The specialized microenvironment or niche where stem cells reside provides regulatory input governing stem cell function. We tested the hypothesis that targeting the niche might improve stem cell-based therapies using three mouse models that are relevant to clinical uses of hematopoietic stem (HS) cells. We and others previously identified the osteoblast as a component of the adult HS cell niche and established that activation of the parathyroid hormone (PTH) receptor on osteoblasts increases stem cell number. Here we show that pharmacologic use of PTH increases the number of HS cells mobilized into the peripheral blood for stem cell harvests, protects stem cells from repeated exposure to cytotoxic chemotherapy and expands stem cells in transplant recipients. These data provide evidence that the niche may be an attractive target for drug-based stem cell therapeutics.  相似文献

BACKGROUND: Annexin 1 is a 37-kDa protein that has complex intra- and extracellular effects. To discover whether the absence of this protein alters bone development, we monitored this event in the annexin-A1 null mice in comparison with littermate wild-type controls. METHODS: Radiographic and densitometry methods were used for the assessment of bone in annexin-A1 null mice at a gross level. We used whole-skeleton staining, histological analysis, and Western blotting techniques to monitor changes at the tissue and cellular levels. RESULTS: There were no gross differences in the appendicular skeleton between the genotypes, but an anomalous development of the skull was observed in the annexin-A1 null mice. This was characterized in the newborn annexin-A1 null animals by a delayed intramembranous ossification of the skull, incomplete fusion of the interfrontal suture and palatine bone, and the presence of an abnormal suture structure. The annexin-A1 gene was shown to be active in osteocytes during this phase and COX-2 was abundantly expressed in cartilage and bone taken from annexin-A1 null mice. CONCLUSIONS: Expression of the annexin-A1 gene is important for the normal development of the skull in mice, possibly through the regulation of osteoblast differentiation and a secondary effect on the expression of components of the cPLA2-COX-2 system.  相似文献

N,N'-diarylsquaramides were prepared and evaluated as antagonists of CXCR2. The compounds were found to be potent and selective antagonists of CXCR2. Significant differences in SAR was observed relative to the previously described N,N'-diarylurea series. As was the case in the N,N'-diarylurea series, placing sulfonamide substituent adjacent to the acidic phenol significantly reduced the clearance in rat pharmacokinetic studies.  相似文献

Recent literature has suggested the benefit of selective PPARdelta agonists for the treatment of atherosclerosis and other disease states associated with the metabolic syndrome. Herein we report the synthesis and structure-activity relationships of a series of novel and selective PPARdelta agonists. Our search began with identification of a novel benzothiophene template which was modified by the addition of various thiazolyl, isoxazolyl, and benzyloxy-benzyl moieties. Further elucidation of the SAR led to the identification of benzofuran and indole based templates. During the course of our research, we discovered three new chemical templates with varying degrees of affinity and potency for PPARdelta versus the PPARalpha and PPARgamma subtypes.  相似文献

Batesian mimicry occurs when a palatable species (the mimic) gains protection from predators by resembling an unpalatable or otherwise protected species (the model). While some mimetic species resemble their models closely, other species ('imperfect mimics') are thought to bear only a crude likeness. In an earlier study, pigeons (Columba livia) were trained to recognize wasp images in one experiment and non-mimetic (NM) fly images in another by rewarding the pigeons for pecking on the respective image types. These pigeons were subsequently presented with different images, including seemingly wasp-like hoverfly species, and the recorded peck rates on these images were used as a measure of the pigeons' perception of the hoverflies' mimetic similarity. To identify a candidate set of morphological features that the pigeons used when assessing this mimetic similarity, we first extracted a range of biometrical measurements from images originally presented to the pigeons. We then repeatedly optimized an empirical model in an attempt to match the recorded pigeon peck rates while using as few biometrical features as input as possible. Our models were able to fit the pigeon peck rates with considerable accuracy even while excluding many input features. Antennal length, a feature commonly used to discriminate between flies and wasps, was regularly retained as an input variable, but overall a different set of biometrical features was important for predicting the peck rates of pigeons rewarded for identifying wasps compared to those rewarded for identifying NM flies. In highlighting the importance of specific biometrical features in promoting mimicry and the irrelevance of others, our optimized models provide an explanation as to why certain species that appear to be poor mimics to humans are judged to be good mimics by birds.  相似文献

