The arrangement of proteins into complexes is a key organizational principle for many cellular functions. Although the topology of many complexes has been systematically analyzed in isolation, their molecular sociology in situ remains elusive. Here, we show that crude cellular extracts of a eukaryotic thermophile, Chaetomium thermophilum, retain basic principles of cellular organization. Using a structural proteomics approach, we simultaneously characterized the abundance, interactions, and structure of a third of the C. thermophilum proteome within these extracts. We identified 27 distinct protein communities that include 108 interconnected complexes, which dynamically associate with each other and functionally benefit from being in close proximity in the cell. Furthermore, we investigated the structure of fatty acid synthase within these extracts by cryoEM and this revealed multiple, flexible states of the enzyme in adaptation to its association with other complexes, thus exemplifying the need for in situ studies. As the components of the captured protein communities are known—at both the protein and complex levels—this study constitutes another step forward toward a molecular understanding of subcellular organization.  相似文献

The ability of animals to perform a complex motor task representing a model of compensatory discrete tracing was investigated in dogs and cats. Elongation of a light spot on an oscilloscope screen into a vertical line served as a signal of motor reaction. The reaction consisted in several movements, each of which reduced the initial signal change by 1/2, 1/3, 1/4 or 1/5 of its value. Alimentary reinforcement was given only after a reduction of the signal to its initial level. Signals with a different number of steps of transition to their initial state were presented at random. It was found that learned animals were capable of a stable behaviour of a compensatory discrete tracing type. A hypothesis has been advanced about a probabilistic participation of visual control in the realization of tracing motor behaviour.  相似文献

It has been shown that the dielectric properties of sweat of different zones differ. High therapeutic efficacy of aminoglycosides, quinolones, cephalosporins and rifampicins was demonstrated in experiments performed on monkeys, infected aerogenically by Yersinia pestis 1300. Antibacterials inhibited Y. pestis cells reproduction in the infected animals organisms evaluated by dynamics of bacterial cells isolated from the blood and fauces of the animals. It was shown that antibacterial therapy prevented infection transmission from the infected animals. The time of respiratory tract sanitation was in the range from 12 to 48 hours after the treatment and depended on drug efficacy.  相似文献

Human umbilical vein endothelial cells (EC) were grown on elastic silicone membranes subjected to cyclic stretch, simulating arterial wall motion. Stretching conditions (20% amplitude, 52 cycle/min) stimulated stress fiber formation and their orientation transversely to the strain direction. Cell bodies aligned along the same axis after the actin cytoskeleton. EC orientation response was inhibited by the adenylate cyclase activator, forskolin (10(-5) M), which caused stress fiber disassembly and the redistribution of F-actin to the cortical cytoplasm. Preoriented EC depleted of stress fibers by forskolin treatment retained their aligned state. Thus, stress fibers are essential for the process of EC orientation induced by repeated strain, but not for the maintenance of EC orientation. The monolayer formed by EC grown to confluence in conditions of intermittent strain consisted of uniform elongated cells and was resistant to deformation. In contrast, the monolayer assembled in stationary conditions was less compliant and exposed local denudations on initiation of stretching. When stretched in the presence of 10(-5) M forskolin it rapidly (3-4 h) reestablished integrity but gained a heterogeneous appearance since denuded areas were covered by giant cells. The protective effect of forskolin was because of the stimulation of EC spreading. This feature of forskolin was demonstrated while studying its action on EC spreading and repair of a scratched EC monolayer in conventional culture. Thus mechanical deformation and adenylate cyclase activity may be important factors in the control of endothelium morphology in human arteries.  相似文献

Summary The darkening of lupin plants grown in a sand culture on a nitrogen-free medium at a stage of initial flowering led to a sharply decreased nitrogen fixation intensity which eventually ceased. Decreased intensity of nitrogen fixation in bacteroids was accompanied by an accumulation of poly--hydroxybutyric acid (PHB); in the course of 10–20 h (depending upon temperature) its content increased by 2.5–3.0 times. If, following darkening, the plants were once again exposed to light, an abrupt increase of nitrogen fixation intensity was observed and a simultaneous decrease of PHB content.It has been shown that lupin's exposure to light in14CO2 atmosphere lasting 19 h resulted in the latter's incorporation into PHB, bacteroids and into the entire nodule; these processes developed almost in parallel.During the early period of vegetation growth prior to flowering, the PHB content of bacteroids decreased from 13–14 to 3–4% of dry weight, whereas the intensity of nitrogen fixation was raised. Concurrently increase of the activity of some enzymes connected with the PHB metabolism (acetoacetyl-CoA-reductase, acetyl-CoA-acetyl transferase, PHB-depolymerase, CoA-transferase of 3-ketoacids) occured. The plants' subsequent ageing and reduction of nitrogen fixation intensity led to a noticeable increase of PHB content and a decrease of the above mentioned enzymes' activity. The specific activity of \-hydroxybutyric dehydrogenase involved with PHB catabolism was high and was maintained at a constant level throughout the entire vegetative period.  相似文献
