the plasma resonance point. As for short-wavelength internal waves, they are strongly damped by this effect. It is pointed out that, for annular plasma geometry, which is of interest from the experimental point of view, the spectrum of the surface waves depends weakly on the magnetic field strength in the waveguide. 相似文献

A tobacco clone TPD1 (Tobacco Pollen Development), characterized by an extended flowering period, was obtained using a serial agrobacterial transformation of tobacco (Nicotiana tabacum L., cv. Samsun) by constructing a DNA carrying the kanamycin resistance gene inserted into the binary pBin19 vector. However, the characteristics of the vegetative growth of these plants were similar to those of other tobacco clones and the wild type. In the insertion mutant, pollen was 1.5 times smaller than in the wild type and germinated on the stigmata of neither its own clone nor the wild-type plants. Cytochemical investigation of pollen of the TPD1-mutant did not reveal any activity of respiratory enzymes, succinate dehydrogenase and peroxidase, indicating that the pollen was nonviable. Unlike the wild type, theTPD1 mutant exhibited disturbed trophic interactions within the anther tissues, particularly suppressed starch hydrolysis in the anther wall tissues at the stage of rapidly growing microspores. We conclude that the insertion of T-DNA into the TPD1 gene produced structural and metabolic changes during the development of anther tissues in the mutant clone, resulting in pollen sterility. 相似文献

The role of p38α stress-kinase in the regulation of the premature senescence program induced by the histone deacetylase inhibitor sodium butyrate (NaB) was studied in rodent transformed cell lines. The study was carried out on E1A+cHa-ras transformants obtained from mouse embryonic fibroblasts null for the Mapk14 gene encoding p38α stress-kinase (the mERasp38?/? cell line), or for the PPM1D gene encoding the Wip1 phosphatase (the mERas Wip1?/? cell line), whose absence led to constitutive activation of p38α kinase. It was found that after NaB treatment both cell lines completely stopped proliferation due to irreversible G1/S cell cycle arrest. In both lines a marker of senescence appeared—the activity of β-galactosidase (SA-β-Gal). As well, treatment of the cells with NaB for several days led to morphological cell changes, such as partial readjustment of the actin cytoskeleton, spreading on the substrate, and heterochromatin focus formation (SAHF) in the senescent cell nuclei. These data allow us to suggest that, in the absence of functionally active p38α kinase, the NaB-induced irreversible process of cellular senescence may occur via alternative pathways for downregulation of the cell cycle. 相似文献

A study was made of the content of alpha-fetoprotein (alpha-FP) in the blood serum and of proliferative processes in hepatoma 22A-bearing mice after injection of cyclophosphamide in a dose of 150 mg/kg at varying time of 4-day tumor growth. The data obtained indicate that the time of appearance of alpha-FP in the blood, the pattern of rhythmic fluctuations in protein content and in the number of DNA-synthesizing and mitotic cells of the tumor depend on the time of the cytostatic injection during the day. Besides, cyclophosphamide exerts a more powerful inhibitory effect on the content of alpha-FP in the blood serum and less powerful on the proliferative processes in hepatoma 22A. 相似文献

The dynamics of 3H-thymidine labeled mitosis and diurnal rhythm of proliferative activity was studied. The isotope was injected to BALB/C mice at the peak of diurnal rhythm of DNA synthesis activity of basal layer cells of oesophageus epithelium. It has been established that the increase in the mitotic index during 24 hours depends on the increase in number of cells being in S-period. The data show that the increase of mitotic index at diurnal rhythm occurs at the expense of 75% of new G0-cells which entered into the mitotic cycle, and of 25% of re-entering cells that had divided during the maximal mitotic activity a day before. It is found that the duration of mitotic cycle of cell population which entered into the mitotic cycle synchronously is almost equal to the period of diurnal rhythm of mitotic activity, i.e. 24 hours. 相似文献
