A simple system for the Collection of Leaf-Derived Faecal Pellets of Aquatic Invertebrates

Animals and food (leaves) are confined at a desired temperature in a feeding chamber in the upper portion of the water column of a collector system. Egested material collects in water at 0°C also eliminates coprophagy and physical damage to the pellets.     

The purification of murine histocompatibility antigens (H-2b) from RBL-5 tumor cells using detergents.

A detergent-solubilized form of H-2b (dH-2b) has been purified 1500-fold from RBL-5 tumor cells. The purification was accomplished by deoxycholate solubilization of purified plasma membranes, gel filtration, Lens culinaris lectin affinity chromatography, and affinity chromatography on a sheep anti-dH-2b immunoadsorbent. Both alloantigen and beta 2-microglobulin were monitored by radioimmunoassay during purification. The final product was judged to be greater than 90% pure by the following criteria: 1) sodium dodecyl sulfate-polyacrylamide electrophoresis which showed the expected 2-component structure of histocompatibility antigens, i.e. a heavy chain and beta 2-microglobulin; 2) amino acid composition which was comparable to the known compositions of other H-2 and HLA molecules; 3) NH2-terminal sequencing which gave a unique sequence for the heavy chain, and the reported sequence for beta 2-microglobulin; and 4) immunoprecipitation of the bulk of the preparation by appropriate alloantisera.     

Methods in coccidiosis research: separation of oocysts from faeces.

Factors which may be important in the large-scale extraction of coccidial oocysts from faeces ha.ve been investigated with Eimeria tenella. Age of bird, inoculum, feeding status at the time of inoculation, period of collection, feeding status during collection, collection medium, homogenization and sieving, flotation, washing, sporulation and further purification have all been considered. The aim has been to establish a method to produce the maximum number of oocysts of a required degree of purity and viability, with the expenditure of the minimum amount of physical effort, time, animals and chemicals. In our method, groups of chickens 3-4 weeks of age are inoculated with 5000 oocysts of E. tenella and food is supplied ad lib. Over the period 5-8 days after inoculation, faeces are collected in trays containing 2% (w/v) potassium dichromate solution, while food intake is restricted. The faecal material is homogenized, passed once through 40 and 100 mesh sieves, centrifuged and the oocysts recovered from the sediment by 3 flotations in saturated salt solution. Following washing, oocysts are sporulated by forced aeration at 30 degrees C and may be further purified by hypochlorite treatment, or passage in 5% Tween 80 solution through a glass bead column followed by sucrose density gradient centrifugation. Routine passages along these lines over a 5 year period have given a recovery of 46% of the oocysts excreted by over 7000 birds.     

On functional morphology of the skeleton in lychnisc sponges (Porifera,Hexactinellida)

By comparison with architectural models it can be shown that both the “euretoid” arrangement of fused spicules and lychnisc node itself should enhance the strength of the skeleton of the Lychniscosa. Contrary to expectation however, the Lychniscosa do not inhabit more turbulent waters than the Hexactinosa with a simpler skeletal structure. Possible reasons for this are discussed.     

Hepatic fatty cirrhosis in Texas white-tailed deer

Hepatic fatty cirrhosis (HFC) has been known to occur in certain domestic livestock species since 1931. Early studies in Texas indicated that HFC was restricted to five western counties. Recently HFC was identified in white-tailed deer (Odocoileus virginianus texanus) in South Texas for the first time. Of the deer examined, 25% were affected. The etiology of HFC was not determined.     

Incidence of aneuploidy in a human population

A population of 40,371 individuals consisting of every baby delivered at two Denver hospitals from 1964 to 1974 has been screened from aneuploidy of the sex chromosomes and chromosome 21. The pattern in time with which aneuploidy occurs suggests an epidemic component of the incidence superimposed on an approximately equal constant frequency. The epidemic incidence is most likely to be high for births from May to October, to persist for several consecutive years, and then to be absent for several consecutive years.     

Improved histological localization of GABA-transaminase activity in rat cerebellar cortex after aldehyde fixation

Summary A method for the chemical fixation of the enzyme GABA-transaminase in nervous tissue is described. It is shown that after perfusion with a formaldehyde/glutaraldehyde fixative, activity of the enzyme in cerebellar cortex is demonstrable whilst cellular morphology is preserved. Results from the improved technique have shown new sites of GABA-transaminase activity in cerebellar cortex. In view of these results a special function for glial cells in this area of brain has been suggested.
