Orbitally magnetized organic solids

The preparation is discussed of polymeric organic solids in which large ring-like delocalized electronic orbitals can be made to enclose or trap magnetic flux. The situation is analogous to snapping shut a loop of superconductor while it is in a magnetic field. The operation of the Meissner effect acts to retain the trapped flux and create remant magnetization.     

Linkage Modification with Mixed Random Mating and Selfing: A Numerical Study

Although recombination cannot increase under conditions of random mating or complete selfing in regimes of constant selection, with mixed random mating and selfing, selection for increased recombination can occur. For some fitness regimes there may be selection for reduced recombination with both low and high degrees of selfing but selection for increased recombination with moderate degrees of selfing. With some fitness regimes there is a historical effect: depending on which equilibrium a population starts from, there may be selection for either increased or decreased recombination. In other cases the direction of selection may be determined by the present state of individuals within the population. If recombination is already fairly limited, there may be selection for further reduction. If recombination is already fairly frequent, there may be selection for increased recombination. For certain symmetric viability systems there may be an intermediate value of the recombination fraction between 0 and 0.5 toward which the population will evolve. Although it is not yet possible to classify precisely those fitness matrices that can exhibit selection for increased recombination, it does appear that selection for increased recombination can occur only if at least two of the double homozygotes are less fit than would be expected on the basis of a comparison of the fitnesses of the single and double heterozygotes on an additive scale.     

Modification of the zona-free hamster ova bioassay of boar sperm fertility and correlation with in vivo fertility

These studies were designed to evaluate the ability of the zona-free hamster ova bioassay to detect differences in fertility of boar sperm. In the first study, sperm from two previously infertile boars were compared to sperm from seven previously fertile boars. The percentage of zona-free hamster ova penetrated by sperm from the previously infertile boars was significantly lower than the percentage of ova penetrated by sperm from previously fertile boars (18% of ova penetrated vs. 83%, P < .001). In the 14 ejaculates from the previously infertile boars that had ejaculate motilities of 50% or greater, the percentage of zona-free hamster ova penetrated continued to be lower than in ejaculates from the fertile boars. One of the two previously infertile boars consistently had a normal semen analysis. The only two observed manifestations of his reduced fertility were his zero conception rate and the limited ability of his sperm to penetrate zona-free hamster ova. In the second study, females were inseminated with equal numbers of sperm from two previously fertile males and the paternity of offspring determined at birth. The experiment was replicated with four combinations of six boars. A high correlation was observed between the percentage of offspring sired and the ability to penetrate zona-free hamster ova (R = .89). Neither morphology nor the ability of the sperm to undergo an acrosome reaction during in vitro incubation was correlated with fertility in the competitive mating situation. These results suggest the zona-free hamster ova bioassay can improve the in vitro fertility assessment of fresh boar semen.     

Ineffectiveness of ethylene biosynthetic and action inhibitors in phenotypically reverting theEpinastic mutant of Tomato (Lycopersicon esculentum mill.)

Five-day-old, dark-grown seedlings of theEpinastic (Epi) tomato mutant (Lycopersicon esculentum Mill.) and its parent, cultivar VFN8, were used as a system for assessing the role of ethylene in theEpi phenotype. The distinguishing features ofEpi seedlings are an increase in hypocotyl diameter and reduced hypocotyl length. Treatment of VFN8 seedlings with 0.5 l/liter ethylene closely mimicked theEpi phenotype. The rate of ethylene production by 5-day-old, dark-grownEpi seedlings was double that of VFN8 seedlings. Nevertheless, treatment ofEpi seedlings with inhibitors of ethylene biosynthesis (aminoethoxyvinylglycine or Co²⁺) or ethylene action (silver thiosulfate or norbornadiene) failed to normalize theEpi phenotype.Epi seedlings grown in sealed jars containing ethylene and CO₂ adsorbants also expressed the characteristicEpi phenotype. The results indicate that the physiological lesion resulting from theEpi gene mutation is not simply an overproduction of ethylene.

Irradiation of cells in tissue culture

Summary Cultures of human amnion were employed to check the hypothesis that cell strains with heteroploid chromosome counts regularly produce giant cells within 12 days following treatment with 2000 r and 4000 r of gamma irradiation from a cobalt source, while this response has not been obtained from primary cultures whose cells were presumed to be diploid.The giant cell reaction not only was obtained from two transfer passage lines of a well-established amnion strain developed at Berkeley (No A 185-21C-26 and No A 185-21C-45) but was also found for a 20-day second passage culture of amnion. Since this line has continued to reproduce at a rapid rate, it is presumed to have assumed the features of a typical strain within the period of observation. This impression was reinforced by the finding that the chromosome number for 32 cells fixed on the 35th day had a modal value of 67.In contrast, both untrypsinized and trypsinized spindle cells in primary cultures as well as unaltered epithelial elements which had not been subcultured gave no evidence of giant cell formation 12 days after exposure to 2000 r and 4000 r from a Cobalt⁶⁰ source.These data lend evidence that giant cell formation is related to the chromosomal constitution of the irradiated elements.This research was supported by funds provided under Contract AF 18(600)-1263 with the School of Aviation Medicine, USAF, Randolph Air Force Base, Texas.Fellow of the Instituto de Alta Cultura and the Fundacão Calouste Gulbenkian of Lisbon, Portugal.Tobacco Industry Research Committee Fellow.     

Pollen frequencies reflect vegetation patterns in a great basin (U.S.A.) mountain range

Five indices are used to quantify the relationship between vegetation and pollen in a mountain range in the arid Great Basin. Computations are based on vegetation coverage and pollen percentages from 63 stands. Association is a measure of whether the presence of the pollen type in a surface sample is an indication of the presence of the parent plant in the local vegetation. Over-representation and under-representation measure tendencies for pollen to occur where the parent plants are absent and vice versa. The correlation coefficient measures the relationship between plant and pollen in stands where both are present. Twenty-nine trees, shrubs, and herbs accurately reflect local vegetation conditions. A percentage diagram shows elevational trends in abundant pollen types. Regional pollen types are used to compute the accumulation rate of pollen in the surface samples. A diagram of pollen accumulation rates shows trends similar to those shown in the percentage diagram. The moss polsters used in this study may collect pollen over a fifteen-year interval.     

IDENTIFICATION OF SEROTONIN WITHIN VITAL-STAINED NEURONS FROM LEECH GANGLIA

Abstract— We have measured serotonin (5-HT) within large and small neurosomata which are vitally stained by Neutral Red dye. A micro-radioenzymatic technique which is sensitive to 50fmol of 5-HT was employed on intact ganglia, 75 μm Retzius Cells (RZ) and a 10 μm ventro-lateral cell (VL) taken from the leech Macrobdella decora. The stain does not affect the levels of 5-HT in either ganglia or RZ. The VL cell body contains 5-HT at concentrations of at least 100 m m . Microspectrofluorometry of all the ganglionic neurosomata which fluoresce following the Falck-Hillarp formaldehyde condensation reaction detected rapidly-fading emission peaks of 509–523 nanometers. We conclude that all seven fluorescent neurons in the leech ganglion very probably contain serotonin.
