The influence of Calacarus heveae Feres on physiological processes was evaluated in two rubber tree clones. Experiments were conducted in a greenhouse with 5-month-old potted seedlings of RRIM 600 and GT 1 clones, that were either infested with C. heveae or not (non-infested control). The level of photosynthetic pigments, net photosynthetic rate, stomatal conductance, transpiration rate, changes in relative humidity between leaf surface and ambient air (Δw) and intercellular CO(2) concentration (Ci CO(2)) were evaluated. Infested plants showed significant reductions in the rate of transpiration, the rate of photosynthesis, stomatal conductance and Δw. RRIM 600 seedlings showed more pronounced physiological damage than GT 1 seedlings, indicating a lower physiological tolerance of the former clone to the mite. However, carotenoid levels were reduced only in GT 1 seedlings. Photosynthesis was probably reduced due to a decrease in stomatal opening, as indicated by reductions in transpiration rate and stomatal conductance and by the absence of differences in chlorophyll levels between treatments. Our results indicate that populations of C. heveae reduce the productivity of rubber trees. Thus, farmers must to be aware to control this mite pest in rubber tree plantations.  相似文献

The mycotoxin aflatoxin B1 (AFB1) is a carcinogenic food contaminant which is metabolically activated by epoxydation. The metabolism of mycotoxins via the mercapturate metabolic pathway was shown, in general, to lead to their detoxication. Mercapturic acids thus formed (S-substitued-N-acetyl-l-cysteines) may be accumulated in the kidney and either excreted in the urine or desacetylated by Acylase 1 (ACY1) to yield cysteine S-conjugates. To be toxic, the N-acetyl-l-cysteine-S-conjugates first have to undergo deacetylation by ACY 1. The specificity and rate of mercapturic acid deacetylation may determine the toxicity, however the exact deacetylation processes involved are not well known. The aim of this study was to investigate the role of ACY1 in the toxicity of some bioactive epoxides from Aflatoxin B1. We characterized the kinetic parameters of porcine kidney and human recombinant aminoacylase-1 towards some aromatic and aliphatic-derived mercapturates analogue of mycotoxin-mercapturic acids and 3,4-epoxyprecocene, a bioactive epoxide derivated from aflatoxin. The deacetylation of mercapturated substrates was followed both by reverse phase HPLC and by TNBS method. Catalytic activity was discussed in a structure-function relationship. Ours results indicate for the first time that aminoacylase-1 could play an important role in deacetylating mercapturate metabolites of aflatoxin analogues and this process may be in relation with their cyto- and nephrotoxicity in human.  相似文献

doi: 10.1111/j.1741‐2358.2011.00587.x The influence of double flask investing on tooth displacement in dentures processed by microwave irradiation Objective: This study evaluated the influence of the bimaxillary flask (BMF) and two different investing materials on first molar inclination in dentures processed by microwave irradiation. Background: The BMF may minimise tooth displacement, saving time and improving occlusion. Methods and materials: Forty pairs of dentures were randomised into four groups: stone wall in monomaxillary flask; silicone wall in BMF; stone wall in BMF; acrylic resin retentions and silicone in BMF. Dentures were processed by microwave irradiation. Two referential points were established on tooth surface. A microscope and a digital pachymeter were used to measure the distance between these points, and the angles α (right maxillary molar), β (left maxillary molar), α′ (right mandibular molar) and β′ (left mandibular molar) were calculated by the law of cosines. Data were submitted to Kruskal–Wallis (5% significance). Results: No difference was observed among the groups (p > 0.05). In the intra‐group analysis, α was significantly different for groups I, II and III; α′, for groups II and IV; β, for all groups; β′, for groups III and IV. Conclusion: First molar inclination was similar for monomaxillary and BMFs. The use of stone or silicone as investing materials presented the same effect on tooth inclination.  相似文献