We calculate posterior probabilities for candidate genes as a function of genomic location. Posterior probabilities for quantitative trait loci (QTL) presence in a small interval are calculated using a Bayesian model-selection approach based on the Bayesian information criterion (BIC) and used to combine QTL colocation information with sequence-specific evidence, e.g., from differential expression and/or association studies. Our method takes into account uncertainty in estimation of number and locations of QTL and estimated map position. Posterior probabilities for QTL presence were calculated for simulated data with n = 100, 300, and 1200 QTL progeny and compared with interval mapping and composite-interval mapping. Candidate genes that mapped to QTL regions had substantially larger posterior probabilities. Among candidates with a given Bayes factor, those that map near a QTL are more promising for further investigation with association studies and functional testing or for use in marker-aided selection. The BIC is shown to correspond very closely to Bayes factors for linear models with a nearly noninformative Zellner prior for the simulated QTL data with n > or = 100. It is shown how to modify the BIC to use a subjective prior for the QTL effects.  相似文献

Neutral models are often used as null models, testing the relative importance of niche versus neutral processes in shaping diversity. Most versions, however, focus only on regional scale predictions and neglect local level contributions. Recently, a new formulation of spatial neutral theory was published showing an incompatibility between regional and local scale fits where especially the number of rare species was dramatically under‐predicted. Using a forward in time semi‐spatially explicit neutral model and a unique large‐scale Amazonian tree inventory data set, we show that neutral theory not only underestimates the number of rare species but also fails in predicting the excessive dominance of species on both regional and local levels. We show that although there are clear relationships between species composition, spatial and environmental distances, there is also a clear differentiation between species able to attain dominance with and without restriction to specific habitats. We conclude therefore that the apparent dominance of these species is real, and that their excessive abundance can be attributed to fitness differences in different ways, a clear violation of the ecological equivalence assumption of neutral theory.  相似文献

Supplementation of infant formulas with prebiotic ingredients continues the effort to mimic functional properties of human milk. In this double-blind, controlled, 28-day study, healthy term infants received control formula (control group; n = 25) or control formula supplemented with polydextrose (PDX) and galactooligosaccharide (GOS) (4 g/liter) (PG4 group; n = 27) or with PDX, GOS, and lactulose (LOS) (either 4 g/liter [PGL4 group; n = 27] or 8 g/liter [PGL8 group; n = 25]). A parallel breast-fed group (BF group) (n = 30) was included. Stool characteristics, formula tolerance, and adverse events were monitored. Fecal bacterial subpopulations were evaluated by culture-based selective enumeration (Enterobacteriaceae), quantitative real-time PCR (Clostridium clusters I, XI, and XIV, Lactobacillus, and Bifidobacterium), and fluorescence in situ hybridization (FISH) (Bifidobacterium). Fecal bacterial community profiles were examined by using 16S rRNA gene PCR-denaturing gradient gel electrophoresis. The daily stool consistency was significantly softer or looser in the BF group than in all of the groups that received formula. The formulas were well tolerated, and the incidences of adverse events did not differ among feeding groups. Few significant changes in bacterial subpopulations were observed at any time point. The bacterial communities were stable; individual profiles tended to cluster by subject rather than by group. Post hoc analysis, however, demonstrated that the bacterial community profiles for subjects in the BF, PG4, PGL4, and PGL8 groups that first received formula at a younger age were less stable than the profiles for subjects in the same groups that received formula at an older age, but there was no difference for the control group. These data indicate that formulas containing PDX, GOS, and LOS blends are more likely to influence gut microbes when administration is begun in early infancy and justify further investigation of the age-related effects of these blends on fecal microbiota.  相似文献

Summary .  Little and An (2004,  Statistica Sinica   14, 949–968) proposed a penalized spline of propensity prediction (PSPP) method of imputation of missing values that yields robust model-based inference under the missing at random assumption. The propensity score for a missing variable is estimated and a regression model is fitted that includes the spline of the estimated logit propensity score as a covariate. The predicted unconditional mean of the missing variable has a double robustness (DR) property under misspecification of the imputation model. We show that a simplified version of PSPP, which does not center other regressors prior to including them in the prediction model, also has the DR property. We also propose two extensions of PSPP, namely, stratified PSPP and bivariate PSPP, that extend the DR property to inferences about conditional means. These extended PSPP methods are compared with the PSPP method and simple alternatives in a simulation study and applied to an online weight loss study conducted by Kaiser Permanente.  相似文献
