Genetic variants of cytoplasmic malate dehydrogenase (MDH:EC:1.1.1.37) in populations in England and the Indian subcontinent

Summary A new variant malate dehydrogenase is described, designated S-MDH^Ind to indicate its discovery in the Indian subcontinent pending full comparison with all other variants. It occurred during a survey of the incidence of variant S-MDH phenotypes in 4149 subjects in north-east England and 1494 subjects from several populations in the Indian region. The variants previously thought to be restricted to the New Guinea region and to African populations occurred in three English subjects in northeast England. The incidence of variant S-MDH phenotypes in other populations is summarised.

HIGH AFFINITY l-[3H]GLUTAMATE BINDING TO POSTSYNAPTIC RECEPTOR SITES ON RAT CEREBELLAR MEMBRANES

l [³H]glutamate binding was investigated in membrane preparations derived from rat cerebellum, an area of the brain where it is likely that a high density of postsynaptic glutamate receptors occurs. Glutamate was hound specifically and, in freshly prepared membranes, was optimal under physiological conditions of pH and temperature and was associated with the synaptic membrane fraction of the cell. Specific binding occurred through a single, high-affinity process with a K_D, of 744 nM and a capacity of 73 pmol/mg protein. Unlike the findings reported for GABA, the specific binding of glutamate to fresh membranes did not involve an uptake site. Comparison of the potencies of a wide range of compounds with known pharmacological activities, demonstrated that their ability to displace specific glutamate binding was consistent with specific interactions with glutamate receptors.     

A Predictive Study of Congenital Heart Disease and Need for Care

For long-term planning in the delivery of health care, prevalence data are essential for budget estimates in terms both of distribution and training of manpower and fiscal responsibility. From incidence figures, from the knowledge of the natural history of congenital heart disease and from predicted population estimates it is possible to construct a model that reflects the prevalence of congenital heart disease. This has been done for the state of California; the methods used and the data gathered should prove useful nationally.It is estimated that there were in 1975 in California 17,531 children under 21 years of age with congenital heart disease; 24 percent of these had ventricular septal defects and 23 percent had pulmonary stenosis, 11 percent had atrial septal defects and 9 percent had aortic stenosis; the other forms of congenital heart disease constituted the remaining 33 percent. Based on these estimates it is then possible to plan the medical resources necessary for optimal care.     

Plane penetration of uterine muscle by intrauterine shields

A finite element approach is used to predict stress and deformation states of uterine muscle tissue under plane strain indentation by a flexible shield. Realistic, one-dimensional "punch" elements at the shield edges assure that muscle shear stresses remain bounded within experimentally measured values. For typical tissue, bearing pressure, deformation flow fields and edge slip stresses leading to tissue damage are calculated. Penetration depth to shield width ratios are up to 3.0. A piecewise linear, elastic approximation to the highly variable, nonlinear mechanical behavior of the tissue is used. Results are applied to the prediction of possible tissue damage by a flexible shield intrauterine contraceptive device, in place and in equilibrium with typical multiparous uteri.     

Proposed molten globule intermediates in fd phage penetration and assembly

The fd filamentous phage can be contracted to short rods called I-forms and to spheroidal particles called S-forms. The conversions from fd----I-forms----S-forms were previously suggested to mimic steps in fd penetration. The same conversions, in reverse order, were suggested to mimic steps in fd assembly. The I-forms and S-forms bind the hydrophobic probe, 1-anilino-napthalene-8-sulfonate (ANS); under the same conditions, fd binds this probe very poorly. Rigidly packed side chains in fd and nonrigidly packed side chains in I-forms and S-forms would explain the differences in ANS binding. A compilation of the properties of I-forms and S-forms indicate that: (i) they have compact structures; (ii) they have secondary structures of the same type as native phage; (iii) they have non-native morphologies; and (iv) they may have nonrigid side chain packing. These are the properties of molten globules.     

A model for fd phage penetration and assembly

Below 15 degrees C, chloroform causes fd phage to contract to I-forms, which are compact structures about 1/3 as long as the original phage. Above 15 degrees C, chloroform causes I-forms to contract to even more compact spheroidal S-forms. Here we show that the coat protein structure in I-forms is the same as the protein structure in the phage and the protein structure in S-forms is the same as the protein structure in bilayers. The conversions from fd----I-forms----S-forms are therefore suggested to mimic steps in fd penetration. The same conversions, in reverse order, are suggested to mimic steps in fd assembly.
