Changes in the nitric oxide (NO) system of the rat cerebral cortex were investigated by immunohistochemistry, immunoblotting, and NO synthase (NOS) activity assays in adult rats submitted for 30 min to hypoxia, in a hypobaric chamber at a simulated altitude of 38,000 ft (11000 m) (154.9 mm Hg). The cerebral cortex was studied after different survival times, 0 and 24 h, 5, 8, 15, and 30 days of reoxygenation. This situation led to morphological alterations in the large type I interneurons, as well as immunoreactive changes in the appearance and number of the small neurons (type II), both containing neuronal NOS (nNOS). Some of these small neurons showed immunoreactive cytoplasm and short processes; others, the more numerous during all reoxygenation periods, contained the immunoreactive product mainly related to a perinuclear ring. Ultrastructurally, these small neurons exhibited changes in nuclear structures as in the shape of the nuclear membrane, in the distribution of heterochromatin, and in the nucleolar morphology. The reaction product for nitrotyrosine, as a marker of protein nitration, showed modifications in distribution of the immunoreactive product. No expression was found for inducible NOS (iNOS). All these modifications were accompanied by increased nNOS and nitrotyrosine production as demonstrated by Western blotting and calcium-dependent activity, returning to control conditions after 30 days of reoxygenation, suggesting a reversible NO mechanism of action.  相似文献

The objective of the present investigation was to verify ifAnabaena spiroides exopolysaccharides sustains growth and reproductionof Ceriodaphnia cornuta. The animals were raised in controlledlaboratory conditions and fed exclusively on A. spiroides exopolysaccharides,which were obtained from filtered stationary growth phase A.spiroides cultures. A molecular weight cut-off of 12–14kDa was used to separate the exopolysaccharides from low molecularweight materials. The lyophilized exopolysaccharides were dissolvedin filtered and autoclaved reservoir water at a concentrationof 15 mg L–1 (0.22 ± 0.01 mg C animal–1day–1).These results were compared with animals fed on seston at naturalconcentration obtained by filtering (68 µm) Barra BonicaReservoir water (0.23 ± 0.01 mg C animal–1day–1).Several life history parameters (duration of embryonic development,generation time, reproductive performance, age-specific length,primipara age and length) were obtained during 15-day experiments.Growth rate (GR) and weight-specific secondary production werealso quantified. Exopolysaccharide-fed organisms had bettergrowth and reproductive performance (0.091 ± 0.004 GR;51 ± 4.59 eggs produced per female) than seston-fed individuals(0.078 ± 0.010 GR; 30.90 ± 7.25 eggs producedper female). These results show that such macromolecules area potential food source, fulfilling nutritional requirementsto sustain growth and reproduction of C. cornuta and supportstrophic relationships in aquatic ecosystems.  相似文献

The use of Assisted Reproductive Technologies (ARTs) in modern cattle breeding is an important tool for improving the production of dairy and beef cattle. A frequently employed ART in the cattle industry is in vitro production of embryos. However, bovine in vitro produced embryos differ greatly from their in vivo produced counterparts in many facets, including developmental competence. The lower developmental capacity of these embryos could be due to the stress to which the gametes and/or embryos are exposed during in vitro embryo production, specifically ovarian hormonal stimulation, follicular aspiration, oocyte in vitro maturation in hormone supplemented medium, sperm handling, gamete cryopreservation, and culture of embryos. The negative effects of some ARTs on embryo development could, at least partially, be explained by disruption of the physiological epigenetic profile of the gametes and/or embryos. Here, we review the current literature with regard to the putative link between ARTs used in bovine reproduction and epigenetic disorders and changes in the expression profile of embryonic genes. Information on the relationship between reproductive biotechnologies and epigenetic disorders and aberrant gene expression in bovine embryos is limited and novel approaches are needed to explore ways in which ARTs can be improved to avoid epigenetic disorders.  相似文献