doi: 10.1111/j.1741‐2358.2011.00454.x Influence of surface treatments on the flexural strength of denture base repair Objective: The purpose of this study was to evaluate the flexural strength of repairs made with autopolymerising acrylic resin after different treatments of joint surfaces. Material and Methods: Fifty rectangular specimens were made with heat‐polymerised acrylic resin and 40 were repaired with autopolymerising acrylic resin following joint surface treatments: group 1 (intact specimens), group 2 (chemical treatment: wetting with methyl‐methacrylate for 180 s), group 3 (abraded with silicon carbide paper), group 4 (abraded and wetting with methyl‐methacrylate for 180 s) and group 5 (without surface treatment). The flexural strength was measured by a three‐point bending test using a universal testing machine with a 100 Kgf load cell in the centre of repair at 5 mm/min cross‐head speed. All data were analysed using one‐way anova and Tukey HSD test for multiple comparisons (p < 0.05). Results: Among repaired specimens, groups 2 and 4 had 66.53 ± 3.4 and 69.38 ± 1.8 MPa mean values and were similar. These groups had superior flexural strength than groups 3 and 5 that were similar and had 54.11 ± 3.4 and 51.24 ± 2.8 MPa mean values, respectively. Group 1 had a mean value of 108.30 ± 2.8 MPa being the highest result. Conclusion: It can be concluded that the treatment of the joint surfaces with methyl‐methacrylate increases the flexural strength of denture base repairs, although the strength is still lower than that observed for the intact denture base resin. Abrasion with sandpaper was not able to influence the flexural strength of repaired denture bases.  相似文献

Evaluation of Vickers hardness of different types of acrylic denture base resins with and without glass fibre reinforcement Objective: To evaluate the Vickers hardness of different acrylic resins for denture bases with and without the addition of glass fibres. Background: It has been suggested that different polymerisation methods, as well as the addition of glass fibre (FV) might improve the hardness of acrylic. Materials and methods: Five types of acrylic resin were tested: Vipi Wave (VW), microwave polymerisation; Vipi Flash (VF), auto‐polymerisation; Lucitone (LT), QC20 (QC) and Vipi Cril (VC), conventional heat‐polymerisation, all with or without glass fibre reinforcement (GFR) and distributed into 10 groups (n = 12). Specimens were then submitted to Vickers hardness testing with a 25‐g load for 30 s. All data were submitted to anova and Tukey’s HSD test. Results: A significant statistical difference was observed with regard to the polymerisation method and the GFR (p < 0.05). Without the GFR, the acrylic resin VC presented the highest hardness values, and VF and LT presented the lowest. In the presence of GFR, VC resin still presented the highest Vickers hardness values, and VF and QC presented the lowest. Conclusions: The acrylic resin VC and VW presented higher hardness values than VF and QC resins. Moreover, GFR increased the Vickers hardness of resins VW, VC and LT.  相似文献

doi: 10.1111/j.1741‐2358.2011.00463.x Effect of water storage and heat treatment on the cytotoxicity of soft liners Objective: To evaluate the effect of water storage time on the cytotoxicity of soft liners. Methods: Sample discs of soft liners Dentusoft, Dentuflex, Trusoft, Ufi‐Gel‐P and denture base acrylic resin Lucitone‐550 were prepared and divided into four groups: GN: No treatment, G24: Stored in water at 37°C for 24 h; G48: Stored in water at 37°C for 48 h, GHW: Immersed in water at 55°C for 10 min. To analyse the cytotoxic effect, three samples of each group were placed in tubes with Dubelcco’s Modified Eagle Mediums and incubated at 37°C for 24 h. During this period, the toxic substances were leached to the culture medium. The cytotoxicity was analysed quantitatively by the incorporation of radioactivity 3H‐thymidine checking the number of viable cells (synthesis of DNA). The data were statistically analysed using two‐way anova and Tukey’s honestly significant difference tests (α = 0.05). Results: Treatments did not reduce the cytotoxicity effect of the soft liners (p > 0.05). It was found that Ufi‐Gel‐P had a non‐cytotoxic effect, Trusoft had a slightly cytotoxic effect, Dentuflex had a moderated cytotoxic effect, Dentusoft alternated between slightly and non‐cytotoxic effect, and Lucitone‐550 had non‐cytotoxic effect when stored in water for 48 h. Conclusion: The effect of water storage and the heat treatment did not reduce the cytotoxicity of the soft liners.  相似文献

