Signal-dependent sorting of proteins in the early secretory pathway is required for dynamic retention of endoplasmic reticulum (ER) and Golgi components. In this study, we identify the Erv41–Erv46 complex as a new retrograde receptor for retrieval of non–HDEL-bearing ER resident proteins. In cells lacking Erv41–Erv46 function, the ER enzyme glucosidase I (Gls1) was mislocalized and degraded in the vacuole. Biochemical experiments demonstrated that the luminal domain of Gls1 bound to the Erv41–Erv46 complex in a pH-dependent manner. Moreover, in vivo disturbance of the pH gradient across membranes by bafilomycin A1 treatment caused Gls1 mislocalization. Whole cell proteomic analyses of deletion strains using stable isotope labeling by amino acids in culture identified other ER resident proteins that depended on the Erv41–Erv46 complex for efficient localization. Our results support a model in which pH-dependent receptor binding of specific cargo by the Erv41–Erv46 complex in Golgi compartments identifies escaped ER resident proteins for retrieval to the ER in coat protein complex I–formed transport carriers.  相似文献

In the preseason soccer training, morning and afternoon training sessions often are scheduled daily. The high frequency of training sessions could place heavy strain on biological systems, and it is necessary to apply proper recovery strategies for improving the players' capability to regain an adequate working state for subsequent training units. However, the effect of recovery interventions following soccer training units is debatable, due to a lack of studies performed in field situations. The aim of this study was to examine, during a 21-day preseason soccer training, the most effective recovery intervention (i.e., passive, dry-aerobic exercises, water-aerobic exercises, electrostimulation) on anaerobic performances (i.e., squat jump, countermovement jump, bounce jumping, and 10-m sprint) and subjective ratings (i.e., perceived exertion and muscle pain), with the conditions before the intervention controlled and standardized. Twelve young (age: 18.1 +/- 1.2 years) elite soccer players participated. Data were collected on 4 occasions 2 days apart and at the same time of the day. Activity and dietary intake were replicated on each occasion. After baseline measurements, participants performed a standardized training during which their heart rates and ratings of perceived exertion were recorded. This was followed by a 20-minute recovery intervention. After a 5-hour rest, athletes' ratings of muscle pain were recorded prior to a second test session. There were no significant differences in exercise intensities and baseline anaerobic performances. Significantly (p < 0.01) better performances were observed in the afternoon. Although no main effect of recovery intervention was observed on anaerobic performances, dry-aerobic exercises (0.6 +/- 0.9) and electrostimulation (0.6 +/- 1.2) were more beneficial (p < 0.01) than water-aerobic exercises (2.1 +/- 1.1) and passive rest (2.1 +/- 1.7) for reducing muscle pain, which could affect the player's working ability.  相似文献

Background  

The first step in biofilm formation is bacterial attachment to solid surfaces, which is dependent on the cell surface physico-chemical properties. Cell wall anchored proteins (CWAP) are among the known adhesins that confer the adhesive properties to pathogenic Gram-positive bacteria. To investigate the role of CWAP of non-pathogen Gram-positive bacteria in the initial steps of biofilm formation, we evaluated the physico-chemical properties and adhesion to solid surfaces of Lactococcus lactis. To be able to grow in milk this dairy bacterium expresses a cell wall anchored proteinase PrtP for breakdown of milk caseins.  相似文献

Species translocations are increasingly being used as a management tool to mitigate population losses due to such factors as habitat degradation and fragmentation, but post‐introduction follow‐up is relatively sparse. Post‐translocation telemetry can assess success by identifying activity, emigrations, survival, habitat usage, and reproductive events, aiding in the continued management of translocated populations and informing future efforts. This study assessed movement of translocated adult lake sturgeon (Acipenser fulvescens) immediately post‐release and a decade later, and tested for associations between environmental variables and spawning movements. Prior to their translocation in 2002, 13 of 51 adult lake sturgeon were surgically implanted with radio telemetry tags and tracked for 1 year. In 2011 and 2013, eight additional adults were captured within the reintroduction site and implanted with radio‐tags. Six of the 13 sturgeon tagged in 2002 dispersed downstream over a dam during the early post‐release period. In spring 2014, tagged adults were tracked to the spillway at the release area's inflow, and spawning was confirmed by larval captures. Movement data for tagged adults differed between the two tracking periods, showing marked differences in behaviour over time. Water velocity was correlated with upstream and downstream spawning movements, with water temperature also correlated with downstream movement. Research regarding post‐translocation movement and dispersal provides insight on behavioural responses following translocation, and may improve outcomes by informing future efforts.  相似文献
