New genetic markers were characterized for the broad-snouted caiman (Caiman latirostris) by constructing libraries enriched for microsatellite DNA. Construction and characterization of these libraries are described in the present study. One microsatellite marker was developed from a (ACC-TGG)(n)enriched microsatellite DNA library, and 12 microsatellite markers were developed from a (AC-TG)(n)enriched microsatellite DNA library. These markers were tested in wild-caught animals, and these tests resulted in ten new polymorphic microsatellites for C. latirostris.  相似文献

In theory, geographic scale is related to genetic variation at the population level, whereas microgeographic scale may reveal intra-population structure such as social groups and families. In the present work, both levels of genetic variation in the broad-snouted caiman (Caiman latirostris) were evaluated in small wetlands associated with the Piracicaba River and some of its tributaries in the state of S?o Paulo, Brazil. Genetic variation was determined using microsatellite DNA markers originally developed for the American alligator (Alligator mississipiensis) and previously tested in pedigreed captive broad-snouted caimans. Using these markers, we were able to detect variability among individuals from different sites, even those within a small geographic distance. Genetic results suggest that the groups sampled at each site are composed predominantly of related individuals. A possible combination of high mortality and low natality rates results in a low number of successfully dispersed individuals per generation. Future studies using a recently constructed Caiman latirostris microsatellite library (Zucoloto et al., 2002) might help us to understand metapopulation processes that may be occurring within this species.  相似文献

Low-level laser energy has been increasingly used in the treatment of a broad range of conditions and has improved wound healing, reduced edema, and relieved pain of various etiologies. This study examined whether 635-nm low-level lasers had an effect on adipose tissue in vivo and the procedural implementation of lipoplasty/liposuction techniques. The experiment investigated the effect of 635-nm, 10-mW diode laser radiation with exclusive energy dispersing optics. Total energy values of 1.2 J/cm(2), 2.4 J/cm(2), and 3.6 J/cm(2) were applied on human adipose tissue taken from lipectomy samples of 12 healthy women. The tissue samples were irradiated for 0, 2, 4, and 6 minutes with and without tumescent solution and were studied using the protocols of transmission electron microscopy and scanning electron microscopy. Nonirradiated tissue samples were taken for reference. More than 180 images were recorded and professionally evaluated. All microscopic results showed that without laser exposure the normal adipose tissue appeared as a grape-shaped node. After 4 minutes of laser exposure, 80 percent of the fat was released from the adipose cells; at 6 minutes of laser exposure, 99 percent of the fat was released from the adipocyte. The released fat was collected in the interstitial space. Transmission electron microscopic images of the adipose tissue taken at x60,000 showed a transitory pore and complete deflation of the adipocytes. The low-level laser energy affected the adipose cell by causing a transitory pore in the cell membrane to open, which permitted the fat content to go from inside to outside the cell. The cells in the interstitial space and the capillaries remained intact. Low-level laser-assisted lipoplasty has a significant impact on the procedural implementation of lipoplasty techniques.  相似文献

Studies from our laboratory have demonstrated rapid ( < 1 min) non-genomic activation of Na(+)-H(+) exchange, K(+) recycling, PKC activity and a PKC-dependent Ca(2+) entry through L-type Ca(2+) channels specifically by mineralocorticoids in distal colon. Aldosterone directly stimulates the activity of the PKC alpha isoform (but not PKC delta, PKC epsilon and PKC zeta) in a cell-free assay system containing only purified commercially available enzyme, appropriate substrate peptide, co-factors and lipid vesicles. The primary ion transport target of the non-genomic signal transduction cascade elicited by aldosterone in epithelia is the Na(+)-H(+) exchanger. In isolated colonic crypts, aldosterone produced a PKC alpha sensitive intracellular alkalinisation within 1 min of hormone addition. Intracellular alkalinisation upregulates an ATP-dependent K(+) channel, which is involved in K(+) recycling to maintain the electrical driving force for Na(+) absorption, while inhibiting a Ca(2+) -dependent K(+) channel, which generates the charge balance for Cl(-) secretion. The non-genomic response to aldosterone in distal colon appears to enhance the capacity for absorption while down-regulating the potential for secretion. We have also demonstrated rapid (< 1 min) non-genomic activation of Na(+)-H(+) exchange, K(+) recycling, PKC alpha activity, and a PKC delta- and PKA-dependent Ca(2+) entry through di-hydropyridine-blockable Ca(2+) channels specifically by 17beta-estradiol in distal colon. These rapid effects are female gender specific and are insensitive to inhibitors of the classical estrogen receptor (ER). 17 beta-Estradiol directly stimulated the activity of both PKC delta and PKC alpha (but not PKC epsilon or PKC zeta) in a cell-free assay system. E2 rapidly inhibited basolateral K(Ca) channel activity which would be expected to result in an acute inhibition of Cl(-) secretion. Physiological concentrations of E2 (0.1-10 nM) reduced both basal and secretagogue-induced Cl(-) secretion. This anti-secretory effect of E2 is sensitive to PKC inhibition, intracellular Ca(2+) chelation, and is female gender specific and insensitive to inhibitors of the classical ER. These observations link rapid non-genomic activation of second messengers with a rapid gender-specific physiological effect in the whole tissue. Aldosterone and E2 differ in their protein kinase signal transduction and both hormones stimulate specific PKC isoforms indicating both common and divergent signalling systems for salt-retaining steroid hormones. The physiological function of non-genomic effects of aldosterone and estradiol is to shift the balance from net secretion to net absorption in a pluripotential epithelium.  相似文献