Embryonic development and larval morphology of Chromis crusma was described from five nests sampled between 21 and 25 m depth in central Chile (33°S). From each nest, a set of c. 100 randomly selected eggs were hand-collected and transported in seawater to the laboratory. Subsets of c. 30 eggs per nest were maintained in 50 ml glass containers at a constant ambient temperature of c. 12°C (range 11.5–12.9°C). Egg length (L) and width (W) and larval notochordal length (LN) were measured from photographs. Geometric morphometric analyses were performed in newly hatched and 1 week old larvae to quantify shape changes. Ellipsoid eggs had an average (mean ± SE) size of 1.12 ± 0.05 mm L and 0.67 ± 0.02 mm W, with volume being similar throughout 15 developmental stages (i.e., ellipsoid-shaped; 0.27 mm3). Planktonic larvae hatched between 5 and 11 days at 12°C and had a mean LN of 3.13 ± 0.25 mm, a yolk sack volume of 0.03 mm3 and an oil droplet volume of 0.005 mm3. Morphological traits at hatching included: (a) lack of paired fins and jaws; (b) single medial fin fold; (c) lack of eye pigmentation; (d) yolk sac present near anterior tip; (e) melanophores distributed along ventral surface with one pair over the forehead. In order to generate an up-to-date summary of developmental traits within Pomacentridae, we reviewed literature on egg development (e.g., shape and number of oil droplets), hatching and larval traits (e.g., morphology, pigmentation patterns). Thirty-two publications accounting for 35 species were selected, where eggs, embryonic development, hatching and larval traits were found for 26, 21, 24 and 34 species, respectively. In order to evaluate potential phylogenetic and environmental relationships within the early stages of Pomacentridae, cluster analyses (Bray Curtis similarity, group average) were also performed on egg and larval traits of 22 species divided by subfamily (Stegastinae, Chrominae, Abudefdufinae, Pomacentrinae) and thermal ranges (i.e., low: 16.5°C (range: 12–21°C), medium: 24.5°C (range:21–28°C) and high: 27°C (range: 26–28°C)), suggesting that early developmental patterns can be segregated by both temperature and phylogenetic relationships.  相似文献

Using an extensive database compiled by scientific observers aboard commercial fishing operations between 1984 and 2014, we describe the maturity and size structures of white warehou Seriolella caerulea and silver warehou Seriolella punctata from by-catch of the trawling industrial fisheries operating in the austral zone off Chile. Macroscopic maturity stages and gonadosomatic (IGS) index show mature females throughout the year and a pronounced spawning period in both austral autumn and winter seasons, with an IGS peak in July for S. punctata and August for S. caerulea. Reproductive patterns in both species show an extended spawning season (July to September) across the area between 43 and 47° S. Length–mass relationships showed significant differences between sexes in both species, where females reach a larger size. Fork length at 50% maturity was 43.5 cm for S. caerulea and 37.2 cm for S. punctata. Female catch composition is dominated by adult fish (96% for S. caerulea and 86% for S. punctata). Currently, both species are exploited with no assessment and management-decision framework. Therefore, information regarding reproductive biology and demographic traits becomes an important baseline to ensure adequate fisheries management for both species.  相似文献

