The Fine Structure of Some Retinal Photoreceptors

An electron microscope study has been made of octopus and amphibian photoreceptors, after fixing with KMnO₄ and embedding in araldite. What has previously been seen as a single dense stratum bounding the tubular compartments (octopus) or the double membrane discs (rods and cones), now shows a double structure. We interpret this as showing that these tubules and discs have similar bounding surfaces, which are probably directly related to the cell membrane. This is confirmed by the finding that the tubules and discs are (at least occasionally) continuous with the cell membrane.     

The Influence of Morphology on Prognosis in Acute Leukemia

The presence of definite cytoplasmic granulation in at least some of the malignant cells was used as the sole criterion to separate 156 patients with acute leukemia into two groups: 110 with myeloblastic (AML), and 46 with lymphoblastic or stem cell leukemia (ALL). The median survival from the onset of symptoms in patients with AML was 20 weeks, and those with ALL 37 weeks. The difference in survival in these two groups is much greater for patients under the age of 25 than for those over the age of 25.

Previous studies in our laboratory led to the elucidation of the covalent structure of a tetraacyldisaccharide 1,4'-bisphosphate precursor of lipid A (designated lipid IVA), that accumulates at 42 degrees C in temperature-sensitive mutants defective in 3-deoxy-D-manno-octulosonic acid (KDO) biosynthesis (Raetz, C. R. H., Purcell, S., Meyer, M. V., Qureshi, N., and Takayama, K. (1985) J. Biol. Chem. 260, 16080-16088). Using [4'-32P]lipid IVA as the probe, we now demonstrate the existence of cytoplasmic KDO-transferases in Escherichia coli capable of attaching 2 KDO residues, derived from CMP-KDO, to lipid IVA. A partial purification has been developed to obtain a cytoplasmic subfraction that adds these 2 KDO residues with a 90% yield. The product is shown to have the stoichiometry of (KDO)2-IVA by fast atom bombardment mass spectrometry and NMR spectroscopy. The partially purified enzyme can utilize alternative lipid-disaccharide cosubstrates bearing five or six fatty acyl chains, but it has an absolute requirement for a monophosphate residue at position 4' of the lipid acceptor. When reincubated with a crude cytoplasmic fraction, a nucleoside triphosphate and Mg2+, (KDO)2-IVA is rapidly metabolized to more polar substances, the identity of which is unknown. The KDO-transferase(s) described in the present study should be very useful for the semisynthetic preparation of complex lipopolysaccharide substructures and analogs.     

A mathematical model for the evaluation of the behaviour during flexion of condylar-type knee prostheses

A 3D knee model was developed in order to evaluate the mechanical behaviour during flexion of condylar-type knee prosthesis. Based on the total energy minimization principle, it takes into account the articular surfaces (the tibial surface being deformable), the body weight, and the patello femoral joint. It generates the kinematics of the joint, the motion of the centre of contact, the quadriceps forces, the pressure distribution on the tibial plateau, and ligament lengths and forces between 0 and 120 degrees of flexion. The results for ten digitized knees and the commercially available prostheses are presented. They are in general agreement with experimental results published in the literature. It is concluded that this computer program may be, within its limitations, a useful tool in the preliminary evaluation of new condylar-type knee prosthesis designs.     

Flow cytometric detection of ornithine decarboxylase activity in epidermal cell subpopulations

Using flow cytometry in combination with membrane permeabilization techniques to enhance binding of antibodies with immunoreactive protein within the cytoplasm, we have developed a method to examine the ornithine decarboxylase (ODC) activity present within subpopulations of epidermal cells following acute and chronic exposure to the phorbol ester tumor promoter 12-0-tetradecanoylphorbol-13-acetate (TPA). The method described has the sensitivity to detect basal levels of ODC as well as increases in ODC at early time points following treatment with TPA and has the additional advantage of allowing subpopulation identification and characterization.     

Brood size and the cost of provisioning nestlings: interpreting Lack's hypothesis

A recent model of parental provisioning (the tradeoff model) suggests that the maximum delivery rate of food to nestlings represents a tradeoff between parental residual reproductive value and nestling survival. In contrast, Lack's hypothesis suggests that maximum provisioning rate determines brood size and therefore delivery rates are limited by shortages of food or foraging time, not by tradeoffs of parental investment. Several authors have examined the shape of the per-nestling feeding curves to test the tradeoff model against Lack's hypothesis. We show that Lack's hypothesis can produce per-nestling feeding curves consistent with the tradeoff model. Therefore, the shape of the per-nestling feeding curve cannot be used to distinguish between the models.     

Immunological Activity of Leptospira Pomona Bacterins

Seven commercially available leptospiral bacterins were compared in guinea pigs and on that basis two of them, the best and the poorest, were further compared in calves. A wide variation was observed in their antigenicity and in their ability to provide protection. Guinea pigs appeared to be satisfactory test animals for evaluation of bacterins prepared for use in cattle.