MOTIVATION: The current data explosion is intractable without advanced data management systems. The numerous data sets become really useful when they are interconnected under a uniform interface--representing the domain knowledge. The SRS has become an integration system for both data retrieval and applications for data analysis. It provides capabilities to search multiple databases by shared attributes and to query across databases fast and efficiently. RESULTS: Here we present recent developments at the EBI SRS server (http://srs.ebi.ac.uk). The EBI SRS server contains today more than 130 biological databases and integrates more than 10 applications. It is a central resource for molecular biology data as well as a reference server for the latest developments in data integration. One of the latest additions to the EBI SRS server is the InterPro database-Integrated Resource of Protein Domains and Functional Sites. Distributed in XML format it became a turning point in low level XML-SRS integration. We present InterProScan as an example of data analysis applications, describe some advanced features of SRS6, and introduce the SRSQuickSearch JavaScript interfaces to SRS.  相似文献

The foraging ecology of hummingbirds involves the exploitation of a high number of patchily distributed flowers. This scenario seems to have influenced capabilities related to learning and memory, which help to avoid recently visited flowers and to allocate exploitation to the most rewarding flowers, once learning has occurred. We carried out two field experiments with the green‐backed firecrown hummingbird (Sephanoides sephaniodes, Trochilidae) in order to examine the ability of birds, first, to recall a nectar location, and secondly, to remember the location of the most rewarding flower among lower quality flowers. The first experiment showed that subjects were able to recall the location of nectar among flowers of identical appearance. In the second experiment, hummingbirds were also able to recall the location of the most rewarding nectar among less rewarding flowers with the same appearance. The results of this study suggest that S. sephaniodes can remember the location of the most rewarding patch, facilitating efficient exploitation of flowers in the absence of visual cues related to nectar quality.  相似文献

Fruit-set and fruit growth in pea (Pisum sativum L.) depend on gibberellins (GAs). The authors have isolated a new pea mutant, gio, which appeared spontaneously within the population of the cultivar Alaska, characterized by unpollinated ovaries much less sensitive to applied GAs. The mutant also has elongated peduncles, and is taller than the wild-type (WT) because the upper plant internodes are longer. Contrary to WT, the gio ovaries respond very little to benzylaminopurine (BAP) and 2,4-dichlorophenoxyacetic acid, but become fully sensitive to GA(3) when this hormone is applied together with BAP. The gio phenotype is determined by a mutation at a single mendelian locus. The mutation is recesive, shows incomplete penetrance, and its expression depends on environmental culture conditions. The sensitivity of the ovaries to GA(3) can be recovered by removing the apical shoot (plant decapitation) and by blocking the transport of indoleacetic acid (IAA) from the apical shoot with 2,3,5-triiodobenzoic acid. The content of IAA in methanolic extracts and phloematic exudates of the apical shoot of gio is about double that in the WT. The rate of transport of [(3)H]IAA applied to the apex of the mutant is also twice that in the WT. This indicates that the insensitivity of the gio ovaries to GAs is due to the inhibitory effect of the higher basipetal IAA transport from the shoot. The interaction between the fruit and the apical shoot mediated by IAA probably also involves cytokinins transported from the basal part of the plant.  相似文献

Commercially avaiable pine bark nuggets (composted) and fresh pine bark were ground into powders and added at rates of 0 to 50 g kg–1 to field soil infested withMeloidogyne arenaria andHeterodera glycines. The treated soil was maintained moist in the greenhouse for 2 weeks, sampled, and planted with Davis soybean (Glycine max.). Eight weeks after planting, numbers ofM. arenaria andH. glycines in soil decreased with increasing amounts of composted or fresh pine bark. No juveniles were present in soil treated with 5% pine bark. The number of galls and cysts g–1 root decreased in proportion to the amount of pine bark added to soil. Gall and cyst formation was completely eliminated at the 5% rate. Numbers of saprophagous nematodes were highest in soil with 4–5% pine bark. The activity of several soil enzymes was correlated with the addition of both composted and fresh pine bark. Fresh pine bark powder caused an increase in soil enzyme activity compared to composted pine bark, but did not provide consistent control of gall and cyst formation.Journal Series No. 18-933579 Alabama Agricultural Experiment Station  相似文献