Chagas disease is one of the most neglected tropical diseases in the world, affecting nearly 15 million people, primarily in Latin America. Only two drugs are used for the treatment of this disease, nifurtimox and benznidazole. These drugs have limited efficacy and frequently induce adverse effects, limiting their usefulness. Consequently, new drugs must be found. In this study, we demonstrated the in vitro trypanocidal effects of a series of four gallic acid derivatives characterized by a gallate group linked to a triphenylphosphonium (TPP+) moiety (a delocalized cation) via a hydrocarbon chain of 8, 10, 11, or 12 atoms (TPP+-C8, TPP+-C10, TPP+-C11, and TPP+-C12, respectively). We analyzed parasite viability in isolated parasites (by MTT reduction and flow cytometry) and infected mammalian cells using T. cruzi Y strain trypomastigotes. Among the four derivatives, TPP+-C10 and TPP+-C12 were the most potent in both models, with EC50 values (in isolated parasites) of 1.0 ± 0.6 and 1.0 ± 0.7 μM, respectively, and were significantly more potent than nifurtimox (EC50 = 4.1 ± 0.6 μM). At 1 μM, TPP+-C10 and TPP+-C12 induced markers of cell death, such as phosphatidylserine exposure and propidium iodide permeabilization. In addition, at 1 μM, TPP+-C10 and TPP+-C12 significantly decreased the number of intracellular amastigotes (TPP+-C10: 24.3%, TPP+-C12: 19.0% of control measurements, as measured by DAPI staining) and the parasite’s DNA load (C10: 10%, C12: 13% of control measurements, as measured by qPCR). Based on the previous mode of action described for these compounds in cancer cells, we explored their mitochondrial effects in isolated trypomastigotes. TPP+-C10 and TPP+-C12 were the most potent compounds, significantly altering mitochondrial membrane potential at 1 μM (measured by JC-1 fluorescence) and inducing mitochondrial transition pore opening at 5 μM. Taken together, these results indicate that the TPP+-C10 and TPP+-C12 derivatives of gallic acid are promising trypanocidal agents with mitochondrial activity.  相似文献

The effects of the red imported fire ant, Solenopsis invicta Buren (Hymenoptera: Formicidae), on cotton aphid, Aphis gossypii Glover, populations and its predation of bollworm, Helicoverpa zea (Boddie), and beet armyworm, Spodoptera exigua (Hübner), (both Lepidoptera: Noctuidae) eggs were evaluated in cotton under field conditions during 2001 and 2002 in central and northern Texas. In central Texas, cotton aphid populations were approximately 5.5 times greater and predation of sentinel bollworm eggs 2 times greater in the presence of S. invicta versus in its absence, although aphid populations did not reach economic levels. Most predation of beet armyworm egg masses, measured via direct nocturnal observations, was due to S. invicta (68%) and cotton fleahopper, Pseudatomoscelis seriatus (Reuter) (21%), where S. invicta was present, and by the mite Abrolophus sp. (52%), spiders (13%), and minute pirate bug (Orius sp.) (13%) where S. invicta was absent. Predation of sentinel bollworm eggs and beet armyworm egg masses was approximately 1.5 and 4.1 times greater, respectively, in the presence of S. invicta versus in their absence. In the presence of S. invicta, the relative frequencies of minute pirate bug and cotton fleahopper were higher, and of S. invicta and native ants lower in beat bucket samples compared with their relative frequencies in nocturnal observations of predation upon beet armyworm egg masses. In the absence of S. invicta seven of eight predators sampled were similarly represented in beat bucket samples and nocturnal observations of beet armyworm egg mass predation, whereas minute pirate bug occurred at a higher frequency in beat bucket samples relative to nocturnal observations. These observations suggested that the relative frequencies of minute pirate bug, cotton fleahopper, S. invicta and native ants in beat bucket samples do not closely reflect the frequency with which these predators prey on noctuid eggs. Overall, the results of this study show that although S. invicta may promote aphid populations early in the growing season, it is an important predator of bollworm and beet armyworm eggs later in the season.  相似文献

The resurrection plant Craterostigma plantagineum Hochst. is used as an experimental system to investigate desiccation tolerance in higher plants. A search for genes activated during early stages of dehydration identified the gene CpEdi-9, which is expressed in mature seeds and in response to dehydration in the phloem cells of vascular tissues of leaves. Elements for the tissue-specific expression pattern reside in the isolated promoter of the CpEdi-9 gene, as shown through the analysis of transgenic plants. The CpEdi-9 promoter could be a suitable tool for expressing genes in the vascular system of dehydrated plants. CpEdi-9 encodes a small (10 kDa) hydrophilic protein, which does not have significant sequence homologies to known genes. The predicted protein CpEDI-9 shares some physicochemical features with LEA proteins from plants and a nematode. Based on the unique expression pattern and on the nucleotide sequence we propose that CpEdi-9 defines a new class of hydrophilic proteins that are supposed to contribute to cellular protection during dehydration.