doi: 10.1111/j.1741‐2358.2011.00568.x Discussions on oral health care among elderly Chinese immigrants in Melbourne and Vancouver Background: This study explored how elderly Chinese immigrants value and relate to how acculturation influences oral health and subsequent service use. Methods: Elders who had immigrated to Melbourne and Vancouver within the previous 15 years were recruited from local community centres and assigned to focus groups of 5–7 participants in Vancouver (4 groups) or Melbourne (5 groups). Results: Following an iterative process of thematic analysis, the discussions revealed that immigrants care about the comfort and appearance of their teeth, and they value Western dentistry as a supplement to traditional remedies, but they have difficulty getting culturally sensitive information about oral health care. Accessing dentistry, they explained, is distressing because of language problems and financial costs that impose on their children. Consequently, many immigrants obtain dental treatment in China when they return for occasional visits. They felt that separation of dentistry from national health care programmes in Canada and Australia disregards natural links between oral health and general health. Conclusions: The similarity of concerns in both cities suggests that dissemination of information and availability of services are the important themes influencing oral health, and that, beliefs developed over a lifetime play an important role in interpreting oral health in the host country.  相似文献

Aedes aegypti control is crucial to reducing dengue fever. Aedes aegypti larvae have developed resistance to organophosporous insecticides and the use of natural larvicides may help manage larval resistance by increasing elements in insecticide rotation programs. Here, we report on larvicidal activity of Moringa oleifera flower extract against A. aegypti L(1), L(2), L(3), and L(4) as well as the effect of flower extract on gut trypsin and whole-larval acetylcholinesterase from L(4.) In addition, the heated flower extract was investigated for larvicidal activity against L(4) and effect on larval gut trypsin. Moringa oleifera flower extract contains a proteinaceous trypsin inhibitor (M. oleifera flower trypsin inhibitor, MoFTI), triterpene (β-amyrin), sterol (β-sitosterol) as well as flavonoids (kaempferol and quercetin). Larvicidal activity was detected against L(2), L(3), and L(4) (LC(50) of 1.72%, 1.67%, and 0.92%, respectively). Flower extract inhibited L(4) gut trypsin (MoFTI K(i) = 0.6 nM) and did not affect acetylcholinesterase activity. In vivo assay showed that gut trypsin activity from L(4) treated with M. oleifera flower extract decreased over time (0-1,440 min) and was strongly inhibited (98.6%) after 310 min incubation; acetylcholinesterase activity was not affected. Thermal treatment resulted in a loss of trypsin inhibitor and larvicidal activities, supporting the hypothesis that flower extract contains a proteinaceous trypsin inhibitor that may be responsible for the deleterious effects on larval mortality.  相似文献

We have reported previously that human prostate-derived sterile 20-like kinase (PSK) 1 alters actin cytoskeletal organization and binds to microtubules, regulating their organization and stability. We have shown a structurally related protein kinase PSK2, which lacks a microtubule-binding site, activated c-Jun N-terminal kinase (JNK), and induced apoptotic morphological changes that include cell contraction, membrane blebbing, and apoptotic body formation. Apoptotic stimuli increased the catalytic activity of endogenous PSK2 and JNK, and dominant negative JNK or a physiological inhibitor of JNK blocked these apoptotic morphological responses to PSK2, demonstrating a requirement for JNK. PSK2 also stimulated the cleavage of Rho kinase-1 (ROCK-I), and the activity of ROCK-I was required for PSK2 to induce cell contraction and membrane blebbing. The activation of caspases was also needed for the induction of membrane blebbing by PSK2, which was itself a substrate for caspase 3. PSK2 therefore regulates apoptotic morphology associated with the execution phase of apoptosis, which involves dynamic reorganization of the actin cytoskeleton, via downstream targets that include JNK and ROCK-I. Our findings suggest that PSKs form a subgroup of sterile 20 (STE20)-like kinases that regulate different cytoskeletal processes.  相似文献
